[Review of bioactivities of natural cycloartane triterpenoids].

Cycloartane triterpenoids, which exist widely in nature, are mainly distributed in Astragalus (Leguminosae) species, Trib. Cimicifuga (Ranunculaceae) and Thalictrium (Ranunculaceae) species and possess various bioactivities. Along with the development of isolation techniques of phytochemistry, more and more this kind of compounds are isolated and identified. However, bioactivity researches on the compounds are relatively lagged behind. Most researches are still in screening level, deficient in mechanism elucidation, short of action proven in vivo and SAR analysis. The author summarized the bioactivity of this kind of compounds from all aspects: anti-tumor, anti-virus, antibacterial, anti-inflammation, immune-regulatory, cardiovascular system, hepatic protection and so forth. This will be benefit for the further research and development of the compounds.

[Identification of antiviral activity of Toddalia asiatica against influenza type A virus].

OBJECTIVE: To identify antiviral activity of Toddalia asiatica against influenza virus type A in vitro. METHOD: More than two hundred Chinese medicinal herb extracts were screened for antiviral activities against influenza A/PR/8/34 (H1N1) virus using 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay for virus induced cytopathic effect (CPE) in a primary screening. Positive samples were picked up and were subjected to quantitative real-time polymerase chain reaction (PCR) to quantify reduction of H1N1 virus genomic RNA. RESULT: Toddalia asiatica showed potent antiviral activities against H1N1 virus, with 50% effective concentration (EC50) value of 4.7 mg x L(-1) in MTS assay and 0.9 mg x L(-1) in quantitative PCR assay respectively. The cytotoxicity test of Toddalia asiatica generated a CC50 value of 187.2 mg x L(-1) and a selective index (SI) larger than 206 in quantitative PCR. Although the best antiviral activity of Toddalia asiatica was observed with co-treatment of influenza virus infection, it remained effective even when administrated 24 h before and after the initiation of infection. CONCLUSION: The results suggested that Toddalia asiatica compound extract could be a candidate for anti-H1N1 virus agent in the treatment of influenza.

Differential gene expression profiles in paclitaxel-induced cell cycle arrest and apoptosis in human breast cancer MCF-7 cells.

AIM: To elucidate the molecular mechanism of cell cycle arrest and apoptosis of MCF-7 cells induced by paclitaxel. METHODS: Flow cytometry was used to determine the cell cycle changes of MCF-7 cells upon paclitaxel treatment. Gene expression profiles of MCF-7 cells induced by paclitaxel were obtained by using cDNA microarrays containing 9984 genes and expressed sequence tags (ESTs). RESULTS: Cell cycle analysis showed that 77.8% of cells arrested at G2/M phase and 1.3% of cells underwent apoptosis upon 100 nmol x L(-1) paclitaxel treatment for 24 hours; cDNA microarray results revealed that 27 and 77 genes were differentially expressed upon 12.5 nmol x L(-1) (IC50) and 100 nmol x L(-1) paclitaxel treatment, respectively. CONCLUSION: Paclitaxel stabilized microtubules and caused G2/M cell cycle arrest and apoptotic cell death in a concentration-dependent manner, which is associated with the regulation of selected genes related to microtubule assembly and cytoskeleton, cell cycle regulation, and DNA repair and apoptosis.

[Utilization of spaceflight technology on medicinal plant study].

Utilization of spaceflight technology will benefit the modernization of Traditional Chinese Medicine study. Many countries, such as USA or Russia, have conducted extensive experimental research with plants in the regenerative life-support system in space stations as well as the research on breeding or chromosomal aberration after spaceflight. The space botany is still in its preliminary stage in our country. Research has mainly been focused on breeding under space-environment. In addition, many experiments have been conducted with the spaceflown plants to investigate the growth, development, biochemical and physiological changes, as well as the inheritance and variation. Little has been done with regard to the facility development to contain the spaceflying seeds and no research has been reported on plant growth in spacecrafts. Medicinal plant study has certain characteristics and advantages in our country where small biological vessels are developed for the spaceflown seeds, which are then distinguished to microgravity group and radiation exposed group with analytical instrument. However, research has been carried out in medicinal plant breeding or inheritance. In future research, more effort should be directed to the study of medicinal plants as an important link of a future biological life support system as well as to the health care of astronauts by.
