Development of a high performance liquid chromatographic method for systematic quantitative analysis of chemical constituents in rhubarb.

HPLC methods for the systematic determination of 30 compounds in Rhei Rhizoma (rhubarb) were developed. Using a combination of mobile phase gradient conditions and UV detection at 280 nm, all 30 compounds were separated satisfactorily with low detection limits (0.05-2 microg/ml). The developed methods provided a reliable calibration curve for each compound. By adopting these methods, the determination of 30 compounds in three kinds of rhubarb samples, derived from Rheum tanguticum, R. palmatum and R. officinale, was achieved. The constituent pattern of each rhubarb was clearly characterized through the quantitative composition of 30 major constituents of rhubarb.

Evaluation of variation of acteoside and three major flavonoids in wild and cultivated Scutellaria baicalensis roots by micellar electrokinetic chromatography.

Micellar electrokinetic chromatography (MEKC) conditions were developed to analyze the constituents of Scutellariae Radix (SR) and Scutellaria baicalensis roots. Using the MEKC method, the major flavonoid constituents of baicalin, baicalein and wogonin of wild and cultivated S. baicalensis roots were compared. In a preliminary comparison of electropherogram, one special peak was found in a wild sample but not in a 2-year-cultivated one. The compound corresponding to the peak was isolated and identified as a phenylethanoid glycoside, acteoside, by comparing the 1H- and 13C-NMR spectral data with that of the authentic compound. This is the first time acteoside has been isolated from the Scutellaria genus. It could only be found in SR derived from wild S. baicalensis roots and 4-year-cultivated plants, but not in plant materials cultivated for 3 years. Applying the MEKC method established in this study, rapid and simultaneous determinations of acteoside together with 3 flavonoids in samples were achieved. The method can thus be used for the quality control of SR in a shorter analysis period than HPLC.

[A novel approach to quality evaluation of root of Scutellaria baicalensis by DPPH free radical scavenging].

OBJECTIVE: To develop a simple, reliable approach for evaluating the quality of Huangqin (Scutellaria baicalensis). METHOD: To determine the DPPH free radical scavenging activity and assay of four bioactive components: baicalin, baicalein, wogonin and wogonin-7-O-glucuronide by HPLC. RESULT: The correlative relationship between DPPH free radical scavenging activity and baicalin content was obtained. CONCLUSION: Bioassay of DPPH free radical scavenging activity could be used as one of the methods for quality evaluation of Chinese drug Huangqin.
