We looked for the earliest signs of the onset of myopia in 194 emmetropic (-0.25 to +0.75 D) eyes by following them for 1 to more than 2 years. Visual acuity examination, cycloplegic retinoscopy, and A-scan ultrasonography were performed at intervals of 6 to 12 months. Of the 194 eyes 64 became myopic during the observation time. Our results show that during the progression from emmetropia to myopia the change in visual acuity is not a good indicator of the onset of myopia. When visual acuity decreased to below 6/6 (20/20) cycloplegic retinoscopy showed that most of the 64 eyes had become truly myopic and A-scan ultrasonography revealed a significant increase in vitreous chamber depth and axial length as compared to the non-myopic eyes. In the early stage of myopia development the anteroposterior diameter of the eye elongates. However, changes in anterior chamber depth and lens thickness did not differ between the myopic and non-myopic group. Our results suggest that routine ocular refraction should be conducted in addition to the school vision screening. For those who have visual acuity 6/6 (20/20) and ametropia of -0.25 D or more minus we would like to be able to prevent or at least retard the development of myopia.
An immunosuppressed rat model was established by injecting cortisone acetate 25 mg/rat twice a week for 4 weeks and 12.5 mg/rat for another 2 weeks subcutaneously. A development of Pneumocystis carinii pneumonia (PCP) was found at the end of the 6th week in all rats. These rats were injected intramuscularly with artemether at 100 mg/kg once a day for 5 consecutive days. All rats were necropsied at the end of the 8th week. The lung impression smears were stained with Gomori's stain, and Pneumocystis carinii cysts were counted. The ultrastructural changes of trophozoite, precyst and cyst were investigated by transmission electron microscopy on the 7th day after treatment.
BALB/c mice were repeatedly inoculated subcutaneously with purified rat source Pneumocystis carinii (P.c.) cysts. Six weeks later, the spleen cells were fused with NS1 myeloma cells, and a mouse hybridoma producing monoclonal antibody against P.c. was established and cloned, which was named as 4D7 McAb. Its mean of chromosome was 82. The 4D7 McAb was shown to be IgG1 subclass. It reacted mainly with 54 kDa polypeptide of P.c. in EITB. 4D7 McAb could clearly recognize P.c. by immunohistochemical staining in the specimens from confirmed Pneumocystis carinii pneumonia (PCP) patients, and the sensitivity and specificity were both 100% for diagnosis of PCP (11/11). No cross-reaction of 4D7 McAb was found with Pneumococcus, Candida albicans and Cryptococcus neoformans, as well as all examined normal human tissues.
Rat Pneumocystis carinii cyst soluble antigen (CSA) was analysed by SDS-PAGE, and more than 20 polypeptide bands were found. The molecular weights of the major bands were > 100kDa, 85-94kDa, 67kDa, 52kDa and 34kDa, respectively. The specific IgG antibodies in the sera from healthy blood donors recognizing the 110kDa, 105kDa, 85kDa, 67kDa, 52kDa and 46kDa polypeptide antigens were identified by immunoblot analysis. The total positive rate of IgG specific antibodies was 56.7% (59/104). The 85kDa polypeptide antigen could also react with the monoclonal antibody derived from human Pneumocystis carinii. The positive rate of the specific anti-85kDa-IgG antibody was 37.5% (39/104), but the specific anti-Pneumocystis carinii IgM antibodies were not detected in all the sera examined. Three of 7 serum samples from the patients with Pneumocystis carinii pneumonia were positive for specific anti-85kDa-IgG antibody (Figs 1-4).
Nuclear transplantation has been proved to be a very powerful method in researching the relationship between nucleus and cytoplasm during embryogenesis. Electrofusion is a new means of cell fusion which is being improved in the past nearly ten years. In this study, we produced interorder nucleocytoplasmic hybrid embryos between mouse and rabbit with these two techniques. Two phenomena, i.e. premature chromosome condensation (PCC) and nuclear swelling, were observed after mouse 8-cell stage blastomeres were fused into rabbit enucleated oocytes. After 4.5-days cultured in mouse oviducts, 5.4% reconstituted embryos developed into blastocysts. By karyotype examination, it was known that only mouse chromosomes were presented in the blastomeres, one blastocyst even had a normal mouse karyotype (2 n = 40, XX). As a conclusion, we consider it is possible to obtain normal early development of interspecies nucleocytoplasmic hybrid embryos between mouse and rabbit.
Twenty Wistar rats were injected with cortisone acetate twice a week subcutaneously for 6-12 wks. By 7 weeks Pneumocystis carinii pneumonia was induced in rats successfully. The cysts of Pneumocystis carinii from infected lungs of cortisonized rats were identified by phase-contrast microscopy, Giemsa's stain, Chalvardjian's stain and Gomori's methenamine silver nitrate stain. The process of isolation and purification of Pneumocystis carinii from infected rat lungs included the following three test steps. First, the tissue was cut into small pieces and homogenized and filtered through #60, #100, #200-gauge wire mesh respectively. Secondly, the homogenate was digested with collagenase, the optimal concentration of collagenase being 0.2%. Thirdly, the discontinuous percoll density gradient centrifugation was used to separate P. carinii cysts. The majority of P carinii cysts were present in a density zone of approximately 1.033g/ml and were essentially free from host lung debris, the latter being removed due to their higher density, 1.040g/ml.
Bilateral electrical lesioning of the hippocampal CA3 region (HCA3-EL) or anterior commissura hippocampi (ACHF-EL) caused marked elevations in plasma basal levels of insulin. 2 weeks later, fasting blood glucose levels were also augmented with decreased glucose tolerance. In contrast, the secretory response of pancreatic B cells to glucose stimulation was markedly enhanced. Following intravenous glucose tolerance test (IVGTT), the relative amounts of glucagon-like and insulin-like immunoreactants were reduced in the pancreatic islets of both HCA3-EL and ACHF-EL rats in comparison with the controls. In the HCA3-EL group, the relative amounts of somatostatin-like immunoreactants and gross numbers of such immunostained cells in islets were also decreased as compared with the control. No difference was seen in pancreatic-polypeptide-like immunoreactivities as assessed by immunohistochemistry plus microphotometry method. The above results suggest strongly that HCA3 and ACHF exert a tonic inhibitory action on the insulin secretion in the rat.
Through a critical study of herbalogy, it is concluded that the mature seeds of Alpinia zermubet were used as caodoukou in ancient times. The original plant of Yutao recorded in Zhiwu Mingshi Tukao may be the species of Alpinia katsumadai. Baidukou used in ancient times may be the seeds of Alpinia katsumadai found in the south of China, or Amomum compactum and Am. kravanh imported from foreign countries.
One hundred and nine clinically diagnosed manic patients were examined and re-diagnosed with SADS/RDC. The results of joint-test and test-retest showed that the reliabilities of the subscales of SADS were good enough for clinical use. The mania subscale score of SADS showed significant negative correlation with GAS score, but positive correlation with SI or BRMS. The results of joint-test and test-retest showed that the diagnosis of mania by the use of SADS/RDC were reliable. The concurrent validity between SADS/RDC and CMA-H was higher. This study indicated that SADA/RDC can be used satisfactorily in clinical practice.
A predominant population of mucosal nerves in the mammalian gastric body has been known to contain large amounts of gastrin-releasing peptide (GRP). The present immunohistochemical study demonstrates the depletion of GRP immunoreactivity from nerves in the oxyntic mucosa of rats with ulcers induced by restraint plus water immersion. Depletion of GRP occurred in a major part of the nerves after 3 h exposure to the stress, and after 6 h exposure only a few nerve fibers could be recognized. In contrast, GRP fibers in the pyloric mucosa did not decrease significantly in number in any of the stressed rats. Since the depletion of GRP immunoreactivity preceded mucosal erosion in the gastric body, the possibility is proposed that GRP released from the nerves may be related to stress-related ulceration in the stomach.
PURPOSE: To evaluate the safety and feasibility of a percutaneous Port-Catheter System (PCS) implanted via the subclavian artery (SCA) for regional chemotherapy or chemoembolization of thoracic, abdominal, and pelvic malignant tumors. METHODS: Percutaneous puncture of the SCA was performed in 256 patients with thoracic, abdominal, or pelvic malignant tumors; then a catheter was inserted into the target artery. After the first transcatheter chemotherapy or chemoembolization with an emulsion of lipiodol and anticancer agents, an indwelling catheter was introduced with its tip placed in the target artery and its end subcutaneously connected to a port. RESULTS: The procedure was successfully completed in all 256 cases (100%). The indwelling catheter tip was satisfactorily placed in the target arteries in 242 cases (98%). Complications attributable to the procedure occurred in 20 (7.8%) cases, including pneumothorax (n = 10, 4%), hemothorax (n = 1, 0.4%), infections in the pocket (n = 4, 1.6%), and hematoma at the puncture site (n = 5, 2%). There were no severe sequelae or deaths. The duration of PCS usage was 1-36 months (median 9.5 months). During the course of treatment, occlusion of the target artery occurred in 20 cases (7.8%). Dislocation of the tip of the indwelling catheter occurred in 12 cases (4.7%); in 10 of the 12, the tip of the indwelling catheter was repositioned into the target artery. In all 10 cases no large symptomatic hematomas developed after the PCS was removed. CONCLUSION: Percutaneous PCS implantation via the left SCA, a relatively new procedure, is a safe and less invasive treatment approach than surgical placement for malignancies.
