Intermediate nonlinear evolution of the Parker instability: formation of convection-induced discontinuities and absence of finite-time singularities.

Direct numerical simulations of the line-tied Parker (or magnetic Rayleigh-Taylor) instability, based on the fully compressible ideal magnetohydrodynamic equations, are presented. In the intermediate nonlinear phase, the instability continues to grow exponentially in time and the plasma tends to develop convection-induced discontinuities in the form of shocklike coherent structures. No evidence of finite-time singularities is seen.

Nonlinear compressional waves in a two-dimensional Yukawa lattice.

A modified Korteweg-de Vries (KdV) equation is obtained for studying the propagation of nonlinear compressional waves and pulses in a chain of particles including the effect of damping. Suitably altering the linear phase velocity makes this equation useful also for the problem of phonon propagation in a two-dimensional (2D) lattice. Assuming a Yukawa potential, we use this method to model compressional wave propagation in a 2D plasma crystal, as in a recent experiment. By integrating the modified KdV equation the pulse is allowed to evolve, and good agreement with the experiment is found. It is shown that the speed of a compressional pulse increases with its amplitude, while the speed of a rarefactive pulse decreases. It is further discussed how the drag due to the background gas has a crucial role in weakening nonlinear effects and preventing the emergence of a soliton.

Long-term effects of fertilization on soil organic carbon changes in continuous corn of northeast China: RothC model simulations.

Soil organic C (SOC) content can increase by managing land use practices in which the rates of organic C input exceed those of organic C mineralization. Understanding the changes in SOC content of Black soils (mainly Typic Halpudoll) in northeast China is necessary for sustainable using of soil resources there. We used the RothC model to estimate SOC levels of Black soils under monoculture cropping corn in a long-term fertilization trial at Gongzhuling, Jilin Province, China. The model outputs for the changes in SOC were compared with measured data in this long-term fertilization/manure trial. The sound performance of model in simulating SOC changes suggests that RothC is feasible with Black soils in the temperate climatic region of northeast China. The modeled and measured results indicated that the treatment without fertilizer/farmyard manure (FYM) addition led to a continuous decline in SOC during the study period and N and NPK fertilization were inadequate to maintain the SOC levels in the plow layer (upper 20 cm) unless FYM was added under the current conventional management associated with no above-ground crop residues returning into the soil. Soil organic carbon could follow the same path of decline if the same management practices are maintained. Model results indicate that returning above-ground crop residues to the soil from 2002 to 2022 would increase SOC by 26% for the treatment without fertilization addition, 40% for N treatment, 45% for NPK treatment, and 38% and 46% for N and NPK treatments with FYM addition, compared to the levels in the corresponding treatments in 2002. The simulation results suggest that the RothC model is a feasible tool to assess SOC trend under different management practices, and returning above-ground crop residues into the soil would lead to a remarkable increase in SOC of Black soils in the region.

In silico analysis indicates a similar gene expression pattern between human brain and testis.

OBJECTIVES: Previous data has reported similarity between human brain and testis gene expression patterns. Brain is the most important tissue in human speciation. Hence, it means that human testis could also play a crucial role in human speciation if these two tissues exhibit similar gene expression patterns. However, previous reports were based on only limited and scattered data. Determining the large scale anatomy of gene expression patterns of various human tissues could draw a more convincing conclusion, and better our understanding of the correlation/inter-correlation among different tissues. Furthermore, it could also provide a clue for evolutionary study. METHODS: To obtain gene expression information for large-scale data analysis, expression data of 760 Unigenes in seventeen human tissues (liver, lung, testis, brain, ovary, uterus, colon, stomach, heart, eye, kidney, spleen, gall bladder, breast, thymus, prostate and pancreas) were retrieved by DDD (differential digital display) analysis, and this expression data was subjected to clustering analysis. These Unigenes represent a wide range of genes classified according to their characterization and function. RESULTS: Among the 17 tissues, the highest similarity in gene expression patterns was between human brain and testis, based on DDD and clustering analysis. Genes contributing to the similarity include ribosomal protein (RP) genes as well as genes involved in transcription, translation and cell division. CONCLUSIONS: Present results provide evidence to support the proposal that human testis and brain share the highest similarity of gene expression patterns. The implications of the similarity regarding that both brain and testis contributed to human speciation are discussed.

Mortality in 7B2 null mice can be rescued by adrenalectomy: involvement of dopamine in ACTH hypersecretion.

The serine protease prohormone convertase 2 (PC2), principally involved in the processing of polypeptide hormone precursors in neuroendocrine tissues, requires interaction with the neuroendocrine protein 7B2 to generate an enzymatically active form. 7B2 null mice express no PC2 activity and release large quantities of uncleaved ACTH, resulting in a lethal endocrine condition that resembles pituitary Cushing's (Westphal, C. H., Muller, L., Zhou, A., Bonner-Weir, S., Schambelan, M., Steiner, D. F., Lindberg, I. & Leder, P. (1999) Cell 96, 689). Here, we have compared the 7B2 and PC2 null mouse models to determine why the 7B2 null, but not the PC2 null, exhibits a lethal disease state. Both 7B2 and PC2 nulls contained highly elevated pituitary adrenocorticotropic hormone (ACTH); the neurointermediate lobe content of ACTH in 7B2 nulls was 13-fold higher than in WT mice; that of the PC2 null was 65-fold higher. However, circulating ACTH levels were much higher in the 7B2 null than in the PC2 null. Because hypothalamic inhibitory dopaminergic control represents the major influence on intermediate lobe proopiomelanocortin-derived peptide secretion, dopamine levels were measured, and they revealed that 7B2 null pituitaries contained only one-fourth of WT pituitary dopamine. Adrenalectomized 7B2 null animals survived past the usual time of death at 5 weeks; a month after adrenalectomy, they exhibited normal levels of pituitary dopamine, circulating ACTH, and corticosterone. Elevated corticosterone, therefore, seems to play a central role in the lethal phenotype of the 7B2 null, whereas a 7B2-mediated dopaminergic deficiency state may be involved in the actual ACTH hypersecretion phenomenon. Interestingly, adrenalectomized 7B2 nulls also developed unexpectedly severe obesity.

Valproic acid defines a novel class of HDAC inhibitors inducing differentiation of transformed cells.

Histone deacetylases (HDACs) play important roles in transcriptional regulation and pathogenesis of cancer. Thus, HDAC inhibitors are candidate drugs for differentiation therapy of cancer. Here, we show that the well-tolerated antiepileptic drug valproic acid is a powerful HDAC inhibitor. Valproic acid relieves HDAC-dependent transcriptional repression and causes hyperacetylation of histones in cultured cells and in vivo. Valproic acid inhibits HDAC activity in vitro, most probably by binding to the catalytic center of HDACs. Most importantly, valproic acid induces differentiation of carcinoma cells, transformed hematopoietic progenitor cells and leukemic blasts from acute myeloid leukemia patients. More over, tumor growth and metastasis formation are significantly reduced in animal experiments. Therefore, valproic acid might serve as an effective drug for cancer therapy.

Transient-resonance Raman and density functional theory investigation of 4-biphenylylnitrenium, 2-fluorenylnitrenium, and diphenylnitrenium ions.

We present transient-resonance Raman spectra for the 4-biphenylylnitrenium, diphenylnitrenium, and 2-fluorenylnitrenium ions. These spectra display a number of fundamental vibrational bands whose frequencies exhibit good agreement with those computed using BPW91/cc-PVDZ density functional theory calculations for the singlet ground states of the 4-biphenylylnitrenium, diphenylnitrenium, and 2-fluorenylnitrenium ions. Comparison of these arylnitrenium ions with each other and with previous results for structurally similar biphenyl radical cations indicates that the degree of iminocyclohexadienyl character observed in these arylnitrenium ions depends on the relative orientation of the two phenyl rings, the nature of the nitrenium ion moiety, and the ability of the biphenyl-like group to accommodate positive charge through formation of a more planar-like structure with quinoidal-like character.

[Quantitative analysis of human herpes virus type 8 and expression of its genes in patients with multiple myeloma].

OBJECTIVE: To investigate if infection of human herpesvirus type 8 (HHV8) exists in Chinese patients with multiple myeloma (MM), and to identify the relationship between clinical manifestation and the viral load in bone marrow biopsy samples. METHODS: Real-time quantitative PCR was conducted to define the viral load of HHV8 in bone marrow biopsy samples from 23 patients with MM. Nested PCR was conducted to amplify the KS330(233)Bam fragment of HHV8 from peripheral blood mononuclear cells, bone marrow aspirates and bone marrow biopsy samples. RT-PCR was used to study the expression of suspected oncogenes: vIL-6 and vIRF-1 genes. Bone marrow biopsy samples of 5 patients with blood diseases other than MM and 5 patients with non-blood system diseases were used as controls. PCR product of BCBL-1 cell line was used as positive control. RESULTS: (1) The positive rate of HHV8 in bone marrow biopsy samples from patients with MM was 69.6% (16/23) by real-time fluorescence quantitative PCR, and 82.6% (19/23) by nested-PCR. 16 bone marrow samples positive by fluorescence quantitative were positive too by nested-PCR. However, 3 bone marrow samples positive by nested-PCR were negative by fluorescence quantative. HHV8 was positive in only one of the 25 bone marrow aspirates from patients with MM (4%) and in none of the 20 peripharal blood samples from patients with MM. PCR showed negative result in 7 bone marrow aspirates taken form 13 MM patients with positive result by fluorescence quantative PCR. HHV8 was negative in bone marrow biopsy samples from patients with other diseases. HHV8 was negative in the peripheral blood samples from 16 MM patients, among which 9 out of the 11 cases with bone marrow biopsy samples taken showed HHV8 positive by fluorescence quatative PCR. Bone marrow biopsy samples of all patients with other blood diseases and non-blood system diseases were HHV8 negative. RT-PCR showed that among 12 MM patients the expression rate of vIRF-1 was 83.3% (10/12), and expression rate of vIL-6 was 0. The PCR positive products tallied with the sequence of HHV8 DNA. Clinical analysis showed that all of the patients with negative results had had chemotherapy. CONCLUSION: There is a certain association between multiple myeloma pathogenesis and HHV8. In detection of HHVb, nested-PCR is more sensitive than fluorescence quantitaative PCR. The viral gene vILR-1 plays a certain role in the pathogenesis of MM.

Genetic analysis of conservation and variation of lipooligosaccharide expression in two L8-immunotype strains of Neisseria meningitidis.

Neisseria meningitidis strains A1 and M978 both express the lipooligosaccharide (LOS) L8 immunotype [Gu et al., J. Clin. Microbiol. 30 (1992) 2047-2053]. Under different growth conditions, strain A1 did not change its LOS profile whereas strain M978 produced variable LOS profiles on SDS-PAGE. To understand the genetic basis of LOS conservation and variation, their lgt locus encoding glycosyltransferases responsible for the biosynthesis of the alpha-chain of LOS was analyzed. Strain A1 possessed only two genes, lgtA and lgtH, at the lgt locus. The lgtA gene was inactivated due to a frameshift mutation; thus strain A1 expressed only L8 LOS. In contrast, strain M978 contained five genes lgtZ, lgtC, lgtA, lgtB and lgtE at this locus, thus it had a potential to express L1, L3,7 in addition to the L8 LOS. The data showed that strain A1 is a better reference strain for the L8 immunotype because of the stability of L8 LOS expression resulting from its unique lgt locus. In addition, these two strains had two new genetic organizations, lgtAH and lgtZCABE, compared to the reported gene organization at the lgt locus in N. meningitidis.

[Expression of CCR5 and its ligand in joint fluid and synovium of patients with rheumatoid arthritis].

OBJECTIVE: To investigate the expression and distribution of C-C chemokine recepter 5 (CCR5) and its ligand in peripheral blood, synovium, and synovial fluid so as to study the mechanism of selective accumulation of Th1 cells in rheumatoid joints. METHODS: Synovial fluid mononuclear cells (SFMC) and peripheral blood mononuclear cells (PBMC) were isolated from 15 patients with active rheumatoid arthritis (RA). The expressions of intracellular cytokines, CCR5 and CXCR3 on cytomembraine of SFMCs and PBMCs, and MIP-1 immunofluorescence labelling, and flow cytometry. RESULTS: (1) The selective pattern of intracellular cytokines in SFMCs was drifted towards Th1 subset. (2) The expression rate of CCR5 was 52% Y 8%, the expression rate of CXCR3 was 61% Y 12% in synovial fluid, significantly higher than those of PBMCs. (3) In RA tissues, most of the infiltrating T cells (especially CD4+ T cells), monocyres/microphages, and B cells expressed macrophage inflammatory protein, ligand of CCR5. CONCLUSION: Infiltrating T cells (especially CD4+ T((MIP)-1 cells), monocyres/microphages, and B cells in the synovium of inflammatory, RANTES, and other cytokines which cause the accumulation of CCR5+ Th1 cells.

[Characteristics of malignant clone from acute lymphocytic leukemia].

OBJECTIVE: To identify the characteristics of malignant clones in leukemia. METHODS: VH region of B lymphocytes from children with ALL, patients with MM, and tissues from fetuses obtained by induced labor at different gestational stages was amplified using RT-PCR method with one primer located in VH1, VH2, VH3, VH4, VH5, or VH6 and another primer at the join region. The PCR products were then separated on sequencing gel and their VH gene fingerprints were obtained. The fingerprints from tissues of fetal liver, spleen, thymus and bone marrow, and bone marrow B cell from 15 cases of ALL and one case of MM were compared, and VH repertoire information were deduced from those fingerprints. RESULTS: The VH6 family was the dominant family in fetal liver and the VH3 family occurred predominantly in fetal spleen and bone marrow. Monoclonal proliferation of B lymphocytes was found in 9 of the 15 patients with ALL. Of these 9 cases, proliferation of VH1 was found in 3 cases, proliferation of VH6 was found in 3 cases, and proliferation of both VH3 and VH4 in 2 cases. The VH families other than leukemic clone(s) in these patients were in normal distribution or suppression status. Oligoclonality was found in one ALL case with malignant growth of VH1, VH3 and VH6. Polyclonality with nearly normal spectrum of VH1-VH6 was detected in 6 of the 15 cases. DNA sequencing of a VH3 clone from an ALL case demonstrated a similarity to the sequence of embryonic clone from a fetus of gestational age of 13 weeks. The VH sequence of one adult MM case, with hypermutation of somatic cell, was remarkably different from that of the germline. CONCLUSION: Rearrangement of VH1, VH3, VH4 and VH6 can be frequently found in ALL. It is unlikely that leukemia in childhood originates from stimulation of foreign antigens.

Experimental determination of critical threshold in electron transport on Tore Supra.

In Tore Supra plasmas with fast wave electron heating, a critical threshold in the electron temperature gradient (inverted DeltaT(e)) is clearly observed, i.e., a finite value of inverted DeltaT(e) for which the turbulent heat diffusivity vanishes. The radial profile of this critical gradient is experimentally determined from a set of discharges characterized by similar plasma parameters with fast wave powers ranging from 0.75 to 7.4 MW. The dependence of the electron heat flux on the gradient length is found to be offset linearly. The offset term increases linearly with the ratio of the local magnetic shear to the safety factor.

Fit genotypes and escape variants of subgroup III Neisseria meningitidis during three pandemics of epidemic meningitis.

The genetic variability at six polymorphic loci was examined within a global collection of 502 isolates of subgroup III, serogroup A Neisseria meningitidis. Nine "genoclouds" were identified, consisting of genotypes that were isolated repeatedly plus 48 descendent genotypes that were isolated rarely. These genoclouds have caused three pandemic waves of disease since the mid-1960s, the most recent of which was imported from East Asia to Europe and Africa in the mid-1990s. Many of the genotypes are escape variants, resulting from positive selection that we attribute to herd immunity. Despite positive selection, most escape variants are less fit than their parents and are lost because of competition and bottlenecks during spread from country to country. Competition between fit genotypes results in dramatic changes in population composition over short time periods.

Transient resonance Raman and density functional theory investigation of the 2-fluorenylnitrenium ion.

We report a transient resonance Raman spectrum for the 2-fluorenylnitrenium ion obtained after photolysis of 2-azidofluorene. The 10 experimental Raman band frequencies of the transient spectrum show very good agreement with the computed frequencies from BPW91/cc-PVDZ density functional theory calculations for the 2-fluorenylnitrenium ion. Our results confirm the assignment of the approximately 460 nm transient absorption band formed after photolysis of 2-azidofluorene in water/acetonitrile or water solution to the singlet ground electronic state 2-fluorenylnitrenium ion. Our study indicates the 2-fluorenylnitrenium has a large degree of iminocyclohexadienyl cation character with significant delocalization of the charge over both phenyl rings of the fluorene moiety. We compare our results for the 2-fluoreneylnitrenium ion to those previously reported for several other arylnitrenium ions.

Biochemical, phenotypic and neurophysiological characterization of a genetic mouse model of RSH/Smith--Lemli--Opitz syndrome.

The RSH/Smith--Lemli--Opitz syndrome (RSH/SLOS) is a human autosomal recessive syndrome characterized by multiple malformations, a distinct behavioral phenotype with autistic features and mental retardation. RSH/SLOS is due to an inborn error of cholesterol biosynthesis caused by mutation of the 3 beta-hydroxysterol Delta(7)-reductase gene. To further our understanding of the developmental and neurological processes that underlie the pathophysiology of this disorder, we have developed a mouse model of RSH/SLOS by disruption of the 3 beta-hydroxysterol Delta(7)-reductase gene. Here we provide the biochemical, phenotypic and neurophysiological characterization of this genetic mouse model. As in human patients, the RSH/SLOS mouse has a marked reduction of serum and tissue cholesterol levels and a marked increase of serum and tissue 7-dehydrocholesterol levels. Phenotypic similarities between this mouse model and the human syndrome include intra-uterine growth retardation, variable craniofacial anomalies including cleft palate, poor feeding with an uncoordinated suck, hypotonia and decreased movement. Neurophysiological studies showed that although the response of frontal cortex neurons to the neurotransmitter gamma-amino-n-butyric acid was normal, the response of these same neurons to glutamate was significantly impaired. This finding provides insight into potential mechanisms underlying the neurological dysfunction seen in this human mental retardation syndrome and suggests that this mouse model will allow the testing of potential therapeutic interventions.

Crypt1, an active Ac-like transposon from the chestnut blight fungus, Cryphonectria parasitica.

A moderately repetitive element was identified previously in the nuclear genome of the chestnut blight fungus, Cryphonectria parasitica, and has been used as a probe for population studies of the fungus. We report here that the repetitive element is a class II transposon of the hAT family of Activator (Ac)-like transposable elements. The element, named Crypt1, has a size of 3563 bp, including 21-bp terminal inverted repeats. A unique 8-bp direct repeat sequence flanking Crypt1 was identified in each of three clones examined. A single large ORF with the potential to encode a putative transposase of 946 amino acid residues was deduced from the sequence of Crypt1. Based on amino acid sequence alignments, Crypt1 is most closely related to other Ac-like transposons of filamentous ascomycetes. A single transcript of approximately 3.0 kb was identified by Northern hybridization experiments from Crypt1-containing isolates, suggesting that Crypt1 is an active element. An isolate containing a single, possibly defective, copy of Crypt1 was identified in C. parasitica isolates from China; no Crypt1 transcript was identified in this isolate. Transposition of Crypt1 was inferred from Southern and inverse PCR analyses of C. parasitica isolates maintained in the laboratory, but transposition appears to be a rare event.

Combined blockade of endothelin-1 and thromboxane A(2) receptors against postischaemic contractile dysfunction in rat hearts.

1. Endothelin-1 (ET-1) may play a role in myocardial ischaemia/reperfusion injury because both the release and vasoconstrictor effect of ET-1 are increased after ischaemia. Since the increased vasoconstrictor effect of ET-1 can be mediated by ET-1-induced release of thromboxane A(2) (TXA(2)), the aim of this study was to test whether combined blockade of ET and TXA(2) receptors protects the coronary flow, contractile performance, and cardiac energy metabolism during ischaemia and reperfusion. 2. Bosentan (antagonist for ET(A) and ET(B) receptors, 1 microM based on concentration-response curves of ET-1), SQ 30,741 (antagonist of TXA(2) receptors, 0.1 microM), or the combination thereof was administered to isolated perfused rat hearts undergoing 15 min of global ischaemia and 60 min of reperfusion. 3. Neither bosentan or SQ 30,741 alone, nor the combination thereof, improved the incomplete postischaemic recovery of coronary flow, left ventricular developed pressure, phosphocreatine, or ATP. However, they attenuated ischaemia-induced acidosis but this did not translate into a measurable effect on haemodynamic or metabolic variables. 4. Thus, combined blockade of ET and TXA(2) receptors does not protect the coronary flow, contractile performance, and cardiac energy metabolism during ischaemia and reperfusion in isolated perfused rat hearts. This finding suggests that neither ET-1 nor ET-1-induced release of TXA(2) play a major role in the postischaemic recovery of the cardiac contractile function and energy metabolism.

Vasorelaxing effect of the potassium (K+)-channel opener pinacidil in isolated porcine ciliary arteries.

PURPOSE: The aim of this study was to investigate the vasorelaxing properties of pinacidil (a potassium-channel-opener) in isolated porcine ciliary arteries. METHODS: Isometric contractions of isolated porcine ciliary arteries were measured with a myograph system. The vessels were first precontracted with the thromboxane A2 analog U 46619 (0.1 microM), and then exposed, in a cumulative manner, to increasing concentrations of pinacidil (1 nM-100 microM). Relaxations have been expressed in percent of the maximal contraction evoked by U 46619 (0.1 microM). RESULTS: Pinacidil showed a pronounced concentration-dependent relaxing effect in isolated porcine ciliary arteries. The difference between pinacidil and time-control experiments was significant (100 microM: 112 +/- 2% vs. 18 +/- 6%, P < 0.0001). The half-maximal concentration (pD50) value was 5.3 +/- 0.1 (-log M). CONCLUSIONS: The present study indicates that pinacidil has marked vasorelaxing properties in isolated porcine ciliary arteries.
