Synthesis and pharmacological activity of derivatives of exo-trimethylenenorbornane. I.

Lithium aluminum hydride reduction of exo-5,6-trimethylenenorbornan-2-one oxime gave in good yield exo-5,6-trimethylenenorbornan-2-endo-amine, from which a number of amides were prepared by reaction with acyl chlorides. Lithium aluminum hydride reduction of the above-mentioned amides afforded N-substituted exo-5,6-trimethylenenorbornan-2-endo-amines in good yields. Some amides showed CNS depressant and anti convulsant activity, and an amine convulsant activity in mice.

Amphetamine↗

Synthesis and pharmacological activity of derivatives of exo-trimethylenenorbornane. II.

By reaction of secondary amines with 3-hydroxymethylene-exo-5,6-trimethylenenorbornan-2-one, synthesized from exo-5,6-trimethylenenorbornan-2-one and sodium/ethyl formate, a series of N,N-disubstituted 3-aminomethylene-exo-5,6-trimethylenenorbornan-2-ones has been obtained. These compounds gave N,N-disubstituted 3-aminomethyl-exo-5,6-trimethylenenorbornan-2-ones by LiAlH4 or catalytic reduction. Starting from the same alpha-hydroxymethyleneketone, pyrazole derivatives (V) and (VI) have been also synthesized; the former showed anticonvulsant activity in mice.

Amphetamine↗

Synthesis and pharmacological activity of derivatives of exo-trimethylenenorbornane. III.

Two synthetic routes to (exo-5,6-trimethylenenorbornan-2-ylidene)acetonitrile starting from exo-5,6-trimethylenenorbornan-2-one are described. From the above-mentioned nitrile, 2-(exo-5,6-trimethylenenorbornan-2-ylidene)ethanamine and 2-(exo-5,6-trimethylenenorbornan-2-yl)ethanamine, as well as N,N-dimethyl and N,N-diethyl derivatives of the latter, were prepared.

Derivatives of 1,2,3,3-tetramethyl-2-azabicyclo[2.2.2]octan-5-endo-ol with antiarrhythmic and other activities. II.

Cyanoethylation of 1,2,3,3-tetramethyl-2-azabicyclo[2.2.2]octan-5-endo-ol, followed by LiAlH4 reduction and reaction with acyl chlorides, gave a series of amides derived from 5-endo-(3-aminopropoxy)-1,2,3,3-tetramethyl-2-azabicyclo[2.2.2]octane. The syntheses of N-methyl, N,N-dimethyl and N,N-dipropargyl derivatives of the afore mentioned amine, as well as of 5-endo-methylcarbamoyloxy- and 5-endo-phenylcarbamoyloxy-1,2,3,3-tetramethyl-2-azabicyclo[2.2.2]octane, are also described. Strong antiarrhythmic and local anesthetic activity was observed in two amides; an urethane showed antiarrhythmic activity only.

Nitrogen mustards derived from alpha-hydroxymethyleneketones.

alpha-[Bis-(2-chloroethyl)]aminomethyleneketones were prepared starting from alpha-hydroxymethyleneketones and N-(2-chloroethyl)-2-chloroethanamine. Also 3-[2-(2-chloroethylthio)ethyl]aminomethylene-2-bornanone was synthesized from 2-(2-chloroethylthio)ethanamine and 3-hydroxymethylene-2-bornanone. This compound and 3-[bis-(2-chloroethyl]aminoemthylene-2-norbornanone were active against P388 lymphocytic leukemia in mice.

Reaction of dichloroketene and sulfene with N,N-disubstituted 6-aminomethylene-5,6,8,9-tetrahydro-7H-benzocyclohepten-7-ones. Synthesis of 5H-benzo(5,6)cyclohepta(2,1-b)pyran and 5H-benzo(5,6)cyclohepta(1,2-e)-1,2-oxathiin derivatives.

The 1,4-cycloaddition of dichloroketene to N,N-disubstituted 6-aminomethylene-5,6,8,9-tetrahydro-7H-benzocyclohepten-7-ones afforded N,N-disubstituted 4-amino-3,3-dichloro-3,4,10,11-tetrahydro-5H-benzo[5,6]cyclohepta[2,1-b]pyran-2-ones only in the case of full or partial aromatic N-substitution. The relative adducts gave N,N-disubstituted 4-amino-3-chloro-10,11-dihydro-5H-benzo[5,6]cyclohepta[2,1-b]pyran-2-ones by dehydrochlorination. The 1,4-cycloaddition with sulfene occurred readily in the case of both aliphatic and aromatic N-substitution to give N,N-disubstituted 4-amino-3,4,10,11-tetrahydro-5H-benzo[5,6]cyclohepta[1,2-e]-1,2-oxathiin-2,2-dioxides. These compounds, as well as the pyran derivatives, can be considered as linear heterocyclic analogues of the Amitriptyline ring system.

2- and 5- substituted 5-amino-2,6,6-trimethyltetrahydropyrans.

Reaction of N,N-disubstituted 2-carboxyamido-2,6,6-trimethyltetrahydropyran-5-carboxylic acids with diphenyl phosphorazide in the presence of triethylamine and benzyl alcohol gave N,N-disubstituted 5-benzyloxycarbonylamino-2-carboxyamido-2,6,6-trimethyltetrahydr-pyrans, which in turn yielded N,-disubstituted 2-aminomethyl-5-methylamino-2,6,6-trimethyltetrahydropyrans by LiA1H4 reduction. 5-Benzyloxycarbonylamino-2-carboxypyrrolidino-2,6,6-trimethyltetrahydropyran gave 5-amino-2,6,6-trimethyl-2--pyrrolidinomethyltetrahydropyran by hydrogenolysis followed by LiA1H4 reduction. Among the amides prepared from the latter, the diphenylacetic acid amide showed antiarrhythmic and local-anesthetic activity.

(III) showed antiinflammatory activity and (VIII) an anticonvulsant action.

[N 1-nicotinoyl and isonicotinoylpyrazoles].

Synthesis of twelve N 1-nicotinoyl and isonicotinoylpyrazoles by reaction of 4,5-tetra-, penta-, hexamethylenepyrazoles, dihydronaphtopyrazole, bornylenepyrazole and cineolylenepyrazole with nicotinoyl and isonicotinoylchloride, is described. The structure of N 1-derivatives of these compounds was ascertained by comparison of N.M.R. spectra with those of 1-nicotinoyl and 1-isonicotinoyl-4,5-pentamethylene-5-hydroxy-2-pyrazoline, obtained by reaction of 2-hydroxymethylenecycloheptanone with nicotinoyl and isonicotinoylhydrazide, respectively. N 1-isonicotinoylbornylenepyrazole shows weak C.N.S. depression, muscle relaxation and anticonvulsant activity.
