Establishment of induced pluripotent stem cell line TRNDi045-A-38 carrying homozygous DOK7-related Congenital Myasthenia patient-mutation knock-in variant from parental KOLF2.1J.

DOK7-related Congenital Myasthenic Syndrome (CMS) is a rare genetic neuromuscular junction disorder. This is one of the most common of the recessive forms of CMS, often presenting with more static proximal weakness (hence also referred to as limb girdle CMS). Whole-genome sequencing of affected patients implicates frameshift duplication mutations in DOK7 as drivers of impaired neuromuscular-junction signaling. In this study, we generated a human induced pluripotent stem cell (hiPSC) line TRNDi045-A-38 from the KOLF2.1J reference line, engineered to carry homozygous DOK7 c.1124_1127dupTGCC mutation knock-in using CRISPR/Cas9. This iPSC line could be used for in vitro disease modeling to study disease pathophysiology and for therapeutic development.

Quantum-sized carbon dots for bright and colorful photoluminescence.

We report that nanoscale carbon particles (carbon dots) upon simple surface passivation are strongly photoluminescent in both solution and the solid state. The luminescence emission of the carbon dots is stable against photobleaching, and there is no blinking effect. These strongly emissive carbon dots may find applications similar to or beyond those of their widely pursued silicon counterparts.

Nurse-led management of carpal tunnel syndrome: an audit of outcomes and impact on waiting times.

INTRODUCTION: This article describes the outcome of a nurse-led service developed to manage patients referred with a presumptive diagnosis of carpal tunnel syndrome. PATIENTS AND METHODS: We developed a rapid-access service in response to unacceptable waiting times for patients with carpal tunnel syndrome. The service was developed around the role of a nurse practitioner providing a single practitioner pathway from first clinic appointment, through surgery to discharge. RESULTS: Waiting times improved considerably whilst the standard and quality of care was maintained. CONCLUSIONS: There is a role for nurses to perform certain surgical procedures within a well-defined environment.

Direct conjugation of semiconductor nanoparticles with proteins.

Nanocrystalline CdS particles directly conjugated with bovine serum albumin (BSA) protein were prepared by applying the supercritical fluid processing technique, rapid expansion of a supercritical solution into a liquid solvent. The direct conjugation takes advantage of the unique features of the process for nanoparticle formation. The BSA-conjugated CdS nanoparticles in stable aqueous suspension or in the solid state were characterized by using microscopy, X-ray diffraction, and optical spectroscopy methods. The results show that well-dispersed CdS nanoparticles are coated with BSA in a core-shell-like arrangement and that the protein species associated with the nanoparticles remain functional according to the modified Lowry assay. These BSA-conjugated CdS nanoparticles are also strongly luminescent, with the luminescence spectrum contributed to primarily by the exciton emission.

Polymeric nanoparticles from rapid expansion of supercritical fluid solution.

In this concept paper we highlight applications of supercritical fluid technology in particle formation and production, especially some recent advances in the rapid expansion of supercritical solutions (RESS) processing technique. We also highlight the simple but significant modification to the traditional RESS by using a liquid solvent or solution at the receiving end of the supercritical solution expansion, or the rapid expansion of a supercritical solution into a liquid solvent (RESOLV), and applications of the technique to the preparation of nanoparticles. In particular, successes and challenges in the use of RESOLV for nanoscale (<100 nm) polymeric particles and the subsequent protection of the suspended nanoparticles from agglomeration are discussed.

Supercritical fluid technology for enhanced drug delivery.

The rapid advances in the development of formulation and delivery systems based on micron-sized and nanoscale drug particles will create significant benefits to the pharmaceutical industry. Complementary to traditional methods, supercritical fluid techniques have found many useful, and sometimes unique, applications in the production and processing of drug particles. In this article background information is provided on a variety of supercritical fluid techniques relevant to drug formulation and delivery, recent advances and novel applications are highlighted, and the successful development of a new supercritical fluid rapid expansion technique for producing exclusively nanoscale drug particles will be discussed. Challenges and opportunities for further development and future applications are also reviewed.

Nanosizing drug particles in supercritical fluid processing.

The supercritical fluid-processing technique, rapid expansion of a supercritical solution into a liquid solvent (RESOLV), was applied to the nanosizing of water-insoluble drug particles. Selected for demonstration were antiinflammatory drugs Ibuprofen and Naproxen, for which CO2 and CO2-cosolvent systems were used. The RESOLV process produces exclusively nanoscale (less than 100 nm) Ibuprofen and Naproxen particles suspended in aqueous solutions, and the aqueous suspensions of the drug nanoparticles are protected from particle agglomeration and precipitation by using common polymeric and oligomeric stabilization agents.
