Neurophathological findings are reported in a 15-year-old boy who died 8 months after acute rubella encephalitis. There are widespread bilateral necroses in large areas of the cerebral cortex with a diffuse macroglial and microglial reaction. In basal ganglia, degenerative changes and a more or less pronounced rarefication of nerve cells as well as very small cystic areas and glial scars are observed. In the white matter, neither foci of demyelination nor perivascular cuffings nor focal gliosis occurred. The results are compared with other observations in the literature. It is concluded that rubells encephalitis is a disease of a variety of pathogenetic mechanisms and has a variety of clinical and morhpological features. It may evidently be caused by direct viral invasion of the CNS or by hypersensitivity reaction.
The transplacental activity of N-ethyl-N-nitrosourea (ENU) was tested in rats, rabbits, Syrian golden hamsters, Dzungarian dwarf hamsters, guinea pigs, dogs, and rhesus monkeys. In the offspring of rats, multiple tumours of the central and peripheral nervous system were found in 100 per cent, whereas in rabbits kidney tumours developed. In Syrian golden hamsters, the application led mostly to neoplasms of the thyroid gland. Dzungarian dwarf hamsters and monkeys prooved to be resistant. 9 out of 15 dogs died shortly after birth. In two dogs a carcinoma of the thyreoid gland and one of an ovary were found. Because of an infection, the experiments with guinea pigs were to be interrupted untimely. Therefore, a final answer is impossible. Own results were compared with the findings of other authors. The significance of the different susceptibility in distinct species is discussed.
Application of methyl- and/or ethylnitrosourea during intrauterine or postnatal life did not induce tumours in Macaca mulatta. Most of the animals survived 5 to 10 years after the beginning of the experiments and are in good health at present. Complete postmortem examinations were performed in all monkeys who died.
Repeated intraperitoneal injections of methylnitrosourea (MNU, 10 mg/kg b. w.) lead to tumours of the nervous system in 32 out of 44 hooded rats (72.7 per cent) and to extraneural growths in 38 animals (86.4 per cent). After combined application of MNU and intramuskular injections of prednisolute (0.56 mg/kg b. w.), in 11 out of 42 rats (26.2 per cent) tumours of the nervous system were found. In this group, there were extraneural tumours in 17 animals (40.5 per cent). The results were compared with findings of other authors obtained after combined application of chemical carcinogens and corticosteroid hormones.
