A method for sequence-specific deletion mutagenesis.

We describe a novel procedure for the construction of deletion mutants. Existing exonuclease-based protocols are efficient at producing randomly positioned deletions over large regions of DNA, but are of limited use in targetted mutagenesis due to their inherent sequence-specificity. We have taken advantage of the Exonuclease III-resistant nature of alpha-thio-dNTPs, incorporated into the target DNA template by a primer extension reaction, to generate base-specific alpha-thio-dNTP terminated products. Following removal of the 5' overhanging strands, the products can be cloned to generate a nested set of deletions with single base-pair increments. We demonstrate the utility of this technique by isolating multiple deletions over a 40bp region of the human beta-interferon promoter.

Takayasu's arteritis in a pregnant woman. A case report.

Takayasu's arteritis in a pregnant white patient is described. This case highlights the fact that, irrespective of race, any patient who presents for the first time in pregnancy with pulseless hypertensive disease or other features suggestive of Takayasu's arteritis, should have their management in labour determined by the number of complications that are present. These are retinopathy, arterial aneurysms, hypertension and aortic regurgitation. These prognostic criteria will result in a classification of patients that will lead to appropriate management.

Legal regulation of human gene therapy.

The adequacy of existing legal mechanisms to regulate clinical trials of human gene therapy is examined. Existing legal controls include the federal Guidelines for Research Involving Recombinant DNA Molecules and federal regulations for the protection of human subjects. Another significant mechanism is provided by judicial oversight, i.e., judicial decisions involving recombinant DNA research. Human gene therapy does raise new issues that still must be resolved. At least two weaknesses exist in the present regulatory system: first, Recombinant DNA Advisory Committee (RAC) only has authority over federally funded research, not work done with private support, and second, RAC is not mandated to focus on difficult ethical issues, e.g., germ-line therapy, that arise from human genetic engineering technology.

Human papillomavirus infection in the cervix of pregnant females in Hong Kong.

Routine cytological examination and HPV-DNA probing were performed on cervical cells obtained from 215 pregnant Chinese females. The procedure utilized the specific binding of a mixture of complementary [32P]-labelled RNA probes to membrane-bound target HPV DNA types 6, 11, 16, 18, 31, 33 and 35. Positive results for HPV-DNA were obtained in 11/215 (5%) of the cases. The cervical smears were routinely processed and in 3/215 (1.5%) cases a cytological diagnosis of wart virus infection was made. All three were included in the 11 probe-positive cases. Using similar molecular biological techniques, pregnant Western females have an incidence of HPV infection of the cervix which is up to five times that of the present series. This is an unexpected finding in view of the fact that the incidence of cervical cancer is higher in Chinese women than American women.

Estimation of noncompartmental parameters: a technical note.

Noncompartmental pharmacokinetic parameters are usually expressed in terms of the area under the so-called first moment curve and the area under the plasma concentration-time curve. These areas are normally computed by numerical integration. There is an inconsistency in the way in which they are presently estimated. This inconsistency is detailed and a consistent approach to the computation of noncompartmental parameters is outlined. The difference between the two methods is illustrated using some data from the literature.

Two rare cases of 6p partial deletion.

Two rare cases of 6p partial deletion (6p23----pter) are described. Both patients are at or past the adolescent stage, with severe mental retardation and severe to moderate developmental retardation. Physical dysmorphic features that stand out are: short forehead, borderline microcephaly, low-set malformed ears, hyperplastic nares, dental anomalies and short terminal phalanges. The diversity of the phenotypic features has considerable variations in patients with ring of 6, apparently reflecting the relative loss of p and q arms. A case of a larger terminal deletion and a report of an interstitial deletion is also reviewed.

Comparison of an adapted milk with whole cows milk.

Two groups of babies, one having SMA, one homogenised milk, were studied. Haemoglobin was measured in the neonatal period, and at 3, 6, 10 1/2 and 15 months. Five babies of the homogenised milk group became anaemic. Stools were tested for occult blood, with more strongly positive test results found in babies having homogenised milk. Serum lipids were measured at 3, 6, 10 1/2 and 15 months. Serum cholesterol was significantly higher at three and six months in the homogenised milk group. There were no significant differences in growth between the groups. Review of progress at five years of age discovered higher serum cholesterol in those children of social class three who had homogenised milk compared with the similar five year olds who had had SMA.

Caecal herniation through the foramen of Winslow.

The 28th reported case of caecal herniation through the foramen of Winslow is presented. A review of the literature reveals a mortality of 49% in lesser-sac herniae due to late diagnosis and operative difficulties. The comparative ease of hernial reduction after deflating trapped bowel by small-bore needle aspiration is discussed.

Age differences in verbal mediation: a structural and functional analysis.

Five experiments were conducted to investigate potential structural and functional differences in verbal mediators given by young and elderly adults to CVC material. Experiment 1 found no significant differences in the complexity and flexibility of mediator formation by the two age groups. Experiment 2 replicated these findings using a different sample and task, and different materials. Additionally, this study also indicated a significant rank order correlation for the particular types of mediators used. Experiment 3 indicated that under some situations young subjects may be able to discriminate young from old mediators while Experiment 4 showed that younger subjects had no preference for which mediator would be most helpful to them in a learning task. Experiment 5 found that, in a paired associate learning task, young subjects learned faster the mediators given by other young subjects. The data were discussed in terms of current methodolical practices and in terms of localizing age-related memory deficits in structure or process variables.

Multi-laboratory comparison of three heparin-Mn2+ precipitation procedures for estimating cholesterol in high-density lipoprotein.

Plasma high-density lipoprotein is commonly estimated by measuring the cholesterol remaining in plasma supernatant solutions after other lipoproteins, which contain apolipoprotein B, are precipitated with heparin and Mn2+. The method (method I) now in use by the Lipid Research Clinics, in which Mn2+ is at 46 mmol/liter final concentration, is reasonably accurate, but precipitation and sedimentation of lipoproteins other than high-density lipoproteins is often incomplete. We evaluated two modifications of method I. In method II, the Mn2+ concentration was doubled; the second modification (method III) included the increased Mn2+ concentration in a combined heparin Mn2+ reagent, decreased sample volume (2 ml), and a shorter incubation time (10 min at room temperature). The percentages of samples with turbid supernates (i.e., incomplete sedimentation) by methods I, II, and III were 9, 3, and 2%, respectively. Among non-turbid supernates, the percentages of samples containing measurable apolipoprotein B (incomplete precipitation) were 79, 19, and 16%, respectively. We conclude that method III is the most convenient and accurate of the three procedures.
