[Diagnosis of non-typical myocarditis by applying immunohistochemical method of CD68].

OBJECTIVE: To make a distinction between myocarditis and the reaction to some pathological state of myocardium. METHODS: Myocardium of 26 cases with sudden cardiac death were stained and LM light microscopies with immunohistochemical method 10 cases with normal myocardium were contrasted. RESULTS: A great deal of stained positive monocyte of immunohistochemistry emerged in the parasetions of myocarditis patients with various farms and stacking(> 15). CONCLUSION: The stain of immunohistochemistry can be used as one of the indications for diagnosing non-typical myocarditis.

[Brainstem changes in death from head injury].

The cortex and brainstem changes in death from head injury were observed. There were hemorrhage, edema and neuron necrosis in brain injury group, and also in disputed group of brainstem injury.

Increase of ocular blood flow by some phytogenic compounds.

The effects of a number of phytogenic compounds on ocular blood flow were measured by the colored microsphere technique in rabbits. Methyltyramine and coumarin increased blood flow in all eye tissues tested. The blood flow of iris, ciliary body-iris root, retina and choroid was increased by methyltyramine for 255%, 322%, 189%, and 189%, respectively, and by coumarin for 375%, 193%, 139% and 299%, respectively, at 120 minutes after drug administration. Rescinnamine, apocynin and hesperetin also increased ocular blood flow in all eye tissues except retina. The blood flow of iris, ciliary body-iris root, and choroid was increased by rescinnamine for 344%, 234%, and 199%, respectively; by apocynin for 355%, 152%, and 414%, respectively; and by hesperetin for 265%, 136%, and 204%, respectively, at 120 minutes after drug instillation. These results indicate that these compounds could be used to treat ischemic eye diseases in the future.

Diagnostic value of ions as markers for differentiating antemortem from postmortem wounds.

In the present study, the diagnostic value of ions as markers for differentiating antemortem from postmortem wounds was investigated. To accomplish this, skin and muscle samples were taken from antemortem and postmortem wounded cadavers and the contents of ions (Fe, Zn, Mg, Cu, K and Na) were determined with atomic absorption spectrophotometry. Our results show that the concentrations of Fe in antemortem wounded skin and muscle were significantly higher as compared with the control skin and muscle and the K/Na ratio in antemortem wounded muscle was reduced significantly compared to control muscle. However, the concentrations of Zn, Mg, Cu in antemortem wounded skin and muscle, and the K/Na ratio in antemortem wounded skin, were not significantly different from their controls. The concentrations of all these ions in postmortem wounded skin and muscles were not significantly different from their controls. It is indicated that the concentrations of Fc in skin and muscle and K/Na ratio in muscle are useful markers for differentiating antemortem from postmortem wounds. However, their routine use in forensic practice needs further investigation, since there are some overlaps of the ionic concentrations between normal and wounded skin and muscle.

Prevention and treatment of ocular inflammation with a new class of non-steroidal anti-inflammatory agents.

New non-steroidal anti-inflammatory agents (NSAIAs) were tested on lens protein-, endotoxin- and interleukin-1-induced ocular inflammation. It was found that most NSAIAs, including REV 5901, mefenamic acid, indomethacin, CK-17 and CK-102, inhibited lens protein-induced inflammation. Endotoxin induced inflammation indirectly through the release of IL-1 which was inhibited by fewer agents, including CK-17, CK-102 and prednisolone. However, the direct effect of IL-1 can only be suppressed by CK-17 and prednisolone. Therefore, CK-17 could become an important NSAIA which acts similarly to corticosteroids yet produces no steroidal side effects. CK-17 was different from most NSAIAs as it affected little, if any, arachidonate metabolism. Most importantly, CK-17 was found to be 2-fold more potent than prednisolone in inhibiting IL-1-induced uveitis, while no side effects were noted at doses tested to date.

Prevention of ocular inflammation induced by lens protein, endotoxin, and interleukin-1 with synthetic interleukin-1 blockers.

It is well known that corticosteroids are potent anti-inflammatory agents, yet they produce serious side effects. Although arachidonate metabolite blockers have been developed for the treatment of inflammation, they are much less potent than corticosteroids. Furthermore, they still process serious side effects. In search of potent and safe non-steroidal anti-inflammatory agents (NSAIA), interleukin-1 (IL-1) blockers have been developed. Among 121 CK-analogs studied, CK-17, CK-101A and CK103A have been identified as promising anti-inflammatory agents as potent as prednisolone in inhibiting lens proteins-induced inflammation and twice as potent as prednisolone in inhibiting endotoxin-and IL-1-induced uveitis. No serious side effects could be noticed with the doses of these compounds tested to date. These results indicate that the development of potent NSAIAs is feasible. Moreover, these compounds are not related to arachidonate metabolites.

Inhibition of crystallins-induced inflammation in rabbit eyes with five phytogenic compounds.

Ocular inflammation was induced by injection of crystallins (lens protein) intracamerally and endotoxin intravitreously into rabbit and rat eyes, respectively, and was measured with fluorophotometry by quantitating the amount of fluorescein which entered into the globe. Five compounds isolated from anti-inflammatory Chinese herbs were studied for their effects on ocular inflammation. It was found that lens protein-induced inflammation was inhibited significantly by the topical instillation of pulegone (0.5%), friedelin (0.5%), and sabinene (1%), but not by dihydrojasmon or naringin at concentrations up to 1%. However, none of these compounds inhibited endotoxin-induced posterior uveitis.

Inhibition of ocular inflammation by chalcone derivatives and prednisolone.

Posterior uveitis was induced by injection of 10 micrograms endotoxin intravitreally into rat eyes and anterior ocular inflammation was induced by injection of 0.75 mg of lens protein intracamerally into rabbit eyes. Four chalcone derivatives, RVC-556 (2'-hydroxychalcone), RVC-574 (2'-hydroxychalcone hydrazone), RVC-574P (2'-hydroxychalcone phenyl hydrazone) and RVC-588 (4,4'-dihydroxy chalcone) were studied along with prednisolone at a dose of 10 mg/kg i.p. t.i.d. for their anti-inflammatory actions. RVC-574 was more active than prednisolone in inhibiting posterior uveitis by 65% and 43%, respectively. RVC-556, RVC-574P, and RVC-588 did not affect the posterior uveitis in rats. On the other hand, anterior ocular inflammation was inhibited by 1% eyedrops of RVC-556, RVC-574P and RVC-588 but not by RVC-574. RVC-556 was more active than; RVC-574P was less active than; and RVC-588 was about equiactive as prednisolone in inhibiting anterior ocular inflammation by 77%, 47%, 69%, and 64%, respectively.
