Attachment state of mind and perceptual processing of emotional stimuli.

This study aimed to investigate the relationship between attachment state of mind and perceptual processing of social and non-social, affective, and neutral material. A total of 57 young adults completed the Adult Attachment Interview (AAI) plus an experimental task in which their perceptual thresholds to different types of pictures were assessed. Significant correlations were found between the AAI dimensions and perceptual thresholds for social stimuli such as social interactions or human faces displaying emotional expressions. As expected, no relationships were found between the AAI and perception of neutral stimuli. The pattern of correlations was especially clear for the dismissing dimension. The results suggest that higher vigilance to social stimuli is related to dismissing attachment tendencies and, to a milder degree, to preoccupied tendencies.

The sulphur oxygenase reductase from Acidianus ambivalens is a multimeric protein containing a low-potential mononuclear non-haem iron centre.

The SOR (sulphur oxygenase reductase) is the initial enzyme in the sulphur-oxidation pathway of Acidianus ambivalens. Expression of the sor gene in Escherichia coli resulted in active, soluble SOR and in inclusion bodies from which active SOR could be refolded as long as ferric ions were present in the refolding solution. Wild-type, recombinant and refolded SOR possessed indistinguishable properties. Conformational stability studies showed that the apparent unfolding free energy in water is approx. 5 kcal x mol(-1) (1 kcal=4.184 kJ), at pH 7. The analysis of the quaternary structures showed a ball-shaped assembly with a central hollow core probably consisting of 24 subunits in a 432 symmetry. The subunits form homodimers as the building blocks of the holoenzyme. Iron was found in the wild-type enzyme at a stoichiometry of one iron atom/subunit. EPR spectroscopy of the colourless SOR resulted in a single isotropic signal at g=4.3, characteristic of high-spin ferric iron. The signal disappeared upon reduction with dithionite or incubation with sulphur at elevated temperature. Thus both EPR and chemical analysis indicate the presence of a mononuclear iron centre, which has a reduction potential of -268 mV at pH 6.5. Protein database inspection identified four SOR protein homologues, but no other significant similarities. The spectroscopic data and the sequence comparison led to the proposal that the Acidianus ambivalens SOR typifies a new type of non-haem iron enzyme containing a mononuclear iron centre co-ordinated by carboxylate and/or histidine ligands.

Attachment representations and characteristics of friendship relations during adolescence.

Attachment theory proposes that experiences with the primary caregivers are an important basis for the development of close social relationships outside the parent-child relationship. This study examined the association between representations of attachment, as assessed with the Adult Attachment Interview (AAI), representations of friendship and peer relations, as assessed with an interview in a sample of 43 adolescents. Secure attachment representations were significantly related to interview-based assessments of close friendships, friendship concept, integration in a peer group, and emotion regulation within close friendships. Attachment experiences reported during the AAI, their integration, and their coherency were related to friendship quality and friendship concept. Results show the close associations between attachment representations and friendship relationships during adolescence. The associations between peer relations and attachment representations differed depending on whether an interview approach or a questionnaire approach was used.

An improved method for checking HTS/uHTS liquid-handling systems.

An efficient method is presented to determine precision and accuracy of multichannel liquid-handling systems under conditions near to application. The method consists of gravimetrical determination of accuracy and optical determination of precision based on the dilution of absorbing and fluorescent dye solutions in microplates. Mean delivery volume per well can be determined with precision better than a 0.04% coefficient of variation (CV). Optical signal precision, CV(S), is improved by multiwavelength measurements. Precision of absorbance measurement yields a better resolution than precision of fluorescence measurement (0.3% and 1.5%, respectively), indicating that absorbance measurements should be preferred. From CV(S), an upper bound of the precision of the volumes delivered is derived. Method performance is demonstrated with the dispenser CyBi-Drop and the pipettor CyBi-Well using different ejection principles; with commonly used fluids; with 96-, 384-, and 1536-well microplates; and with photometric and fluorometric indicators. Precision of the volumes delivered, as obtained with optimized methods, all plate formats, and both devices, is better than 2% CV with 2 microL set volume and about 1% CV with higher set volumes.

Regulation of the expression of gas vesicle genes in Haloferax mediterranei: interaction of the two regulatory proteins GvpD and GvpE.

The gas vesicle formation in Haloferax mediterranei occurs in the stationary growth phase and involves the 14 genes mc-gvpACNO and mc-gvpDEFGHIJKLM. The appearance of the two regulatory proteins GvpD and GvpE, and also of GvpF, was investigated during the growth of H. mediterranei. GvpD was only found during the stationary growth phase, GvpE was present from the late exponential to stationary growth phase, and GvpF was present only during the exponential growth, although the three genes were co-transcribed. The impact of GvpD and GvpE on the activity of the promoter of the mc-gvpACNO gene cluster encoding the gas vesicle structural proteins was analysed in H. volcanii transformants containing the mc-gvpA gene or a fusion of the mcA promoter with the bgaH reading frame encoding a halobacterial beta-galactosidase as reporter. The experiments proved that GvpE is a transcriptional activator, whereas GvpD is involved in the repression. Protein-protein affinity chromatography was used to search for putative binding partners of GvpD and GvpE. Both proteins were synthesized in Escherichia coli as his-tagged proteins, isolated under denaturing conditions and refolded by dialysis against buffers containing decreasing urea and increasing KCl concentrations up to 2.5 M. The Ni-NTA matrix tagged with GvpD-his or GvpE-his was incubated with soluble proteins of gas vesicle producing H. mediterranei cells. A 21 kDa protein was purified using the matrix tagged with GvpD-his which proved to be GvpE by Western analysis. Vice versa, GvpD was purified using the GvpE-his-Ni-NTA matrix. These results strongly suggested that GvpD and GvpE were able to interact and might constitute a regulatory system.

Echocardiographic monitoring during induction of general anesthesia with a miniaturized esophageal probe.

UNLABELLED: Standard transesophageal echocardiography (TEE) does not allow cardiac monitoring during the induction of anesthesia because standard probes would limit the oropharyngeal space and impair mask ventilation and tracheal intubation. We hypothesized that a prototype, miniaturized TEE probe could be safely introduced transnasally in awake patients and that mask ventilation and orotracheal intubation could be performed while continuously monitoring left ventricular (LV) function during the induction of anesthesia. Forty-five patients were studied prospectively. The transnasal TEE probe was introduced through one of the nares and advanced until a transverse plane image of the LV at the level of the papillary muscles was seen. Anesthesia was induced, and the patients were ventilated with a mask that had previously been threaded over the TEE probe via a central perforation. Probe insertion was successful in 12 patients under local anesthesia alone and in an additional 31 patients with a combination of local anesthesia and mild sedation. In two cases, probe placement was unsuccessful. Overall, hemodynamic variables did not change significantly during insertion. No case of significant mucosal bleeding was seen. In one patient, regurgitation of gastric contents occurred without affecting the perioperative outcome. The two-dimensional echocardiogram image quality of the LV during the induction of anesthesia was good or acceptable in 95% of patients. We conclude that transnasal TEE can effectively be used for cardiac monitoring during the induction of general anesthesia. IMPLICATIONS: This study demonstrates that it is feasible and generally safe to introduce a miniaturized transesophageal echocardiography probe transnasally in awake cardiac risk patients to monitor cardiac performance during the induction of general anesthesia.

2p Photoionization of atomic Ni: a comparison with Ni metal and NiO photoionization.

The photoabsorption spectrum of the 2p-3d resonances and the 2p photoelectron spectrum of atomic Ni were investigated both experimentally and theoretically. The analysis of the spectra takes into account that at the evaporation temperature of the metal at about 1800 K the fine structure states 3d(8) 4s(2) 3F and 3d(9) 4s 3D of both configurations 3d(8) 4s(2) and 3d(9) 4s are populated. The population of these two configurations offers the unique possibility for a comparison with the corresponding spectra in the condensed phase (Ni metal and NiO) where current configuration interaction calculations use the mixture of the configurations (3d,4s)(10). By using our theoretical description of the 2p photoionization new insight into these condensed phase spectra is found.

Stability of attachment representations during adolescence: the influence of ego-identity status.

The purpose of this study was to examine two core assumptions of attachment theory. First, internal working models of attachment should increase in stability during the course of development. Second, attachment is related to the adaptive solution of stage-salient issues, in adolescence, specifically to identity formation. Adolescence as a developmental period of change and reorganization might lead to changes in attachment representations by re-evaluating one's life experiences in the course of general identity development. In a longitudinal study, the stability of attachment representations (assessed by the Adult Attachment Interview) between ages 16 and 18 was examined. In addition, the concurrent and longitudinal associations of identity status at age 16 with attachment representations at ages 16 and 18 were tested. Results showed high stability of attachment representation during that age period. Secure attachment representation was positively associated with the identity status achievement, whereas a dismissing status was positively associated with identity diffusion. Longitudinally, identity diffusion at age 16 predicted attachment representation at age 18 even after controlling for earlier attachment representation.

Regulation of gas vesicle formation in halophilic archaea.

The halophilic archaea Halobacterium salinarum and Haloferax mediterranei produce gas vesicles depending on the growth phase and on environmental factors such as light, salt, or oxygen. Fourteen different gvp genes (gvpACNO and gvpDEFGHIJKLM) are involved in their formation, and the regulation of gvp gene expression occurs at the transcriptional and translational level. Haloferax volcanii offers a clean genetic background for the functional analysis of gas vesicle genes by transformation experiments. Such experiments show that the promoter of the gvpA gene encoding the major gas vesicle structural protein is activated by the endogenous basic leucine-zipper protein GvpE. On the other hand, the GvpD protein, which contains a p-loop motif, is involved either directly or indirectly in the repression of the gvpA promoter activity. Eight of the fourteen p-gvp genes (p-gvpAO and p-gvpFGJKLM) enable gas vesicle formation in Hf. volcanii transformants and thus constitute the minimal p-vac region.
