[Preparation, identification and application of anti-P450scc antibody].

AIM: To prepare rabbit anti-rat P450scc antibody, and to detect the expression of rP450scc in rat brain tissue and nerve cells. METHODS: An eight-branched polypeptide, rP450scc-16 was synthesized using solid phase synthesis (Fmoc) method. A Newzealand rabbit was immunized with rP450scc-16 and the serum was separated from the whole blood 5 days after the last immunization. The titer and specificity of the antiserum were evaluated, and the expression of rP450scc in normal rat brain and primary rat astrocytes was detected by using ELISA, Western blot and immunocytochemcal staining. RESULTS: The titer of the antiserum was 1:6,400. Western blot analysis showed that the rP450scc protein in rat brain, testis and adrenal gland homogenate was recognized by the antiserum as a single band at M(r) being 50,000, which indicated a high specificity of the antiserum. Hypothalamus, cerebral cortex, and hippocampus of rat brain and the cytoplasm of cultured rat astrocytes were positively stained by the antiserum. CONCLUSION: A highly specific anti-rP450scc antibody was prepared. The antibody can be used to study the expression and distribution of rP450scc in rat brain.

[Three-dimensional breast non-contact measurement].

OBJECTIVE: To develop a system for three-dimensional (3D) non-contact measurement of breast using computer-aided projection fringe measurement (CAPFM). METHODS: A system of 3D non-contact breast measurement was developed based on CAPFM and used for measurement of a plastic model mimicking human breast. RESULTS: The CAPFM system was capable of non-contact 3D measurement with rapid data collection (within 0.5 s) and measurement (no longer than -3 min). This system was highly automatic, and with software-controlled measurement, the sensitivity was as high as 0.6 mm (for linear measurement) and 1 ml (for volume measurement), to well meet the clinical requirements. The measurement results could be stored and expressed in numerical values or represented in vivid 2D figures and 3D images, which facilitated subsequent studies. CONCLUSION: The 3D non-contact breast measurement system on the basis of CAPFM can be helpful for medical assessment of the breast.

[Myocardial protective effect of puerarin injection in children with severe pneumonia].

OBJECTIVE: To investigate the myocardial protective effect of puerarin injection (PI) in children with severe pneumonia. METHODS: One hundred children with pneumonia enrolled were divided into two groups according to their condition of illness. Forty-eight children of mild condition in the common group (CG) were treated with conventional treatment and 52 children of serious condition in the severe group (SG) were treated with conventional treatment with additional PI or fructose-1,6-disphosphate (FDP) for myocardial protection. The parameters of serum myocardial zymogram, ECG and cardiac blood flow were measured and the effects of PI and FDP in patients of serious condition were compared. RESULTS: Change of serum myocardial zymographic parameters was indirectly proportional to the severity of childrens condition. The reduction of myocardial contractive function was demonstrated by the change of cardiac blood flow parameters in Ultrasound Doppler examination (P < 0.01). Both PI and FDP had the effect in protecting myocardium and improving heart function, showing insignificant difference between their efficacy (P > 0.05), but PI had less stimulation to vessels and without any obvious adverse reaction. CONCLUSION: Severe pneumonia in children can lead to myocardial damage and heart failure, and early applying of myocardial protective measure can significantly improve the prognosis of patient. Myocardial zymographic parameters, especially the CK-MB, are sensitive and important indexes for detecting myocardial injury. PI has effects in protecting myocardium and improving heart function, and therefore, is suitable for spreading and applying in clinical practice.

[A study of making use of computer-aided projection fringe system to measure breast dimension].

OBJECTIVE: To measure breast basic dimension by using computer-aided projection fringe system. METHODS: A system has been developed for measuring breast basic dimension based on computer-aided projection fringe measurement and programming software. Plastic manikins breast's SN-N (sternal notch to nipple distance), N-ML (nipple to midline distance), N-N (internipple distance), MBW (base width of breast) and N-IMF (nipple to inframammary fold distance) are measured with this system. At the same time, these items are also measured with routine ruler. RESULTS: This study indicate that the system has some merits: (1) non-touching measurement; (2) it is very rapid, the patient measured need hold his breath only 0.5 second, and all the time it takes is about 2.5 minutes; (3) the measurement's sensitivity is as high as to 0.6 mm, which meets the clinic requirement entirely; (4) the measurement's accuracy of the system is not significantly when comparing to the routine ruler's. CONCLUSIONS: Computer-aided projection fringe system for measuring breast basic dimension is feasible and advanced.

[Roles of bryophyte in nutrient cycling in dark coniferous forest of Changbai Mountains].

This paper measured the nutrient contents of five main bryophytes in the dark coniferous forest of Changbai Mountains, and estimated their values per unit area. The results showed that bryophyte had a nitrogen content 54.371 kg x hm(-2), being 12.22% of total nitrogen content of arbor, shrub and herb, and its potassium and phosphorous content was 12.042 kg x hm(-2) and 31.679 kg x hm(-2), occupying 5.63% and 70.57% of the total, respectively. The phosphorous content of bryophyte was higher than that of arbor, and 2.4 and 792 times as much as that of herb and shrub, respectively, indicating that bryophyte was an important phosphorous pool. Soil nitrogen and potassium content under different coverage of bryophyte had little difference, while soil phosphorous content was significantly different. The soil with no bryophyte coverage had the highest phosphorous content (0.419 +/- 0.023 g x kg(-1)), while that with 80%-100% bryophyte coverage had the lowest one (0.346 +/- 0.017 g x kg(-1)), suggesting that soil phosphorous content was decreased with increasing bryophyte coverage.

[Phototherapeutic keratectomy of diffuse corneal foreign bodies caused by gunpowder explosion].

PURPOSE: To investigate the clinical effects of phototherapeutic keratectomy (PTK) on the treatment of diffuse corneal foreign bodies caused by gunpowder explosion. METHODS: From Dec, 1999 to Dec, 2003, 39 eyes of 25 patients with diffuse foreign bodies on cornea caused by gunpowder explosion were investigated in this study. All foreign bodies located in one third of superficial cornea and conjunctiva. Besides slit-lamp biomicroscope examination and cornea thickness measurement, ultra-sound B, VEP and ERG were used to exclude the possibility of damage of posterior ocular segments. Then the PTK was performed at every 100-150 pulse-interval. In case of damaging to deep cornea, the total pulse was less than 500 and corneal thickness was measured during the operation. After operation, vision acuity, recovery of corneal epithelium, haze, intraocular pressure and ametropia were checked and the patients were followed up for 6 months. RESULTS: Compared with traditional therapies used for diffused corneal foreign bodies, PTK generally took shorter time (10 minutes). It could erase most foreign bodies on cornea and had little injury to normal corneal tissue. Commonly, the corneal epithelial layer recovered completely within one week. The vision acuity after PKT was improved notablely. The mean value of vision acuity at 10th day of post-operation is 0.6 (vs < 0.3 in all patients before operation). There were 38 eyes with gentle hyperopia and 1 eye with moderate hyperopia. The complication of PTK can be controlled during operation or after operation by drugs. CONCLUSION: PTK is a facile, effective, hypo-cost method for the treatment of diffused foreign bodies on cornea damaged by gunpowder explosion.

Humoral immune response induced by oral administration of S. typhimurium containing a DNA vaccine against porcine reproductive and respiratory syndrome virus.

The ORFs-encoded major envelope glycoprotein (GP5) of porcine reproductive and respiratory syndrome virus (PRRSV) is one of the major structural proteins of this virus. In this report, we described the induction of a PRRSV GP5-specific immune response by oral vaccination of mice with eukaryotic expression vectors containing the GP5 gene of PRRSV (pcDNA3-GP5), delived by attenuated Salmonella typhimurium aroA. It demonstrated that oral administration of the transformants resulted in expression of the GP5 transcript in the intestinal epithelium. The level of serum neutralization antibodies to PRRSV was not significantly different between the mice immunized with the transformants and the naked plasmid DNA. But the neutralizing antibody titres in sera of the mice immunized with SL7207/pcDNA3-GP5' and pcDNA3-GP5' (resides 2-25 deletion mutant of GP5) were significantly lower than those immunized with the complete GP5 gene. These results show that oral inoculation of the transformants can induce humoral immune response to PRRSV. The signal peptide of the GP5 protein of PRRSV is associated with the neutralizing epitope of the protein. The attenuated S. typhimurium may be used as a delivery system for oral DNA vaccines containing PRRSV GP5 glycoprotein.

Transgenic nude mouse with ubiquitous green fluorescent protein expression as a host for human tumors.

We report here the development of the transgenic green fluorescent protein (GFP) nude mouse with ubiquitous GFP expression. The GFP nude mouse was obtained by crossing nontransgenic nude mice with the transgenic C57/B6 mouse in which the beta-actin promoter drives GFP expression in essentially all tissues. In crosses between nu/nu GFP male mice and nu/+ GFP female mice, the embryos fluoresced green. Approximately 50% of the offspring of these mice were GFP nude mice. Newborn mice and adult mice fluoresced very bright green and could be detected with a simple blue-light-emitting diode flashlight with a central peak of 470 nm and a bypass emission filter. In the adult mice, the organs all brightly expressed GFP, including the heart, lungs, spleen, pancreas, esophagus, stomach, and duodenum. The following systems were dissected out and shown to have brilliant GFP fluorescence: the entire digestive system from tongue to anus; the male and female reproductive systems; brain and spinal cord; and the circulatory system, including the heart and major arteries and veins. The skinned skeleton highly expressed GFP. Pancreatic islets showed GFP fluorescence. The spleen cells were also GFP positive. Red fluorescent protein (RFP)-expressing human cancer cell lines, including PC-3-RFP prostate cancer, HCT-116-RFP colon cancer, MDA-MB-435-RFP breast cancer, and HT1080-RFP fibrosarcoma were transplanted to the transgenic GFP nude mice. All of these human tumors grew extensively in the transgenic GFP nude mouse. Dual-color fluorescence imaging enabled visualization of human tumor-host interaction by whole-body imaging and at the cellular level in fresh and frozen tissues. The GFP mouse model should greatly expand our knowledge of human tumor-host interaction.

Analysis of coenzyme Q(10) in human plasma by column-switching liquid chromatography.

A new method of determining coenzyme Q10 in human plasma was developed based on column-switching high performance liquid chromatography (HPLC). CoQ10 was quantitatively extracted into 1-propanol with a fast one-step extraction procedure, after centrifugation, the supernatant was cleaned on an octadecyl-bonded silica column and then transferred to reversed-phase column by a column-switching valve. Determination of CoQ10 was performed on a reversed-phase analytical column with ultraviolet detection at 275 nm and the mobile phase containing 10% (v/v) isopropanol in methanol at a flow-rate of 1.5 ml/min. The sensitivity of this method allows the detection of 0.1 microg/ml CoQ10 in plasma (S/N=3). The linearity between the concentration and peak height is from 0.05 to 20 mg/l. The reproducibility (R.S.D.%) of the method is less than 2% (within day) and less than 3% (between day), the average recovery is 100.9 + 2.1%, it takes only 30 min to complete an analysis procedure, suitable for the determination of CoQ10 in human plasma especially for batch analysis in clinical laboratories. Finally, the method was applied to determine the plasma CoQ10 levels in healthy subjects, hyperthyroid and hypothyroid patients.

Cellular dynamics visualized in live cells in vitro and in vivo by differential dual-color nuclear-cytoplasmic fluorescent-protein expression.

We report here the genetic engineering of dual-color fluorescent cells with one color in the nucleus and the other in the cytoplasm that enables real-time nuclear-cytoplasmic dynamics to be visualized in living cells in vivo as well as in vitro. To obtain the dual-color cells, red fluorescent protein (RFP) was expressed in the cytoplasm of HT-1080 human fibrosarcoma cells, and green fluorescent protein (GFP) linked to histone H2B was expressed in the nucleus. Nuclear GFP expression enabled visualization of nuclear dynamics, whereas simultaneous cytoplasmic RFP expression enabled visualization of nuclear cytoplasmic ratios as well as simultaneous cell and nuclear shape changes. Thus, total cellular dynamics can be visualized in the living dual-color cells in real time. The parental HT-1080 and the derived dual-color clones had similar cell proliferation rates, suggesting that expression of GFP and/or RFP does not affect cell cycle progression. The cell cycle position of individual living cells was readily visualized by the nuclear-cytoplasmic ratio and nuclear morphology. Real-time induction of apoptosis was observed by nuclear size changes and progressive nuclear fragmentation. Mitotic cells were visualized by whole-body imaging after injection in the mouse ear. Common carotid artery injection of dual-color cells and a reversible skin flap enabled the external visualization of the dual-color cells in microvessels in the mouse brain where extreme elongation of the cell body as well as the nucleus occurred. Dual-color cells in various positions of the cell cycle were visualized in excised mouse lungs after tail-vein injection of the dual-color cells. In the lung, the dual-color cells were observed frequently juxtaposing their nuclei, suggesting a potential novel form of cell-cell communication. The dual-color cells thus are a useful tool for visualizing living-cell dynamics in vivo as well as in vitro. Drugs that could specifically perturb these processes can now be readily screened in real time in vivo.

Supramolecular self-assembly of conjugated diblock copolymers.

This paper describes the synthesis and characterization of a novel series of copolymers with different lengths of oligo(phenylene vinylene) (OPV) as the rod block, and poly(propylene oxide) as the coil block. Detailed characterization by means of transmission electron microscopy (TEM), atomic force microscopy (AFM), and small-angle neutron scattering (SANS) revealed the strong tendency of these copolymers to self-assemble into cylindrical micelles in solution and as-casted films on a nanometer scale. These micelles have a cylindrical OPV core surrounded by a poly(propylene glycol) (PPG) corona and readily align with each other to form parallel packed structures when mica is used as the substrate. A packing model has been proposed for these cylindrical micelles.

Syngeneic lymph-node-targeting model of green fluorescent protein-expressing Lewis lung carcinoma.

The Lewis lung tumor has been extensively studied in both syngeneic and allogeneic mouse models. However, its metastatic potential and mechanism are poorly understood. The aim of the present study was to develop a highly metastatic lymph-node targeting, imageable model of the Lewis lung carcinoma in a syngeneic host. We report here a syngeneic model of the Lewis lung carcinoma in which the carcinoma cells are labeled with green fluorescent protein (GFP). The tumor cells were transplanted in the dorsal side of the ear of C57-B16 mice in order to give the tumor cells access to the lymphatic system. This model of the Lewis lung carcinoma extensively metastasized to numerous lymph nodes throughout the body of the animal as well as visceral organs, as visualized by fluorescence microscopy using the bright GFP signal. Twenty-one different metastatic sites, including lymph nodes throughout the body, were identified among the cohort of transplanted animals. The data demonstrate a predilection of the Lewis lung carcinoma for lymphatic pathways for metastasis throughout the animal body. The concomitant macrometastases to the visceral organs observed in this model may be remetastasis from the lymph nodes. This model of the Lewis lung carcinoma should be very useful in defining cellular trafficking and targeting mechanisms of metastasis, in particular those involving lymphatic pathways.

Neovascularization and recurrent varicose veins: more histologic and ultrasound evidence.

BACKGROUND: The recurrence of varicose veins is a common and costly consequence of varicose vein surgery. Despite the long history and vast experience of varicose vein surgery, the exact cause of recurrence is still unknown. This study aims to investigate the cause of recurrence further by correlating findings from duplex ultrasound scans, resin casts, and histologic investigation at the recurrence of the saphenofemoral junction. In particular, frequency and neovascularization are evaluated. METHOD: Forty-nine saphenofemoral junctions (SFJs) from 42 patients who presented for re-operation on their varicose veins were examined with duplex ultrasound and physiologic air plethysmography tests before surgery. All patients had reflux at the groin for which surgery was carried out. Specimens taken during surgery were sectioned and stained for conventional histology and immunohistology, and 5 specimens were infused with resin to form a cast of the venous vasculature. RESULTS: All but 3 re-operation specimens (94%) showed multiple vessels at the stump site of the previous SFJ ligation. Neovascular channels of variable size, number, and tortuosity accounted for the ultrasound appearances and reflux to recurrent varicosities in the vast majority of specimens. These new vessels connected to the common femoral vein at the site of the previous SFJ. In 2 incompetent junctions without femoral vein involvement, while small vessels were seen surrounding the femoral stump scar, ultrasound and histology confirmed both neovascular and residual (enlarged collateral) connections from epigastric and pudendal vessels into the thigh. CONCLUSION: Neovascularisation is the major cause for ultrasound-confirmed recurrence of reflux in the groin following varicose vein surgery.
