Neuroendocrine effects of flunarizine treatment in postmenopausal women.

Flunarizine (10 mg/day for 60 days) was given to eight postmenopausal women with common migraine. Plasma LH pulsatility fluctuation, peripheral concentrations of prolactin (PRL), cortisol, beta-endorphin (beta-EP), beta-lipotropin (beta-LPH) and Pain Total Index (PTI) were evaluated before and after treatment. PTI was significantly reduced by flunarizine, which did not affect beta-LPH, beta-EP and cortisol plasma levels. On the contrary, both PRL values and amplitude, and length of LH pulses had increased at the end of treatment. Flunarizine reduced head pain in postmenopausal women. However, the enhancement of both PRL and LH release indicates that this calcium antagonist might interfere with the dopaminergic tonus.

Synchronizing EEG effect of mianserin.

The AA. report on the eegraphic, behavioral and neurovegetative effects caused by the i.v. administration of mianserin, in the rabbit. This substances induces behavioral sedation and EEG synchronization. In addition, these changes are partially reinforced with L-5-hydroxytryptophan (L-5HTP) and inhibited by pretreatment with DL-p-chlorophenylalanine (PCPA). In the light of these data, the AA. discuss the possible interactions between mianserin and neurotransmitter systems, particularly the serotoninergic one.

Headache temporal patterns: towards a chronobiological model.

Rhythmical changes in terrain susceptibility and trigger occurrence can induce periodic temporal patterns in headache patients, with readily foreseeable frequency cycles of attacks. The impaired ability of biological functions to adjust to cyclic environmental changes represents the minus, the handicap, the increased risk of clinical symptomatology (headache); attacks occur when the precarious systems of these patients are forced by cyclical environmental stimuli towards phase shiftings or to cope with new levels of performance. A dyschronic hypothesis of primary headache must be based upon observations demonstrating that along with dysfunctions of the pain control systems there is also a vulnerability of the rhythmic physiological organization of CNS.
