Detection and differentiation of antigenically distinct small round-structured viruses (Norwalk-like viruses) by reverse transcription-PCR and southern hybridization.

Application of reverse transcription (RT)-PCR to detect small round-structured viruses (SRSVs) from fecal specimens of patients with gastroenteritis has been insensitive because of the tremendous sequence heterogeneity between strains. We have designed two RT-PCR primer sets (G-1 and G-2) based on the nucleotide sequence diversity in the RNA polymerase gene of SRSVs belonging to two distinct genogroups represented by Norwalk virus (primers G-1) and Snow Mountain agent (primers G-2). All 22 SRSV strains examined that had been classified previously by solid-phase immune electron microscopy into four antigenic types (UK1, UK2, UK3, and UK4) could be detected by RT-PCR with these two primer sets. The G-1 primer set detected 6 UK2 strains, and the G-2 primers detected 16 strains, including 7 UK1, 5 UK3, and 4 UK4 strains. On the basis of nucleotide sequences of 81-bp fragments of the RT-PCR products from 13 strains determined in this study, together with those previously reported for 17 SRSV strains, we designed four sets of internal oligonucleotide probes (P1-A, P1-B, P2-A, and P2-B) for Southern hybridization, using chemiluminescent detection. The P1-A probe hybridized with PCR products from the UK2 strains; the P1-B probe, with products from two of the seven UK1 strains; the P2-A probe, with four of the remaining five UK1 strains; and the P2-B probe, with products from both UK3 and UK4 strains, as well as with one strain originally typed as UK1 which showed cross-reactivity with UK4 upon retesting by solid-phase immune electron microscopy. RT-PCR with both the G-1 and the G-2 primer sets can increase the detection rate of the many antigenically distinct SRSVs and, when combined with Southern hybridization, may predict the antigenic type of the SRSV associated with infection.

PCR strategy for identification and differentiation of small pox and other orthopoxviruses.

Rapid identification and differentiation of orthopoxviruses by PCR were achieved with primers based on genome sequences encoding the hemagglutinin (HA) protein, an infected-cell membrane antigen that distinguishes orthopoxviruses from other poxvirus genera. The initial identification step used a primer pair of consensus sequences for amplifying an HA DNA fragment from the three known North American orthopoxviruses (raccoonpox, skunkpox, and volepox viruses), and a second pair for amplifying virtually the entire HA open reading frame of the Eurasian-African orthopoxviruses (variola, vaccinia, cowpox, monkeypox, camelpox, ectromelia, and gerbilpox viruses). RsaI digest electropherograms of the amplified DNAs of the former subgroup provided species differentiation, and TaqI digests differentiated the Eurasian-African orthopoxviruses, including vaccinia virus from the vaccinia virus subspecies buffalopox virus. Endonuclease HhaI digest patterns distinguished smallpox variola major viruses from alastrim variola minor viruses. For the Eurasian-African orthopoxviruses, a confirmatory step that used a set of higher-sequence-homology primers was developed to provide sensitivity to discern individual virus HA DNAs from cross-contaminated orthopoxvirus DNA samples; TaqI and HhaI digestions of the individual amplified HA DNAs confirmed virus identity. Finally, a set of primers and modified PCR conditions were developed on the basis of base sequence differences within the HA genes of the 10 species, which enabled production of a single DNA fragment of a particular size that indicated the specific species.

Home care for the urban chronically ill elderly in the People's Republic of China.

This study examines the home care situation of seventy-five urban chronically ill men and women and their caregivers in three cities in the People's Republic of China. It documents the type of home care provided, the severity of illness, and the dependency in activities of daily living in order to appreciate the complexity of these care situations. The findings reveal that women serve as the caregivers in the majority of the cases and often they are elderly spouses with health problems of their own. The physical, social, psychological, and financial problems experienced by these patients create an enormous burden for the caregivers whether spouses or younger family members coming home from work to a second shift. If alternatives to home care, such as nursing homes, were available, 54 percent of these caregivers would be willing to institutionalize their elderly relative. All caregivers express the need for more assistance in providing home care. This study raises questions regarding the role of the family, the state, and specifically the health care system in caring for the elderly chronically ill in light of their increasing numbers in the population.

Effects of the seed sludge on the performance of UASB reactors for treatment of toxic wastewater.

A comparative study of different sludges (anaerobic sewage sludge and anaerobic sewage sludge cultivated for 30 days) as the seed of UASB reactors for treatment of synthetic resin production wastewater was carried out on a laboratory scale. Significant differences in reactor performance were observed. With anaerobic sewage sludge seed, the reactor obtained a 71% COD removal efficiency and produced 2.94 dm3 biogas per dm3 wastewater under an organic loading rate of 4.5 kg COD m-3 day-1. The sludge in the reactor was diffuse and possessed poor settling ability, which led to effluent SS as high as 1200 mg dm-3. However, with anaerobic sewage sludge cultivated with synthetic substrate for 30 days as a seed, the same reactor gave 78% COD removal efficiency and produced 3.5 dm-3 biogas per dm3 wastewater at the same loading rate. Moreover, the sludge in the reactor was completely granulated and possessed good settling ability, which resulted in effluent SS as low as 200 mg dm-3. The granular sludge produced less biomass per gram COD removed than the diffused sludge. Anaerobic sewage sludge cultivated for a short time could form the nuclei of granules for the reactor, allowing granulation to proceed smoothly. Therefore, sludge nuclei are a prerequisite of sludge granulation in UASB reactors for treatment of toxic wastewater.

Design, synthesis and evaluation of bouvardin, deoxybouvardin and RA-I-XIV pharmacophore analogs.

The synthesis and in vitro cytotoxic evaluation of a key set of cycloisodityrosine subunit analogs of deoxybouvardin and RA-VII are detailed and constitute a complete investigation of the natural product pharmacophore. The studies illustrate that the 18-membered ring tetrapeptide potentiation of the cytotoxic activity of cycloisodityrosine is not likely to be due to simple alteration or constraint of the conformation of the 14-membered cycloisodityrosine subunit and that simple derivatization of cycloisodityrosine may not provide the same potentiation.

Effect of albendazole on the larvae and eggs of Necator americanus in golden hamster.

When golden hamsters infected with adult Necator americanus were treated ig with albendazole(Alb) 75 or 150 mg/kg, the efficacy rates were 83.8% and 100%, respectively. Three days before treatment, the mean numbers of larvae recorded from the incubation of feces from 2 treated groups were 29,716 +/- 3,299 and 28,256 +/- 118 per gram of feces, while in the control group, it was 8,828 +/- 2,659. Within 3 days after treatment, no larva was detected in the 2 treated groups, but in the control group the mean larva number was 9,066 +/- 7,416. Furthermore, in 2 treated groups none or only a few degenerated eggs could be found using saturated saline solution method. when infected golden hamsters were treated ig with Alb at a single dose of 150 or 300 mg/kg on day 3, 7 or 14 after infection with the third stage larvae, the egg positive rates in the feces were 0-9.1%, while the worm reduction rates were 96.8-100%.

Spectral-luminosity functions, scalar linearity, and chromatic adaptation.

We report data for three experiments that assess the effect on the luminosity function of chromatic adaptation arising from the measurement stimuli. First, we report spectral-sensitivity functions (wavelength range, 510-640 nm) measured by heterochromatic flicker photometry for a luminance range of 25-5000 Td. The data were fitted to a linear combination of cone fundamentals. The data narrowed and the fits deteriorated with an increase in luminance level, which indicates that at high luminances chromatic adaptation that is dependent on the spectral composition of the standard and test lights is a factor in spectral-luminosity determination. Second, we report heterochromatic modulation photometry as measured with two spectral lights at constant time-averaged chromaticity and luminance for luminances from 1.6 to 1300 Td. For a time-averaged chromaticity of 570 nm, the red-green ratio of the photometric match was invariant with luminance. For a time-averaged chromaticity of 605 nm, the red-green ratio increased by almost 0.3 log unit for a 2-log-unit increase in luminance, which is indicative of chromatic adaptation to the 605-nm chromaticity. Third, we measured flicker increment detection (wavelength range, 510-640 nm) on 570- and 605-nm backgrounds of 25-5000 Td. The data were fitted to a linear combination of cone fundamentals and showed good fits at all luminances. Fits to the 570-nm-background data set showed little variation in the proportions of the cone fundamentals with luminance. Fits to the 605-nm-background data set required an increased weighting of the middle-wavelength-sensitive cone with luminance. These three experiments indicate that luminance-dependent variation in the spectral-luminosity function as assessed by flicker techniques is caused primarily by chromatic adaptation to the measurement stimuli.

Cloning and structural-functional studies for 7.5k promoter of Tiantan strain of vaccinia virus.

The 7.5k promoter of vaccinia virus Tiantan strain has been cloned by DNA polymerase chain reaction (PCR) method. Results of DNA sequencing analysis showed that in comparison with P7.5k of WR strain, three site mutations and 7 bp of natural deletion existed in the 155 bp fragment of P7.5k of Tiantan strain; four of 7 base pairs deleted were in the late transcriptional initiation region. Although the total mutation rate was high to 6.45%, these two 7.5k promoters of Tiantan strain & WR strain were all early-late promoters and not obviously different in their activities and functional phases. The results above confirmed further that it is a mechanism to keep their genetic stability that some genes of vaccinia virus have multiple transcriptional initiation sites and produce many mRNA with heterologous 5' ends.

The nondeletional types of Hb H disease in Guangxi.

Three primers were designed, one specific for alpha 1-globin DNA, a second for alpha 2-globin DNA, and a third that is common for both alpha 1- and alpha 2-globin DNA. These three primers can be applied for selective amplification of the two globin DNA fragments, which is useful for identification of nondeletional types of Hb H disease. Fifty-nine DNA samples of Hb H patients from Guangxi were studied by selective amplification, and 27 cases (45.8%) were confirmed as nondeletional types. Of these, 22 (81.5%) had the Hb Constant Spring (CS) mutation and one had the Hb Quong Sze (QS) mutation; both were identified by hybridization with synthesized oligonucleotide probes. Nondeletional Hb H disease in Guangxi seems to be more severe than the deletional types. The average hemoglobin level of the nondeletional Hb H/CS (--/alpha alpha) is 6.8 g/dl, which is lower than that of the deletional types (7.9 g/dl), while the levels of Hb H and Hb Bart's were much higher in the patients with Hb H/CS than in those with the deletional types.

Molecular cloning of a new variant of human papillomavirus type 6 isolated from a Chinese woman and expression of its L1 gene in E. coli--molecular cloning & expression of HPV6 gene.

A human papillomavirus genome DNA of 7.9 kb from a Chinese woman with genital condyloma acuminata was cloned in BamHI site of pAT153. According to the results obtained from Southern blotting, restriction mapping as well as partial DNA sequencing, the isolated genome (HPV6BV) had obvious variance and was referred to as a new variant of HPV6 found in China the first time. HPV6BV L1 gene was successfully expressed in E. coli as a fusion protein with pUR288. The beta-galactosidase/L1 fusion protein reacted with both beta-galactosidase antiserum and HPV antibody using Western blot technique. The E. coli-produced fusion protein, possessing HPV antigenicity, may provide a reagent for clinical diagnosis and epidemiological survey.

A new human interferon-gamma mutant harboring EGF receptor-interfering sequence possesses high antiproliferative activity.

An interferon gamma (IFN-gamma) mutant gene harboring an additional epidermal growth factor (EGF) sequence at its 3' terminal was constructed and highly expressed in E. coli by using pBV220 as a vector. The new fusion protein-IFN-gamma-EGF possesses not only high antiviral activity, but also high antiproliferative effect as demonstrated by 3H-TdR incorporation method using CN II cells which are rich in EGF receptors.

A novel variant of human interferon alpha 1 gene.

A 520-base pair human IFN-alpha gene was isolated by PCR method twice from chromosome DNA of a Chinese (Han Nationality) fetal liver. The nucleotide sequences were determined. These two separately amplified DNA fragments shared the completely identical nucleotide sequence but possessed C and G at positions 410 and 541, respectively, which differ from those of IFN-alpha 1 and IFN-alpha D previously described. Therefore the deduced amino acid sequence would have an Ala at position 114 and a Val at position 158. At all other sites it has the same amino acids as those in IFN-alpha 1 and IFN-alpha D. We recommend that IFN-alpha D gene, IFN-alpha I gene and IFN-alpha I/158V gene found in our laboratory, be named IFN-alpha 1a gene, IFN-alpha 1b gene and IFN-alpha 1c gene.

High expression of tumor necrosis factor alpha cDNA in E. coli using site-specific deletion mutagenesis.

Using oligonucleotide-directed site specific mutagenesis technique, a TNF-alpha cDNA coding 157 amino acids without signal sequence and initiated with ATG was obtained. Sequencing data showed the precise deletion and insertion of the DNA fragment as designed. The modified TNF gene was expressed in E. coli with a expression vector pBV220. The expression level reached 10(8) units per liter culture. The scanning of the SDS-PAGE gel showed that the amount of the expressed TNF is about 22.8% of total soluble bacterial protein. The TNF activity can be neutralized with TNF monoclonal antibody.

A study on an enhancer-like element in Escherichia coli.

An enhancer-like element of 1.0kb (JM103-M) from E. coli JM103 chromosome was isolated using an enhancer-probing vector. The JM103-M fragment was shown to stimulate the nearby CAT or beta-galactosidase gene expression in E. coli in both directions at a stimulation rate of 3-6. The same fragment was also demonstrated to have promoter function when a promoter-probing vector was tested. Results obtained from sequencing data showed that a sequence TGACTAA homologous to the AP-1 binding DNA motif of SV40 and two GC boxes were present.

The vaccinia virus HindIII K fragment encodes a novel protein belonging to the serpin superfamily.

The 1983-base pair nucleotide sequence of the EcoRI-HindIII fragment of vaccinia virus Tiantan strain HindIII K clone is determined by the dideoxy chain termination method. A search in the NBRF protein sequence database using FASTA and other microcomputer programs reveals that several proteins belonging to the serpin (serine protease inhibitor) superfamily have striking similarities to the protein encoded by the HindIII K1 ORF. On the basis of the dot-matrix analysis and sequence alignment, the K1-encoded protein is shown as a novel member of the serpin superfamily. The putative reactive site and switch sequence of this novel serpin are then compared with those of other serpins. The probable evolutionary and possible functional relationships are discussed.

Vaccinia virus hemagglutinin. A novel member of the immunoglobulin superfamily.

Striking similarities between vaccinia virus hemagglutinin (VVHA) and proteins belonging to the Ig superfamily clearly indicate that VVHA, a 315-amino acid glycoprotein expressed on the surface of the infected cells, is a novel viral protein that can be added to the expanding list of the Ig superfamily. Its deduced amino acid sequence contains one Ig-like domain at the NH2 terminus, followed by two tandem repeating units and a hydrophobic region, suggestive of membrane spanning. The results offer an opportunity for the further study of the probable evolutionary and possible functional relationship between VVHA and other members of the Ig superfamily. Our observation, together with a recent finding that human CMV possibly encodes a protein similar to the MHC class I antigens (13), provides evidence supporting the fact that the viral capture of cellular Ig-related genes is more common than expected in vaccinia and other viruses, and that usage of an Ig-like domain as recognition signals might be extended from higher animals to animal viruses.
