Fenestrations in endothelium of rat liver sinusoids revisited by freeze-fracture.

The freeze-fracture appearance of fenestrations of sinusoidal endothelial cells has been reassessed in perfusion-fixed rat liver. Fenestrations, limited to the attenuated portions of the endothelial cell cytoplasm are usually round or oval in shape, with diameters ranging from 40-600 nm. Within a given fenestrated region, they are disposed in clusters forming sieve plates. In addition, the presence of wide, irregularly shape openings suggest that adjacent fenestrations may coalesce and result in the larger openings. Our data thus confirm the existence of both small and large fenestrae in the endothelial wall, in contradistinction to previous studies showing only regular, medium sized (100 nm) openings.

Encephalopathy subsequent to accidental poisoning with chlorpromazine.

A cerebral biopsy performed in an infant 9 months after accidental poisoning with chlorpromazine was studied. Light microscopy disclosed necrosing encephalopathy, while electron microscopy showed neuronal, glial, myelinic and vascular lesions. The findings in the present study suggest that the various neurologic syndromes appearing in cases of prolonged treatment with neuroleptic drugs and persisting after cessation of therapy would be due to consequent occurrence, in specific areas, of some irreversible cerebral lesions.

Identification of Duchenne muscular dystrophy carriers. Electron microscopical investigation of skeletal muscle.

In order to identify a subclinical dystrophy, muscle biopsy specimens from 15 mothers of boys with Duchenne muscular dystrophy were studied by electron microscopy. Genetic investigations and biochemical determinations were not conclusive for a diagnosis of the carrier state. The fine structural lesions were sufficient to vertify the diagnosis of a subclinical dystrophy, and our findings were in agreement with other investigators. However, original intranuclear filamentous formations were noted in one case.

Some ultrastructural aspects of endemic nephropathy of the Balkans.

Two renal biopsies selected from a large number of specimens taken by renal puncture from patients suffering from endemic nephropathy were studied by electron microscopy. Ultrastructural lesions were seen as electron-dense deposits in the walls of the glomerular capillaries, sometimes associated with glomerular proliferative processes. Such lesions, which are probably the early stages of this obscure renal disease, suggest that an auto-immune mechanism may be involved.

The effect of deprivation of glucose on the ultrastructure and function of the superior cervical ganglion of the rat in vitro.

The superior cervical sympathetic ganglion of the rat kept in vitro in a bicarbonate-buffered Krebs' solution retains its capacity for synaptic transmission and axonal conduction during more than 36 hr. After glucose withdrawal, synaptic transmission is lost in 2(1/2) hr and this loss is irreversible; on the other hand, axonal conduction can still be measured on the postganglionic nerve for more than 24 hr after glucose deprivation. Electrophysiological measurements as well as electron microscope studies revealed specific changes at the level of the presynaptic terminal processes, while the ganglion cells and the satellite cells remained relatively unaltered. The presynaptic lesion due to lack of glucose can be prevented by keeping the preparation in vitro at 6 degrees C. This strongly suggests that this lesion results from a major disturbance of the metabolism of the presynaptic fibers.
