Three 1-thio-beta-D-glucopyranosides from the seeds of Afrostyrax lepidophyllus Mildbr.

Three new 1-thioglycosides namely methylthiomethyl 1-thio-beta-D-glucopyranoside (Afrostyraxthioside A), methylsulfonylmethyl 1-thio-beta-D-glucopyranoside (Afrostyraxthioside B) and methylsulfonylmethylthiomethyl 1-thio-beta-D-glucopyranoside (Afrostyraxthioside C) were isolated from the seeds of Afrostyrax lepidophyllus Mildbr. Their structures were mainly elucidated by using one- and two-dimensional NMR and mass spectroscopies and also by an efficient one-step synthesis. Moreover, Afrostyraxthiosides A, B and C constitute a new subclass of 1-thioglycosides isolated from natural sources.

Structure-activity relationship of natural and synthetic coumarins inhibiting the multidrug transporter P-glycoprotein.

A set of 32 natural and synthetic coumarins were tested in order to evaluate their activity on human leukemic cells (K562/R7) overexpressing P-glycoprotein (P-gp). Their ability to reduce the P-gp-mediated drug efflux of daunorubicin out of cells was evaluated at 10 microM. Four natural compounds, previously isolated from Calophyllum dispar (Clusiaceae) and substituted by a common alpha-(hydroxyisopropyl)dihydrofuran moiety, exhibited a significant inhibitory effect on P-gp when compared to the positive control cyclosporin A. A 3D-quantitative structure-activity relationship (3D-QSAR) analysis of the coumarins was performed using the biological results obtained by comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA) of P-gp. Results showed a favorable electrostatic and steric volume, like the alpha-(hydroxyisopropyl)dihydrofuran moiety, beside C(5)-C(6) or C(7)-C(8) positions. In addition, the analysis revealed an important hydrophobic, neutral charge group, like phenyl, in position C(4) on the coumarinic ring.

Single-scan quantitative T2* methods with susceptibility artifact reduction.

Two imaging methods, MSSAVE (Multiple echo SubSlice AVEraging imaging), based on sub-slice averaging and MGESEPI (Multiple echo Gradient-Echo Slice-Excitation Profile Imaging), based on over-sampling in the slice direction, are proposed for single-scan quantitative T(2)* evaluation with susceptibility artifact compensation. Their potentials in terms of sensitivity, minimum performance time, susceptibility artifact reduction and T(2)* quantitation quality, were compared with existing single-scan methods such as classical FLASH two- or three-dimensional or z-shimmed methods both in vitro and in vivo in normal rat brain. MGESEPI offered good quality T(2)* maps nearly free of artifacts but required a long acquisition time. MSSAVE was faster, but at the expense of reduced artifact compensation and the achievable T(2)* quantitation quality.

New cytotoxic guttiferone analogues from Garcinia virgata from New Caledonia.

There are thirteen endemic species belonging to the genus Garcinia in New Caledonia. Among them, G. virgata is an evergreen tree mainly growing in the rain forests of this island. Fractionation of the cyclohexane extract of the stem bark of this plant produced the known benzophenones guttiferone E and xanthochymol, together with two new guttiferone analogues, namely guttiferones I and J. The structures of these benzophenones were mainly elucidated using 1D and 2D NMR spectroscopy. Compounds and were weakly cytotoxic on the KB cell line with IC50 values of 4.70 and 5.0 microg/mL respectively.

Synthesis and antifungal activity of new thienyl and aryl conazoles.

Recent studies reported that an first generation azole (tioconazole) was active against Candida glabrata petite mutants, a fluconazole- and voriconazole- resistant strain of fungi characterized as most azole resistant yeast by an overexpression of the efflux pumps. Therefore, monosubstituted 1-[2-(2,4-dichlorophenyl)ethyl]-1H-imidazoles differing from tioconazole by the nature of the linker and of the aromatic ring in their side-chain were synthesized and evaluated against the mutant and the wild-type strain of C. glabrata. New 2-aryl-1-azolyl-3-thienylbutan-2-ols were then designed and synthesized, and their antifungal activity was evaluated against both strains of C. glabrata and two other major human pathogenic fungi, C. albicans and Aspergillus fumigatus. These new compounds exhibited a broad spectrum activity, as well as good efficiency against the petite mutant, suggesting that they may overcome the increased expression of the efflux pumps usually observed in clinical yeast isolates resistant to current azoles.

Electrolytic electrospray ionization mass spectrometry of quaterthiophene-bridged bisporphyrins: beyond the identification tool.

Several quaterthiophene-bridged bisporphyrins were analyzed by electrospray ionization mass spectrometry (ESI-MS). The active centers of these molecular assemblies are two porphyrins moieties complexed (Z) or not (H) with a metal ion, typically Zn(2+), and the spacer is a quaterthiophene. The two end-groups were chemically linked to the quaterthiophene spacers by (i) a C--C single bond, (ii) a trans double bond or (iii) a triple bond. The formation of charged species either by protonation ([M + H](+) and [M + 2H](2+)) or electron(s) loss (M(+) and M(2+)), account for the occurrence of electrochemical processes in the basic operation of an electrospray source acting in a non-aqueous solvent. The nature of the observed charged species is correlated with the electro-oxidation properties and proton production by electro-oxidation of residual water. The occurence of these electrochemical reaction is proposed when the electroactivity of the electrosprayed substrates is not sufficient to support the current demand of the ESI source. In this way, the results obtained from the analysed series suggest the occurrence of such a process when the interfacial potential of the metal capillary reaches a value of 0.75 V vs Ag/AgCl. The results of theoretical calculations confirm the importance of the ionization energy with regard to the protonation energy in the course of the ionization reaction. The structural differences at the porphyrin-linker junctions lead to significantly smaller ionization energy in the case of the trans double bond. The MS observation of discharged dimers from molecular assemblies, including two complexed porphyrins ZZ or two free bases HH as end-group and a triple bond as the quaterthiophene-bisporphyrin junction, indicates together with molecular modelling (carried out at the semi-empirical PM3 level), that the planar and symmetric structures favour stacking.

Antileishmanial and antifungal activities of xanthanolides isolated from Xanthium macrocarpum.

Seven xanthanolides, xanthinosin, xanthatin, 4-hydroxyxanthinosin, xanthinin, 4-epiisoxanthanol, 4-epixanthanol, 2-hydroxyxanthinosin and 4-oxobedfordia acid, were isolated from the fruits of Xanthium macrocarpum. A valuation of the antifungal activity of these xanthanolides against Candida albicans, Candida glabrata and Aspergillus fumigatus and of their antileishmanial activity against Leishmania infantum and Leishmania mexicana is presented.

Antimalarial xanthones from Calophyllum caledonicum and Garcinia vieillardii.

The antimalarial activity of 22 xanthones against chloroquino-resistant strains of Plasmodium falciparum was evaluated. Natural caloxanthone C (1), demethylcalabaxanthone (2), calothwaitesixanthone (3), calozeyloxanthone (4), dombakinaxanthone (5), macluraxanthone (6), and 6-deoxy-gamma-mangostin (7) were isolated from Calophyllum caledonicum. 1,6-dihydroxyxanthone (8), pancixanthone A (9), isocudraniaxanthone B (10), isocudraniaxanthone A (11), 2-deprenylrheediaxanthone B (12) and 1,4,5-trihydroxyxanthone (13) were isolated from Garcinia vieillardii. Moreover, synthetic compounds (14-22) are analogues or intermediates of xanthones purified from Calophyllum caledonicum (Oger J.M., Morel C., Helesbeux J.J., Litaudon M., Seraphin D., Dartiguelongue C., Larcher G., Richomme P., Duval O. 2003. The most potent xanthones (2), (3) and (7) (IC50 = c.a. 1.0 microg/mL) are 1,3,7 trioxygenated and prenylated on the positions 2 and 8.

Detailed physicochemical characterization of the 2S storage protein from rape (Brassica napus L.).

Chromatographic, chemical, and spectroscopic techniques were used to characterize the physicochemical properties of napin purified by preparative chromatography. The molar extinction coefficient was determined (epsilon = 0.56), and static and dynamic light scattering measurements enabled the average molecular weight (M(w) = 13919), the second virial coefficient (A(2) = 23.95 x 10(-)(5) mol cm(3) g(-)(2)), and the hydrodynamic radius (R(H) = 1.98 nm) to be determined. No conformational changes were observed by fluorescence and circular dichroism measurements in different buffers at pH 3, 4.6, 7, and 12, confirming the high pH stability of this protein. From MALDI-TOF analysis and after enzymatic digestion, it was found that this purified sample, extracted from the rapeseed variety Express, contained mainly isoform 2SS3_BRANA.

Prenylated xanthones and tocotrienols from Garcinia virgata.

Two xanthones, namely virgataxanthone A and B, have been isolated from the stem bark of Garcinia virgata, together with two formylated tocotrienols and the known delta-tocotrienol, griffipavixanthone and 2,6-dihydroxy-4-methoxybenzophenone (cotoin). Their structures were mainly established using one and two-dimensional NMR and mass spectroscopies. When sufficient material was available, the antioxidant activities of the crude extracts as well as the isolated compounds were evaluated.

Antioxidant xanthones from Garcinia vieillardii.

Two new xanthones, 6-O-methyl-2-deprenylrheediaxanthone B (1) and vieillardixanthone (2), were isolated from the stem bark of Garcinia vieillardii, as were four known compounds (4-7). The structures of 1 and 2 were determined by means of spectroscopic analysis and chemical derivatization. Each isolate was tested for its antioxidant properties based on a scavenging activity study using the stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical.

Investigation of the antifungal activity of caledonixanthone E and other xanthones against Aspergillus fumigatus.

Among the different xanthones previously isolated from the stem bark of Calophyllum caledonicum, caledonixanthone E presented the strongest activity (MIC (80) = 8 microg/mL) in acidic conditions (pH 3) against the human pathogenic fungus Aspergillus fumigatus. Phase-contrast microscopy studies suggested the assembly or synthesis of cell wall components as the target of the drug. Moreover, the use of fluorescent lectins further supported an impact of caledonixanthone E on the synthesis of chitin, the major structural polysaccharide of the fungal wall. These results suggest that caledonixanthone E may be an interesting model for the design of new antifungal drugs.

Photomechanical control of the electronic properties of linear pi-conjugated systems.

Photodynamic molecular architectures have been synthesized by covalent fixation of a photoisomerizable dimethylazobenzene group at two fixed points of conformationally flexible pi-conjugated quater- and sexithiophene chains. Theoretical geometry optimization shows, in excellent agreement with crystallographic structures, that the mode of fixation of the azo group plays a determining role in the geometry of the final molecular architecture and on its ability to perform the expected photoinduced molecular motion. Thus, covalent fixation of meta-dimethylazobenzene on a quaterthiophene chain results in a conformationally locked system in which photoisomerization of the azo group is hindered. However, the experimental results of optical, (1)H NMR spectroscopic, and electrochemical investigations show that when an azobenzene group is connected at the para positions of the phenyl rings, trans-to-cis photoisomerization of the azo group induces a conformational transition and dimensional changes in the underlying pi-conjugated oligothiophene chain. These experimental results unequivocally show that the photochemically induced geometrical changes produce in turn an increase in the HOMO level and a narrowing of the HOMO-LUMO gap. This therefore provides the first evidence of photomechanical control of the electronic properties of linear pi-conjugated systems.

Effect of local molecular structure on the chain-length dependence of the electronic properties of thiophene-based pi-conjugated systems.

Three series of thiophene-based pi-conjugated oligomers built with different combinations of thiophene cycles and double bonds have been synthesized and characterized. The analysis of the chain length dependence of the electronic, electrochemical, and vibrational properties of the three series of oligomers has been carried out using cyclic voltammetry, UV-vis, IR, and Raman spectroscopies. These various investigations provide consistent results showing that incorporation of ethylenic linkages in an oligothiophene structure leads to a faster decrease of the HOMO-LUMO gap with chain extension due to the combined effects of enhanced planarity and lower overall aromatic character of the system. Although the incorporation of two consecutive double bonds in the system leads to a stabilization of the dicationic state, this structural modification does not produce the expected further decrease of the HOMO-LUMO gap at large chain extension. This phenomenon is discussed on the basis of an interplay between aromaticity and bond length alternation.

Photomechanical actuation and manipulation of the electronic properties of linear pi-conjugated systems.

A photodynamic molecular architecture has been synthesized by covalent fixation of a photoisomerizable azobenzene group at two fixed points of a conformationally flexible pi-conjugated quaterthiophene chain. The crystallographic structure shows that the two systems lie in parallel planes with a short interplane distance. Theoretical modelization and experimental analysis by 1H NMR and cyclic voltammetry unequivocally show that trans to cis photoisomerization of the azobenzene group induces dimensional and conformational changes in the underlying pi-conjugated system. These geometrical changes produce, in turn, an increase of the HOMO level and a narrowing of the HOMO-LUMO gap, thus providing a first example of photomechanical control of the electronic properties of the pi-conjugated system.
