Effect of ascorbic acid in the detoxification of the insecticide dimethoate in the bone marrow erythrocytes of mice.

The antimutagenic potential of ascorbic acid in the detoxification of the organophosphorus insecticide dimethoate was evaluated in female Swiss albino mice using the in vivo bone marrow micronucleus test. Groups of three mice were treated with distilled water (control), 1% dimethyl sulphoxide (solvent control), or 86, 129.5 or 259 mg ascorbic acid/kg body weight, with or without the concurrent administration of 150 mg dimethoate/kg. There was a statistically significant increase in the frequency of micronuclei in dimethoate-treated mice. However, in mice that were given dimethoate and ascorbic acid simultaneously, the numbers of micronuclei did not differ significantly from control values, thus indicating the protective role of ascorbic acid.

Effect of glutathione on mitomycin C-induced micronuclei in bone marrow erythrocytes of Swiss albino mice.

The antimutagenic potential of glutathione (GSH) on mitomycin C (MMC)-induced micronuclei was evaluated in Swiss albino mice using the in vivo bone marrow micronucleus test. Six groups of animals were maintained simultaneously. The first group received distilled water only, the second group of animals received 2 mg/kg MMC and the third group was administered 4 doses of GSH, i.e., 20, 40, 80 and 160 mg/kg. The fourth group of animals received GSH and MMC simultaneously. The fifth and sixth groups received a cumulative dose of GSH followed by MMC after 24 h. The fifth group of animals were killed 6 h after the administration of MMC, while the sixth group were killed 24 h after the administration of MMC. The results clearly show a statistically significant increase in micronuclei in MMC-treated animals and also in animals that received GSH followed by MMC. However, there was a decrease in micronuclei in animals that received GSH and MMC simultaneously. The results clearly indicate that GSH exhibits an antimutagenic property in the presence of MMC. It is also observed the treatment with GSH prior to MMC does have some protective effect.

Screening of chromosomal aberrations and sister chromatid exchanges in peripheral lymphocytes of vegetable garden workers.

1. Twenty-five male workers occupationally exposed to DDT, BHC malathion, parathion, dimethoate, fenitrothion, urea and gromor were selected as subjects for the analysis of chromosomal aberrations and sister chromatid exchanges (SCE) in peripheral lymphocytes. 2. Blood samples were collected from 30 normal healthy males from the same age group and socioeconomic class for the control. 3. The frequency of chromosomal aberrations and SCEs increased significantly irrespective of the duration of exposure to pesticides, when compared to controls.

Cytogenetic effects of trifluoperazine in mice.

The cytogenetic effects of trifluoperazine have been studied in male Swiss albino mice using the micronucleus test, chromosomal analysis in germ cells and a sperm morphology assay. The mice were treated by gavage with 80, 120 or 160 micrograms trifluoperazine/kg, divided in each case into two equal doses given 24 hr apart. The dose levels were selected on the basis of standard human therapeutic dosage. Compared with the findings in control mice dosed with distilled water, there were significant increases in the frequency of micronuclei, of chromosomal aberrations in the spermatocytes and of abnormal sperms, at all the levels of trifluoperazine treatment.

The effect of thiram on the germ cells of male mice.

The effects of thiram or tetramethylthiuram disulphide on the germ cells of Swiss albino male mice were evaluated by analysing spermatocytes (derived from treated spermatogonia) for chromosomal aberrations and by the sperm-head morphology assay. The total doses tested were 80, 200 and 320 mg/kg body weight given by gavage in three consecutive daily doses, the top dose being slightly below the LD50 of thiram. There was a significant increase in the frequency of numerical chromosomal aberrations and abnormal sperms in mice treated with thiram at all dose levels. Such results could have implications for man in that they suggest that undue exposure to thiram could result in the birth of human infants with numerical chromosomal aberrations.

Monitoring of workers occupationally exposed to pesticides in grape gardens of Andhra Pradesh.

Studies were undertaken to investigate the effect of pesticides on workers employed in grape gardens of Andhra Pradesh. The data obtained on the reproductive performance showed an increase in the incidence of abortions and stillbirths. Out of 12 employed couples in grape gardens, 2 were found to be sterile. Cytogenetic studies revealed a significant increase in chromatid breaks and gaps in chromosomes of peripheral blood in workers exposed to pesticides. In addition, a high frequency of satellite associations was recorded in these workers.

In vitro effect of trifluoperazine on human lymphocytes.

Trifluoperazine, a phenothiazine group of tranquilizer was studied for its chromosome damaging effect in human lymphocyte cultures. The cells were treated with 10, 15 and 20 micrograms/culture of trifluoperazine for 24, 48 and 72 hours. A high incidence of chromatid breaks and gaps was observed after 48 and 72 hours treatment. The study revealed that the drug acts mostly on the late S and G2 phase of the cell cycle.

Effect of chrysotile asbestos fibers on germ cells of mice.

An Indian form of chrysotile asbestos procured from a local asbestos factory (Hyderabad) was tested for its toxic effects on spermatocytes and sperm of mice. Swiss albino male mice were fed orally with chrysotile asbestos suspended in water. The concentration tested was 20 mg/kg/day. Chronic oral administration of chrysotile failed to induce chromosomal aberrations and abnormal sperms in mice.
