Low-temperature increases the yield of biologically active herring antifreeze protein in Pichia pastoris.

Antifreeze proteins and antifreeze glycoproteins are structurally diverse molecules that share a common property in binding to ice crystals and inhibiting ice crystal growth. Type II fish antifreeze protein of Atlantic herring (Clupea harengus harengus) is unique in its requirement of Ca(2+) for antifreeze activity. In this study, we utilized the secretion vector pGAPZalpha A to express recombinant herring antifreeze protein (WT) and a fusion protein with a C-terminal six-histidine tag (WT-6H) in yeast Pichia pastoris wild-type strain X-33 or protease-deficient strain SMD1168H. Both recombinant proteins were secreted into the culture medium and properly folded and functioned as the native herring antifreeze protein. Furthermore, our studies demonstrated that expression at a lower temperature increased the yield of the recombinant protein dramatically, which might be due to the enhanced protein folding pathway, as well as increased cell viability at lower temperature. These data suggested that P. pastoris is a useful system for the production of soluble and biologically active herring antifreeze protein required for structural and functional studies.

Improved immnunophenotyping of lymphocytes in bronchoalveolar lavage fluid (BALF) by flow cytometry.

BACKGROUND: The immnunophenotyping of lymphocytes in bronchoalveolar lavage fluid (BALF) is of particular importance in the differential diagnosis of interstitial lung disorders. The standard method of lymphocyte phenotyping is peroxidase-anti-peroxidase technique (PAP). However, it was time-consuming and experience-dependent. Flow cytometric (FCM) analysis of BALF lymphocytes was introduced to overcome these disadvantages. Unfortunately, when the number of cells counted was small, FCM could not distinguish lymphocytes from other cells and particles in BALF by light scattering. METHODS: We established a tri-color flow cytometric approach to phenotyping of lymphocytes in BALF. FITC-CD45/PE-CD14 antibodies were used to gate lymphocytes and exclude other contamination. Propidium iodide (PI) was introduced to distinguish lymphocytes from debris. Forty-three BALF species were tested by flow cytometer as well as peripheral blood as a control group by conventional PAP method. RESULTS: The variation of FCM (CV<1.0%) was much lower that that of PAP method (CV>9.8%). Meanwhile, we found that BALF had more clinical significance than peripheral blood in T subset analysis (p<0.01). There were characteristic changes in some lung diseases. Both CD3 and CD4 were significantly increased with decreasing CD8 in sarcoidosis (n=14). Idiopathic pulmonary fibrosis (n=16) demonstrated the reverse tendency: CD8 rose but both CD3 and CD4 dropped. As for lung cancer (n=7), CD3 was normal but the CD4/CD8 ratio declined. Tuberculosis of the lungs (n=6) showed a normal CD3, CD4 and CD8. CONCLUSIONS: The high precision and reliability of tri-color flow cytometric approach to phenotyping of lymphocytes in BALF suggested that it should be used as a routine test, especially of BALF, which was often contaminated by inorganic particles.

Nitric oxide synthase in spinal cord central sensitization following intradermal injection of capsaicin.

Nitric oxide (NO) is believed to be an important messenger molecule in signal transduction pathways that enhance nociceptive transmission in the central nervous system (CNS). The role of nitric oxide synthase (NOS) I and II, which synthesize NO, in central sensitization induced by an intradermal capsaicin injection was investigated. To elucidate whether changes in NOS I and NOS II activities caused by capsaicin injection contribute to behavioral changes, responses to von Frey filaments with two different innocuous bending forces applied on the rat foot were tested. The allodynic responses induced by capsaicin injection in the foot were partially reversed by the administration of either the selective NOS I inhibitor, 7-nitroindazole (7-NINA), or the selective NOS II inhibitor, 2-amino-5,6-dihydro-6-methyl-4H-1,3-thiazine (AMT). To confirm changes at the level of single nociceptive neurons, extracellular recordings were made from rat dorsal horn neurons. The electrophysiological results showed that increased responses to noxious and innocuous stimuli caused by capsaicin injection were blocked by either 7-NINA or AMT delivered through a microdialysis fiber inserted through the dorsal horn. Finally, the expression of both NOS I and NOS II in the spinal cord as demonstrated by Western blots was increased by 20 min following intradermal capsaicin injection in the rat foot. These results suggest that both NOS I and NOS II are upregulated following intradermal capsaicin injection and that both cause NO release that contributes to the secondary hyperalgesia and allodynia following this noxious chemical stimulus.

NMDA or non-NMDA receptor antagonists attenuate increased Fos expression in spinal dorsal horn GABAergic neurons after intradermal injection of capsaicin in rats.

GABAergic neurons play an important role in the generation of primary afferent depolarization, which results in presynaptic inhibition and, if large enough, triggers dorsal root reflexes. Recent electrophysiological studies by our group have suggested that increased excitation of spinal GABAergic neurons by activation of N-methyl-D-aspartate (NMDA) and non-NMDA receptors following intradermal injection of capsaicin results in the generation of DRRs that contribute to neurogenic inflammation. The present study was to determine if changes in the expression of Fos protein occur in GABAergic neurons in the lumbosacral spinal cord following injection of capsaicin into the glabrous skin of one hind paw of anesthetized rats and if pretreatment with an NMDA receptor antagonist, D-(-)-2-amino-7-phosphonoheptanoic acid (AP7) or a non-NMDA receptor antagonist, 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) blocks Fos expression in these neurons. The experiments used western blots and immunofluorescence double labeling staining following capsaicin or vehicle injection. Western blots showed that Fos protein was increased on the ipsilateral side in spinal cord tissue 0.5 h after capsaicin injection. Pretreatment with AP7 or CNQX caused a decrease in capsaicin-induced Fos expression. Immunofluorescence double labeling showed that the proportion of Fos-positive GABAergic neuronal profiles was significantly increased following capsaicin injection (48.8+/-4.8%) compared to the vehicle injection (23.8+/-5.1%) in superficial laminae on the ipsilateral side in lumbosacral spinal cord (P<0.05). However, when the spinal cord was pretreated with AP7 (5 microg) or CNQX (0.2 microg), only 9.1+/-0.6% or 7.1+/-0.8% of GABA-immunoreactive neuronal profiles were stained for Fos following capsaicin injection. The blockade of the capsaicin-evoked Fos staining was dose-dependent. These findings suggest that GABAergic neurons take part in dorsal horn circuits that modulate nociceptive information and that the function of GABAergic neurons following capsaicin injection is partially mediated by NMDA and non-NMDA receptors.

Uncoupling protein1 mRNA, mitochondrial GTP-binding, and T4 5'-deiodinase of brown adipose tissue in euthermic Daurian ground squirrel during cold exposure.

Regulation of thermogenic activity and uncoupling protein1 (UCP1) expression in brown adipose tissue (BAT) were studied in euthermic Daurian ground squirrel after acute and chronic cold exposure at 4 degrees C. The UCP1 concentration was indirectly determined by titration with its specific ligand [3H]-labeled GTP, and Ucp1 mRNA was detected by using a [32P]-labeled antisense oligonucleotide probe. Both acute and chronic cold exposure stimulated up-regulation of Ucp1 mRNA. Although UCP1 concentration is not significantly increased after 24 h of cold exposure, it is markedly elevated by 75% in squirrels after 4-week cold adaptation compared with controls raised at 22 degrees C. Changes in T4 5'-deiodinase activity were closely associated with variations of Ucp1 mRNA level. Ucp1 gene expression is significantly affected by cold exposure in BAT from euthermic Daurian ground squirrels. In addition, the activation of T4 5'-deiodinase may be an important regulatory factor in cold-induced Ucp1 expression.

Epigenetic regulation of gene structure and function with a cell-permeable Cre recombinase.

Studies of mammalian gene function are hampered by temporal limitations in which phenotypes occurring at one stage of development interfere with analysis at later stages. Moreover, phenotypes resulting from altered gene activity include both direct and indirect effects that may be difficult to distinguish. In the present study, recombinant fusion proteins bearing the 12 amino acid membrane translocation sequence (MTS) from the Kaposi fibroblast growth factor (FGF-4) were used to transduce enzymatically active Cre proteins directly into mammalian cells. High levels of recombination were observed in a variety of cultured cell types and in all tissues examined in mice following intraperitoneal administration. This represents the first use of protein transduction to induce the enzymatic conversion of a substrate in living cells and animals and provides a rapid and efficient means to manipulate mammalian gene structure and function.

The Beat generation: a multigene family encoding IgSF proteins related to the Beat axon guidance molecule in Drosophila.

A previous genetic screen led to the identification of the beaten path (beat Ia) gene in Drosophila. Beat Ia contains two immunoglobulin (Ig) domains and appears to function as an anti-adhesive factor secreted by specific growth cones to promote axon defasciculation. We identify a family of 14 beat-like genes in Drosophila. In contrast to beat Ia, four novel Beat-family genes encode membrane-bound proteins. Moreover, mutations in each gene lead to much more subtle guidance phenotypes than observed in beat Ia. Genetic interactions between beat Ic and beat Ia reveal complementary functions. Our data suggest a model whereby Beat Ic (and perhaps other membrane-bound family members) functions in a pro-adhesive fashion to regulate fasciculation, while Beat Ia (the original secreted Beat) functions in an anti-adhesive fashion to regulate defasciculation.

[Study on changes of serum T helper cell type 1 and 2 cytokines after active immunotherapy in women with unexplained habitual abortion].

OBJECTIVE: To investigate the changes of serum T helper cell (TH)1/TH2 type cytokines after the active immunotherapy in unexplained habitual abortion (UHA) women. METHODS: Concentrations of interleukin (IL)-2, IL-12, interferon (IFN)-gamma, IL-4, IL-10 and transforming growth factor (TGF)-beta 1 were measured by enzyme-linked immunosorbant assay (ELISA) method in sera from thirty-three cases of unexplained habitual abortion (UHA) women before and after active immunotherapy. Thirty normal non-pregnancy (NNP) women and thirty normal pregnancy (NP) women were taken as control. RESULTS: (1) Serum concentrations of IL-2 and IL-12 were higher significantly (P < 0.01) in UHA women [(13.3 +/- 13.8) ng/L and (50.5 +/- 25.8) ng/L, respectively] than those in NNP women [(4.6 +/- 6.4) ng/L and (20.3 +/- 28.2) ng/L, respectively]. Serum concentrations of IL-4 and IL-10 were lower significantly (P < 0.01) in UHA women [(13.8 +/- 1.0) ng/L and (13.5 +/- 0.7) ng/L, respectively] than those in NNP women [(14.5 +/- 1.2) ng/L and (14.9 +/- 2.4) ng/L, respectively]. However, there were no significant differences in serum concentrations of IFN-gamma and TGF-beta 1 between UHA women and NNP women. (2) Serum concentrations of IL-2 were lower significantly (P < 0.05) in NP women [(1.6 +/- 4.3) ng/L] than those in NNP women. Serum concentrations of IL-4 were higher significantly (P < 0.01) in NP women [(16.3 +/- 0.8) ng/L] than those in NNP women. But there were no significant differences in serum concentrations of IFN-gamma, IL-10 and TGF-beta 1 between NP and NNP women. (3) After active immunotherapy, serum concentrations of IL-2 and IL-12 were decreased significantly [(5.6 +/- 9.0) ng/L and (28.5 +/- 40.3) ng/L respectively, P < 0.01] whereas serum concentrations of IL-4 and IL-10 were increased significantly [(14.7 +/- 1.2) ng/L and (15.0 +/- 1.8) ng/L respectively, P < 0.01] in UHA women. In addition, no significant changes in serum concentrations of IFN-gamma and TGF-beta 1 were found in UHA women after immunotherapy. (4) As compared to NNP women. there were no significant differences in serum concentrations of the above-mentioned cytokines in UHA women after active immunotherapy. CONCLUSIONS: The results suggest that profile of TH1/TH2 type cytokines tilts to TH2 immunity in normal pregnancy, but to TH1 immunity in UHA. The active immunotherapy could make a shift from TH1 to TH2 immunity, thus favoring the maintenance of pregnancy.

[Effect of organic acids on soil chemical behavior of lead and cadmium and their toxicity to plants].

The interaction between organic acids (citric and oxalic acids) and heavy metals was examined in this paper. Equilibrium dialysis showed that the extent organic acids bound metals depended on the metal involved, and their binding properties affected the adsorption of Pb and Cd by soil, which was increased in the presence of oxalic acid, and decreased in the presence of citric acid. The effect of citric acid on the behavior and toxicity of Pb and Cd to plants was also studied. The presence of citric acid could reduce the toxicity of Pb, and inhibit the uptake of Cd in shoots and roots. However, there was no significant effect on the apparent toxicity of Cd. The interaction between Pb and Cd was found to be complicated. The Pb content in rice was higher in the presence of Cd, while the content of Cd is lower in the presence of Pb.

[Relationship between placenta growth factor and the pathogenesis of pregnancy induced hypertension syndrome].

OBJECTIVE: To determine whether pregnancy induced hypertension syndrome (PIHs) is associated with placenta growth factor (PLGF). METHODS: PLGF level in the maternal and umbilical venous serum collected from 23 PIHs patients and 20 normotensive controls was measured by enzyme linked immunoassay (ELISA), and expression of PLGF and its mRNA in the placenta and decidua from them was assessed by the immuno-histochemistry and reverse transcriptional-polymerase chain reaction (RT-PCR). RESULTS: (1) PLGF levels in the maternal serum from PIHs [(112.7 +/- 63.8) micrograms/L] were significantly lower than those from normal pregnancies [(200.3 +/- 140.9) micrograms/L], (P < 0.05), and they were correlated with infant birth weight and placental mass (r = 0.4, r = 0.6). (2) Expression of PLGF in sycytiotrophoblast and villous mesenchyme and decidua from PIHs significantly decreased respectively (P < 0.01, P < 0.01, P < 0.05), and expression of PLGF in sycytiotrophoblast was associated with the severity of PIHs (P < 0.05). (3) The transcription of total PLGF mRNA in the placenta and decidua from PIHs decreased significantly (P < 0.05 and P < 0.05 respectively). Particularly, the transcription of PLGF-2 mRNA decreased significantly in the placenta from PIHs. CONCLUSION: The abnormal expression of PLGF may play an important role in the pathogenesis of placental ischaemia hypoxia in PIHs.

Therapeutic antitumor response to cervical cancer in mice immunized with U14 vaccines transfected with costimulatory B7 gene.

OBJECTIVE: To investigate the effect of U14 vaccine transfected with the B7 gene in inducing antitumor immune response to murine cervical carcinoma in Chinese 615-strain mice. METHODS: A recombinant retroviral plasmid vector expressing mouse B7-1 gene (pLNSX-mB7) was transfected into 615-strain mouse cervical carcinoma cell line No. 14 (U14) by electroporation to set up a highly-expressed mB7-1 U14 cell clonal strain (B7+ U14). In vivo experiments: (1) B7+ U14 vaccine was primed to protect the 615-strain mice against U14 re-challenge. (2) B7+ U14 vaccine was injected into tumor-bearing mice with different tumor sizes. Lifetimes and tumor sizes were recorded. In vitro cytotoxicity assay: Mice were immunized with B7+ U14 or U14 vaccine and 2 weeks later, spleen cells of those mice were cultured for 2 days. The cytotoxicity of these cells against U14 was detected by 5-diphenyl tetrazolium bromide assay. RESULTS: We obtained several B7-1 high expression clonal U14 lines. In vivo experiment, we did not find tumor growing in 3 of the 6 mice primed by B7+ U14 vaccine during their entire life after re-challenge with U14. The other 3 mice developed tumors and their average survival time was longer than that of the control group (P < 0.01). All 6 mice grew tumors in the control group. When the transplanted tumors became palpable, the mice were randomly divided into 3 groups to be injected with B7+ U14 vaccine. It was effective for tumor-bearing mice only when the tumor diameters were < 3 mm. When the diameters were > or = 3 mm, it was not efficacious to inject B7+ U14 vaccine (P < 0.05). In vitro cytotoxicity assay, cytotoxic T lymphocytes induced by B7+ U14 vaccine had a higher cytotoxicity against U14 than that induced by U14 vaccine (F = 310.8, P < 0.001). CONCLUSIONS: Vaccines of cervical cancer cells transfected with the costimulatory molecule B7 gene can induce antitumor immune protection in host mice against U14 re-challenge. This treatment may cure part of the tumor-bearing mice but be restricted by tumor size. The results suggest that transfecting the B7 gene into cervical cancer as a cell vaccine may be an efficient supplementary method to treat cervical cancer after operation.

A novel mis-sense mutation (G1381A) in the G6PD gene identified in a Chinese man.

OBJECTIVE: To detect new mutations among 29 glucose-6-phosphate dehydrogenase (G6PD) deficient individuals from Yunnan province. METHODS: The nitroblue tetrazolium (NBT) method was used to screen G6PD deficient individuals. Mutation was identified by single strand conformation polymorphism (SSCP), amplification created restriction site (ACRS), amplification refractory mutation system (ARMS) and DNA sequencing. RESULTS: Among 29 cases, 18 cases of G1388A, 1 case of C1004A, and 1 case of G1381A were identified. Nine cases remained to be defined. The G1381A mutation is a novel mis-sense mutation, with a substitution of threonine for alanine (A461T). The resultant G6PD had reduced enzymatic activity. In addition, G1381A caused a restriction site of Stu I to disappear, providing a rapid method for the detection of this mutation. CONCLUSION: A novel mis-sense mutation G1381A was found. This mutation results in a substitution of threonine for alanine, producing enzyme with reduced activity. The loss of the Stu I restriction site offers a rapid method for the detection of this mutation.

[Study on association of vascular endothelial growth factor with the pathogenesis of pregnancy induced hypertension].

OBJECTIVE: To investigate relationship between the vascular endothelial growth factor (VEGF) and the pathogenesis of pregnancy induced hypertension syndrome (PIHs). METHODS: Twenty-three women with PIHs and twenty normal pregnant women were studied. The levels of VEGF in maternal and umbilical venous serum, placenta and decidual tissue from normal pregnancy and PIHs women were detected by enzyme linked immunosorbent assay (ELISA) and immunohistochemistry. Furthermore, the expression of VEGF mRNA in placenta and decidua was examined by reverse transcriptase polymerase chain reaction (RT-PCR). RESULTS: (1) The levels of VEGF in maternal blood were significantly lower in PIHs women than that in normal pregnancies women (10.4 +/- 3.8) ng/L and (17.0 +/- 9.3) ng/L (P < 0.01). (2) The average gray of VEGF was determined on sections of placenta and decidua from PIHs or pregnancies women by computer image analysis. The expression of VEGF in placenta and decidua (69.0 +/- 8.9) and (55.0 +/- 7.3) was decreased significantly in PIHs women than that in normal pregnancies women (75.0 +/- 9.0) and (60.5 +/- 6.4, P < 0.05). (3) The transcription levels of the VEGF mRNA in placenta and decidua were increased significantly in PIHs women than those in normal pregnancies women (4.6 +/- 3.2 vs 2.8 +/- 1.0; 6.3 +/- 2.9 vs 3.9 +/- 1.5, P < 0.01). CONCLUSIONS: These findings suggested that the abnormal expression of VEGF in PIHs occurred within the low-stream of genic transcription. It was considered that the abnormal expression of VEGF may play an important role in the pathogenesis of placental ischemia in PIHs.

[The study on human leucocyte antigen DQ region genes polymorphism in unexplained habitual abortion patients].

OBJECTIVE: To investigate whether human leucocyte antigen DQ (HLA-DQ) genes polymorphism are related to the susceptibility to unexplained habitual abortion. METHODS: HLA-DQA1 and HLA-DQB1 gene typings are performed by polymerase chain reaction-restrictive fragment length polymorphism(PCR-RFLP) technique in 32 cases of unexplained habitual abortion and 54 women with normal pregnancy history. RESULTS: In unexplained habitual abortion group, the frequency of DQB1 * 0604, 0605 allele is increased significantly compared with that of controls(12.5% and 2.8%, P < 0.05) and the frequency of DQB1 * 0501, 0502 allele is decreased significantly (3.1% and 11.1%, P < 0.05). The frequency of DQA1 alleles do not differ significantly between two groups. Furthermore, the frequency of DQA1 * 01/DQB1 * 0604,0605 haplotype in patients is significantly increased than that of controls (12.5% and 2.8%, P < 0.05). CONCLUSION: The results suggest that DQB1 * 0604,0605 allele and DQA1 * 01/DQB1 * 0604,0605 haplotype may be a susceptible gene and haplotype of unexplained habitual abortion, while DQB1 * 0501,0502 allele may be a protective factor for habitual abortion.

[Study on polymorphism of human leukocyte antigen I in patients with endometriosis].

OBJECTIVE: To study the possibility of a correlation between polymorphism of human leukocyte antigen (HLA)-I antigen and occurrence of endometriosis (Em). METHODS: Polymorphism of HLA-I A and B locations were detected by mini-lymphocytotoxin test in 40 surgically proven Em patients and 50 normal controls. The frequency of different antigens on both A and B sites were compared between the 2 groups. RESULTS: No significant difference of the antigen frequency at HLA-I A site was shown between Em and control groups. However, at HLA-I B site, frequency of B46 antigen in Em group was significantly higher [33% Vs 16%, P < 0.05, relative risk (RR) 2.5278] and frequency of B48 antigen in Em group was significantly lower (3% Vs 16% P < 0.05, RR 0.1346) as compared with those of the controls. CONCLUSIONS: The occurrence of Em may be associated with the presence of HLA-I B46 antigen. On the contrary, the HLA-I B48 antigen might play a protective role against Em.

[The combined occluding effect of fluor protector and Nd:YAG laser irradiation on human dentinal tubules].

OBJECTIVE: To evaluate the combined occluding effect of fluor protector and Nd:YAG Laser irradiation on human dentinal tubules. METHODS: Twenty-four dentin specimens with exposed dentinal tubule orifices treated by 37% H3PO4 were randomly divided into four groups. Group B, C and D were vanished by fluor protector, group A served as a control. Group D was lased by Nd:YAG laser. Group C and D were brushed with normal force. RESULTS: Under SEM, group A showed numerous exposed dentinal tubule orifices, the diameter of which is 2-3 microns. Group B showed closure of dentinal tubule orifices. Group C showed that most of the fluor protector were brushed away and group D showed over 80% of the dentinal tubule orifices were occluded. CONCLUSIONS: Fluor protector combined with Nd:YAG laser can make most of the dentinal tubule orifices occluded even after brushed.

Analysis of 133 patients with severe blunt liver injury.

OBJECTIVE: To investigate the treatment and causes of death about severe blunt liver injury. METHODS: The data of 133 patients with severe blunt liver injury (Grade III to Grade V) were analyzed retrospectively. All the patients except 2 underwent operations. Different types of surgical procedures were adopted according to the severity of liver injury. RESULTS: Operations were the major method to treat hepatic injury. The total mortality rate was 21.0% in this study. The mortality rates of Grade III, Grade IV and Grade V of liver injuries were 11.3%, 24.4% and 69.2%, respectively. CONCLUSIONS: Packing treatment plays an important role in the treatment of hepatic injury. The mortality rate is related closely to the severity of hepatic injury, multiple trauma, shock and disturbance of blood coagulation.
