Production of lymphokines by murine cortisone resistant thymocytes stimulated by concanavalin A. III. Effect of cytotoxic pretreatment with anti-Lyt antibodies on interleukin-2 and interferon producing cells.

Lyt phenotypes of interleukin-2 and interferon producing Balb/c cortisone resistant thymocytes, stimulated by concanavalin A in vitro, were studied using cytotoxic anti-Lyt-1.2 and anti-Lyt-2.2 alloantisera and monoclonal antibodies. It was found that while interleukin-2 was produced by both Lyt1+ and Lyt2+ cells, the ability to secrete IFN was limited to Lyt2+ cells only.

Quantitative analysis of tissue distribution of Ly10-like murine alloantigen(s) with antisera and monoclonal antibodies.

Tissue distribution of non-Lyt1.1 ("Ly10-like") antigen or antigens encoded by short chromosomal segment differentiating B6-Ly-1a congenic strain from B6 strain of mice was studied by quantitative absorption of (BALB/c X B6)F1 anti B6-Ly-1a antiserum and by direct cytotoxicity of Ly-10-132-12-26 monoclonal antibody on lymphoid cell populations. Identical strain but not tissue distribution pattern does not allow to conclude whether antiserum and monoclonal antibody detect the same or closely linked antigens. Absorption experiments revealed the highest antigen content in the brain tissue, lower in testis and kidney, still lower in lymphoid organs and the lowest in liver and lung. Among lymphoid cells, bone marrow cells had highest absorbing capacity, followed by thymus, spleen and lymph nodes. Monoclonal antibody lysed almost 100% of thymocytes, 30% bone marrow cells and 10-20% of spleen and lymph node cells (both T-cell and B-cell enriched populations contained the same proportion of positive cells). Cortisone resistant thymocytes showed the same sensitivity as cortical thymocytes to Ly-10-132-12-26 antibody which is distinguishable characteristics of medullary thymocytes from peripheral T cells. Mitogen activated lymphocytes exhibited significantly higher expression of Ly10-like antigen than resting peripheral lymphocytes.

Monoclonal anti-TL antibody recognizing a TL.2-like antigen.

We obtained a monoclonal anti-TL antibody-producing hybridoma by fusion of Sp2/0 hybrid cells with spleen cells from (B6 X A-Tlab) F1 mice immunized with ASL1 leukemia cells. The antibody, TL-22-27-17, reacts in complement-dependent cytotoxicity assay with ASL1 cells as well as with normal thymocytes of TL+ mouse strains (B6-Tlaa, A, BALB/c, DBA/2 and 129) but not with thymocytes from TL- mice (B6--Ly-1a). In addition, it did not react with lymph node cells or Concanavalin A-induced spleen blasts from B6-Tlaa mice; this result shows that TL-22-27-17 recognizes TL and not Qa-1 antigen. The positive reactions with BALB/c (TL.1-2+3-5-6-7+) and 129 (TL.1-2+3-5-6-7-) thymocytes suggest that antibody TL-22-27-17 reacts with TL.2 or TL.2-like antigenic determinant.

Production of lymphokines by murine cortisone-resistant thymocytes, stimulated by concanavalin A. II. Kinetics of interleukin-2 and interferon production in vitro.

The results presented in this study indicate that cortisone-resistant thymocytes stimulated with concanavalin A produce interferon and interleukin-2. The kinetic studies show that the peak of interleukin-2 activity precedes the maximal level of interferon activity which occurs later and does not show as rapid decline as the activity of interleukin-2.

Production of lymphokines by murine cortisone-resistant thymocytes, stimulated by concanavalin A. I. Comparison of optimal culture conditions for proliferative response and interleukin-2 production.

The conditions for optimal proliferation and production of interleukin-2 by cortisone resistant thymocytes stimulated by concanavalin A were compared. The results show that identical culture conditions are optimal for both responses but peak of interleukin-2 production precedes the peak of proliferation.

Failure of in vitro induction of killer cells in C57BL/6 anti-C57BL/6-LYT1.1 strain combination.

Several in vivo preimmunization schedules which are commonly used for induction of in vitro cell-mediated cytotoxicity against weak transplantation antigens have been tested for ability to generate killer cells in congenic anti B6-Lyt1.1 strain combination. Under the conditions studied no cytotoxic cells were generated. Under the same same conditions good cytotoxic response against H-Y antigen was observed.

Studies on cell surface antigens of mouse leukemic and normal lymphocytes. IV. Functional studies on mouse T lymphocyte subpopulations. B. Lyt phenotype of cytotoxic T lymphocytes and their precursors in B6-Lyt-1.1 mice.

Lyt phenotype of in vitro generated alloreactive cytotoxic T lymphocytes (CTL) and their precursors in B6-Lyt-1.1 (Thy-1.2, Lyt-1.1, Lyt-2.2) congenic strain of mice was studied. The generation of CTL in vitro was completely abrogated by pretreatment of responder cells with antisera to Thy-1.2, Lyt-1.1 and Lyt-2.2 antigens and complement. Mixing of anti-Lyt-1.1-pretreated cells with anti-Lyt-2.2-pretreated cells did not restore CTL generation indicating that both Lyt antigens are expressed on the same precursor cell population. The effect of anti-Lyt sera was specific because the generation of CTL was prevented only when cells from strains possessing appropriate Lyt alleles were pretreated. CTL were eliminated by lysis with anti-Thy-1.2 serum and strongly reduced by anti-Lyt-1.1 and anti-Lyt. 2.2 sera. Nevertheless, a small but significant proportion of CTL was insensitive to lysis with anti-Lyt sera. These data indicate that a phenotype of precursor cells and of majority of CTL is Thy-1.2+ Lyt-1.1+ Lyt-2.2+.

Possible association of cytotoxic T-lymphocyte antigen 4 gene promoter single nucleotide polymorphism with acute rejection of allogeneic kidney transplant.

Cytotoxic T-lymphocyte antigen 4 (CTLA-4) molecule is an important inhibitor of T-lymphocyte response. Polymorphisms in the CTLA-4 gene have been described to be associated with numerous autoimmune diseases. However, similar studies in solid organ transplantation have been scarce. Therefore, we examined the distribution of three single nucleotide dimorphisms, namely, -1147T/C, -318C/T, and +49A/G, in two groups of allogeneic kidney graft recipients: (1) those with at least one acute rejection episode ("rejectors"; n = 38) and (2) those with no signs of acute rejection ("nonrejectors"; n = 53). Allele frequencies in both groups of patients were similar in two positions, -1147T/C and +49A/G. However, rejectors showed slight differences from nonrejectors for allele and genotype frequencies in position -318. The -318T allele was two times less frequent among rejectors than nonrejectors, a difference that was close to statistical significance (P = .039; P corrected = .0583), and may reach it when greater numbers of patients are tested.
