Proteins in fluid from the proximal vas deferens of normal fertile and vasectomized men.

Fluids were collected from the proximal vas deferens of 18 normal fertile men and 32 vasectomized men during vasectomy or vasovasostomy, respectively, and the protein concentration and pattern of proteins were then analysed by sodium dodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE). In normal fertile men and vasectomized men there were no significant differences in total protein concentrations between fluids from the left and right vas deferens. The total protein concentration of proximal vas fluid from vasectomized men (82.8 +/- 7.5 mg/ml; mean +/- SEM) was significantly higher than in normal fertile men (36.8 +/- 4.5 mg/ml). In vasectomized men there was no significant correlation between the total protein concentration in vas fluid and the duration of vasectomy. The patterns of protein bands in proximal vas fluid differed from those in seminal plasma. There was no relationship between the pattern of protein bands in vas fluid and the duration of vasectomy. Five major groups of proteins from proximal vas fluid were identified and no new major proteins were found in proximal vas fluid were identified and no new major proteins were found in proximal vas fluid after vasectomy. The percentage of 80K and 67K protein bands in vasectomized men was higher than that in normal fertile men. It is suggested that, after vasectomy, the physiological environment of the proximal vas deferens may be changed. One of these changes may be the higher total protein concentration (especially the 80K and 67K proteins) in vas fluid.

[Clinical study in epidural injection with lappaconitine for post-operative analgesia].

This paper studied the effect and side-effect of Lappaconitine for post-operative analgesia with epidural injection. 50 patients who were operated in the upper abdomen with epidural anesthesia. These patients were randomly divided into 5 groups. The general condition of every group was similar. Group A, B, C was given Lappaconitine 4 ml, 8 ml, 12 ml individually for observation. Group D, E was given 0.9% NS 6 ml and morphine 2 mg individually as control. When the operation was finished the drug was injected into the epidural space with single blind method. The result showed the analgesia of the drug and its effective time was: group E > C > B > A > D. The analgesic effect of Group C was satisfactory. The efficacy and the maintenance time of A, B, C groups were significantly different from that of group D (P < 0.05, P < 0.01). Although the effect of group C was less potent than that of group E (morphine injection), there was no side-effect as morphine had. It suggested that Lappaconitine for post-operative analgesia with epidural injection is effective and safe.

Effect of colchicine on hepatic glycosaminoglycan metabolism in mice infected with Schistosoma japonicum.

AIM: To study the effect of colchicine on hepatic glycosaminoglycan (GAG) metabolism in mice infected with Schistosoma japonicum. METHODS: The amount of GAG was measured in the liver of mice infected with Schistosoma japonicum for 6-16 wk and treated with colchicine. RESULTS: Six weeks after infection, the GAG content of infected mice's livers and uninfected control was 14 +/- 2 micrograms/g liver and 4 +/- 1 microgram/g liver, respectively. The GAG content reached a peak about 6 times of the normal level in the 10th wk after infection, being 56 +/- 9 micrograms/g and 10 +/- 1 microgram/g for infected and uninfected mice respectively, and then declined. Treatment of infected mice with colchicine reduced the total GAG content to 22 +/- 3 micrograms/g liver, but not suppress the GAG content below the normal level in the 10th wk. Microscopic hepatic egg granuloma. CONCLUSION: Colchicine may exerts its GAG reduction effect through the suppression of granuloma formation.

Analysis of spermatozoa from the proximal vas deferens of vasectomized men.

This study assessed the condition of spermatozoa from the proximal vas deferens of men after vasectomy. The fluids of both proximal vas deferens were collected from 67 vasectomized men by cannulating the vas deferens at the time of vasectomy reversal. Selected sperm parameters were analysed after incubation of the spermatozoa for 30 min at 37 degrees C. Sperm concentration in the proximal vas from vasectomized men (16,312 +/- 21,496 million per ml, geometric mean: 7948 +/- 398 million per ml) was significantly higher than that of fertile men and was maintained at a constant level independent of the duration of vas obstruction. The means of sperm motility (36.2 +/- 26.2%), spermatozoa with normal morphology (50.7 +/- 21.7%), sperm viability (53.0 +/- 25.3%) and hypo-osmotic swelling test (HOS-test, 53.9 +/- 21.7%) were statistically lower than the respective values for normal fertile men. There was no significant correlation between the duration of vas obstruction and the above semen parameters. In 46.4% of vas fluids all spermatozoa were immotile and this condition was more common after 3 years of vasectomy. Immotile spermatozoa in the proximal vas fluids at the time of vasectomy reversal may be an important factor for predicting semen quality and fertilizing ability after vasovasostomy. There were no significant differences in the results of sperm-cervical mucus penetration test (CMPT) between spermatozoa from vasectomized and fertile men. Antisperm antibodies on the surface of spermatozoa from the vas of vasectomized men were determined by the immunobead test (IBT; 78.6% for IgG, 32.1% for IgA) and sperm cervical mucus contact test (SCMC, 36.4%).(ABSTRACT TRUNCATED AT 250 WORDS)

[The protective effect of MPEG-SOD on the function of left ventricle during acute hypoxia].

Monomethoxypolyethylene glycol (MPEG) was attached covalently to superoxide dismutase (SOD, EC1.15.1.1). The molecular weight of MPEG-SOD was about 7.0 x 10(5) Dalton determined by Fast Protein Liquid Chromatography (FPLC with Sepharose 6 HR10/30). MPEG-SOD exhibited a sharply enhanced serum half life (more than 30 h) than that of the native SOD (6-10 min). To determine the role of MPEG-SOD in acute hypoxia-induced injury of left ventricular function, heart rate (HR), arterial pressure (AP), left ventricular pressure (LVP) and dp/dt (LV +/- dp/dtmax) were measured in male SD rats divided into three groups: Control group (n = 8, intravenously injected with physiological saline), native SOD group (n = 8,800 U SOD i. v.) and MPEG-SOD group (n = 9,800 U MPEG-SOD i. v.). LVP, LV +/- dp/dtmax, AP and HR showed no significant difference among these three groups before hypoxia. But in acute hypoxia simulating altitude of 6,000-6,500 m, LVP, LV+dp/dtmax and AP were decreased in the control group, while in MPEG-SOD group they were all increased compared with those in control group (P < 0.05, P < 0.01). There were no significant difference between control group and native SOD group. The results indicate that MPEG-SOD has a protective effect on the injury of myocardial function induced by acute hypoxia, and suggest that acute hypoxia cause increase of superoxide free radical (O2.-) and other derivative radicals damaging membrane system of the cell.

Congenital absence of vas deferens treated by artificial spermatocele constructed from the tunica vaginalis; report of six cases.

Six patients with congenital absence of the vas deferens were treated by artificial spermatoceles (ASCs) in an attempt to collect sperm for subsequent artificial insemination (AI). The ASC was constructed from a flap of tunica vaginalis from the patient. Sperm were aspirated from the ASC each month; there was wide variation in the concentration of sperm recovered [33.0 +/- 48.2 x 10(6) mL-1 (range 0.1-200)] and motility and normal morphology of the sperm were 19.0 +/- 17.9% and 48.8 +/- 23.4% respectively. Although a total of 14 cycles of AI was performed using the aspirated sperm, pregnancy did not result. Both poor sperm recovery and poor motility contributed to this failure. The condition of the epididymis observed during scrotum examination is reported. It is suggested that in some patients with congenital absence of vas deferens, the paired Wolffian ducts may differ from each other in their development.

[Rapid ultracentrifugal isolation and imaging detection of cholesterol-phospholipid vesicles].

We have utilized ultracentrifugation of bile-metrizamide density gradients to isolate and to quantitate the cholesterol-phospholipid vesicles (CPV) and identified them by transmission electron microscopy and scanning electron microscopy metrizamide was dissolved in bile and continuous density gradients were performed. The distribution of biliary cholesterol, phospholipid and bile salt was studied. The distribution of biliary phospholipid is similar in different fraction of the gradients. Approximately 70% of total biliary cholesterol was concentrated in the lightest fraction of the gradients (density < 1.035g/ml) and 70% of bile salt was found in the heaviest fraction of the gradients (density > 1.035g/ml). CPV harvested in fractions with density < 1.035g/ml and their diameter was 70-90nm. When 15% Metrizamide was dissolved in bile and centrifuged (50,000rpm, 100min, 37 degrees C), two thirds of total biliary cholesterol was concentrated in the top 0.4ml of the 5ml centrifuge tube. A lot of CPV was found in this part of bile and their diameter was 85nm. The advantage of the procedure described in this study is simple, rapid and accurate for pathological and physiological studies of CPV.

[Effects of obstructive jaundice on immunocompetence in jaundiced patients].

Using CD series monoclonal antibody indirect immune fluorescent assay and LDH enzyme-release assay, we investigated lymphocyte subsets and natural killer cells (NKC) activity in the peripheral blood of 30 patients with obstructive jaundice. The results showed that the percentage of CD3+, CD4+, B cells and that of CD8+ was high. The ratio of CD4/CD8 was low at the first 10 days of jaundice when the serum bilirubin level was less than 171 mumol/L. Our studies suggested that obstructive jaundice might cause depression and impairment of host cellular and humoral immunity. Further studies showed that impaired immune function of jaundiced patients returned to normal 30 days after the disappearance of the jaundice by external or internal biliary drainage. Immune function recovered 30 days after relief of biliary obstruction and external or internal biliary drainage. External or internal biliary drainage was useful in improving host immunity.

[Changes in LAK cell activity in chronic viral hepatitis and treatment with autologous LAK cell transfusion].

We investigated the activity of LAK cells in 10 patients with chronic persistent hepatitis, 20 patients with chronic active hepatitis, 21 patients with post-hepatitic cirrhosis and 21 normal persons. The values were 36.87 +/- 7.44, 30.38 +/- 5.36, 28.84 +/- 4.95 and 44.50 +/- 4.75 respectively (P < 0.05). The LAK cell activity in patients with chronic hepatitis was lower than that of normal persons, especially in those with chronic active hepatitis and post-hepatitic cirrhosis. The causes of lower LAK cell activity in patients with chronic hepatitis were less expression of IL2 receptors on precursors of LAK cells and existing serum LAK cell inhibition factors. We further studied 9 patients with chronic hepatitis B treated with autologous LAK cell transfusions. Factors favoring a more effective result were female sex, ALT elevation after treatment, activity of LAK cells > 10%, and inhibition factor of LAK effector cells existing in serum of patients, with inhibition rate < 50%. These factors might be used as the indications in selecting cases for treatment and predicting the effect of the treatment. As the number of cases observed was rather small, further investigation is needed.

[Eighty cases of alveolar hydatid disease treated with albendazole].

Alveolar hydatid disease occurs frequently in Xiji County of Ningxia. 80 cases of the disease were treated orally with albendazole from 1985 to 1988 and the short-term results proved satisfactory. The treatment consisted of two groups. Group I included 72 cases which were orally given 8-10 mg/kg.d of albendazole 2 to 3 times a day after each meal for 30 days and then stop taking the medicine for 10 days as one treatment course. The treatment continued 3-12 months comprising about 24 courses were given with the highest dosage of 412g. Group II included 8 cases who were given 15.6-20 mg/kg.d of the medicine for 14 days (one course) and every two weeks with a 10 days interval. The treatment of group II continued 3-6 months which totalled 19-24 courses with the highest dosage of 270g. All the treated patients tolerate well except that a few of them felt giddy and had diarrhoea which disappeared after the withdrawal of medicine. Through 3-5 years, follow-up observation, it was found that the gener alsymptoms of the treated patients improved and their body weight increased by 2-9.5 kg, some of them could take part in light physical labour. The jaundice index in 80% of them returned to normal. SGPT in 86.4% decreased to within normal range. The enlarged livers in half of the patients decreased by 1-6 cm. 23.7% of the patients were reexamined by B ultrasound scanning which revealed that their enlarged liver reduced in size while the liquifaction area and the shadows of calcification in the livers in 42.5% patients increased obviously.

[1:2 matched case-control study of congenital deficiency of newborn].

Among 5959 perinatal newborn, 135 cases of congenital deficiency were detected. The frequency was 22.66 per 1,000 newborns. In order to explore possible factors related to congenital deficiency, for 103 cases a 1:2 matched case-control study was performed. Mantel-Haentel chi 2 test and conditional logistic regression model was used to identify factors which associated with the disease significantly in statistics. It was found that six factors, i.e. common cold and fever in the early stage of pregnancy, illiteracy of mother, family history of congenital disease, haemorrhage of genital tract, alcohol drinking and marriage within blood relatives, are risk factors related to congenital deficiency of newborn.

Case-Control Studies

Morphological heterogeneity of esophageal carcinoma.

To clarify the morphological heterogeneity of esophageal carcinoma, the adenocarcinomatous, basaloid, and sarcoma-like components of 178 esophageal carcinomas were studied with regard to histopathology, mucin histochemistry, immunohistochemistry, and ultrastructure. Adenocarcinomatous components with mucicarminophilic cells and/or glandular structures, basaloid components, and sarcoma-like components were found in 55 lesions (30.9%), 17 lesions (9.3%), and five lesions (2.8%) respectively. Carcinoembryonic antigen staining was positive in 52 lesions (29.2%), secretory component staining was positive in 15 (8.4%), and lactoferrin staining was positive in 12 (6.7%). Eight intraepithelial carcinomas were found to have no adenocarcinomatous components, and two intramucosal carcinomas had adenocarcinomatous components in the invasive portions. These findings strongly suggest that the adenocarcinomatous components do not arise from the ductal epithelium, but occur during the process of invasion. There were no significant clinicopathological differences between the carcinomas with adenocarcinomatous components and those without. Ultrastructurally, the adenocarcinomatous components were seen to possess intracellular microcysts, intercellular lumina, and bundles of tonofilaments, having features of both glandular and squamous epithelia. On the basis of the concept that the basaloid components are histological variants of squamous cell carcinoma and that sarcoma-like components arise from mesenchymal metaplasia of squamous cell carcinoma, it is possible that the three components may originate from the squamous component. The present study thus demonstrated a high incidence of histological variation among esophageal carcinomas.

Optimization by stimulating molecular evolution.

Based on the analogy between mathematical optimization and molecular evolution and on Eigen's quasi-species model of molecular evolution, an evolutionary algorithm for combinatorial optimization has been developed. This algorithm consists of a versatile variation scheme and an innovative decision rule, the essence of which lies in a radical revision of the conventional philosophy of optimization: A number of configurations of variables with better values, instead of only a single best configuration, are selected as starting points for the next iteration. As a result the search proceeds in parallel along a number of routes and is unlikely to get trapped in local optima. An important innovation of the algorithm is introduction of a constraint to let the starting points always keep a certain distance from each other so that the search is able to cover a larger region of space effectively. The main advantage of the algorithm is that it has more chances to find the global optimum and as many local optima as possible in a single run. This has been demonstrated in preliminary computational experiments.

Algorithms
