[Comparison of different lysis buffers in the assessment of the efficiency of single cell polymerase chain reaction for preimplantation genetic diagnosis].

OBJECTIVE: To set up a highly efficient cell lysis system to increase the efficiency of single cell polymerase chain reaction (PCR) for preimplantation genetic diagnosis (PGD). METHODS: First we picked up one single lymphocyte or blastomere and lysed it in alkaline lysis buffer (ALB), proteinase K buffer (PK) and a new proteinase K lysis buffer compounded by ourselves. Then we amplified the 3rd exon of Gene ALD using nested PCR and compared the amplification efficiencies. RESULTS: The amplification efficiencies of the single lymphocyte in ALB, PK and the lysis buffer compounded by ourselves were 90%, 74%, and 87%; those of the single blastomere were 98%, 77%, and 96%. CONCLUSION: ALB and the lysis buffer compounded by ourselves are superior to PK in the lysis of cells in clinical PGD.

Coupling parameter in synchronization of diluted neural networks.

We study the critical features of a coupling parameter in the synchronization of neural networks with diluted synapses. Based on simulations, an exponential decay form is observed in the extreme case of global coupling among subsystems and full connection in each network: there exists a maximum and a minimum of the critical coupling intensity for synchronization in this spatially extended system. For partial coupling, we present the primary result about the critical coupling fraction for various linking degrees of the networks.

In situ epitaxial MgB2 thin films for superconducting electronics.

The newly discovered 39-K superconductor MgB2 holds great promise for superconducting electronics. Like the conventional superconductor Nb, MgB2 is a phonon-mediated superconductor, with a relatively long coherence length. These properties make the prospect of fabricating reproducible uniform Josephson junctions, the fundamental element of superconducting circuits, much more favourable for MgB2 than for high-temperature superconductors. The higher transition temperature and larger energy gap of MgB2 promise higher operating temperatures and potentially higher speeds than Nb-based integrated circuits. However, success in MgB2 Josephson junctions has been limited because of the lack of an adequate thin-film technology. Because a superconducting integrated circuit uses a multilayer of superconducting, insulating and resistive films, an in situ process in which MgB2 is formed directly on the substrate is desirable. Here we show that this can be achieved by hybrid physical-chemical vapour deposition. The epitaxially grown MgB2 films show a high transition temperature and low resistivity, comparable to the best bulk samples, and their surfaces are smooth. This advance removes a major barrier for superconducting electronics using MgB2.

[Redox properties and conformational changes of DsbA protein from Escherichia coli periplasm].

DsbA protein, a disulfide bond enzyme in Escherichia coli periplasm, catalyzes mainly disulfide bond formation of proteins. Site-directed mutagenes is combined with Trp-analog labeling technique was used to investigate the redox properties and conformational changes of DsbA. The results show that: (1) DsbA, as an oxidase, can catalyze disulfide bond formation efficiently. The reduced form is more stable than the oxidized form, suggesting that the strong oxidizing force of DsbA comes from the tense conformation of the oxidized form. (2) The alteration of local environment around Trp(76) between redox forms is responsible for the special fluorescence phenomenon of DsbA. (3) The result from (19)F-NMR study provides further evidence that the local conformation of Trp(76) is dramatically affected during the transformation of redox forms, but that of Trp(126) remains unaffected.

Correlation of proliferating cell nuclear antigen expression with the clinical and biological characteristics and prognosis of hypopharyngeal carcinoma.

OBJECTIVE: To study the association of proliferating cell nuclear antigen (PCNA) with the biological characteristics and clinical behavior of hypopharyngeal carcinoma and with the prognosis of the malignancy. METHODS: SP-immunohistochemical method was used to detect the expression of PCNA in 48 cancer tissue specimens and 15 specimens of the adjacent tissue of primary hypopharyngeal carcinomas. The number of PCNA-positive cells was represented by proliferating index (PI) of the antigen. RESULTS: PCNA expression was correlated with the clinical stage, histological grade and clinical recurrence of hypopharyngeal carcinoma as well as the prognosis of the patients, but was not significantly related with the tumor location or the presence of metastasis to the cervical lymph nodes. CONCLUSION: PCNA protein expression may help evaluate the malignancy of hypopharyngeal carcinoma and make prognostic judgement of the patients.

[The change in IL-13 mRNA expression in rat lungs with acute pulmonary injury induced by lipopolysaccharide].

OBJECTIVE: To observe the change in IL-13 mRNA expression in rat lungs with acute lung injury (ALI) induced by lipopolysaccharide (LPS), so as to investigate the significance of anti-inflammatory mechanism in the development of ALI. METHODS: (1) Different doses (2 approximately 8 mg/kg) of LPS were injected via the tail vein to the rats to establish ALI model. (2) The IL-13 mRNA content in pulmonary tissue was determined by RT-PCR. RESULTS: (1) Different degrees of ALI was induced in the rats by different doses of LPS. ARDS was produced in the rats by LPS of 6 mg/kg. (2) The IL-13 mRNA expression in the pulmonary tissue could be enhanced by the induction of LPS, especially when its dose was larger than 6 mg/kg. CONCLUSION: (1) Rat ALI model could be produced by LPS injection via the tail vein. (2) LPS in dose of 6 mg/kg was the threshold for the production of rat ARDS. (3) The incidence of rat ARDS might be closely related to the enhanced expression of IL-13 mRNA in rat pulmonary tissue.

[Changes of plasma interleukin-4, interleukin-10 and interleukin-13 in patients with acute respiratory distress syndrome].

OBJECTIVE: To explore the anti-inflammatory mechanisms of interleukin-4 (IL-4), interleukin-10 (IL-10) and interleukin-13 (IL-13) in acute respiratory distress syndrome (ARDS). METHODS: Enzyme-linked immunosorbent assay (ELISA) was employed to determine the plasma levels of IL-4, IL-10 and IL-13 in patients with ARDS (n = 22) and systemic inflammatory response syndrome (SIRS, n = 8). Ten normal subjects were enrolled as controls. RESULTS: The plasma levels of IL-4, IL-10 and IL-13 were (261 +/- 55) microg/L, (458 +/- 112) microg/L and (5.21 +/- 2.02) microg/L, respectively in patients with ARDS; (155 +/- 26) micro g/L, (260 +/- 54) microg/L and (1.69 +/- 0.39) microg/L respectively in patients with SIRS; and (43 +/- 13) microg/L, (135 +/- 15) microg/L and (0.33 +/- 0.10) microg/L, respectively in normal controls. They were significantly increased in patients with ARDS and SIRS as compared to normal controls, and the levels were higher in patients with ARDS than in patients with SIRS. CONCLUSIONS: Increased release of anti-inflammatory cytokines, such as IL-4, IL-10 and IL-13, may be one of the underlining mechanisms for the development of ARDS and SIRS.

Effects of elevated atmospheric CO2 on nematode trophic groups in a Chinese paddy-field ecosystem.

Soil fauna plays significant roles in the detritus food webs of agroecosystems, they are the essential contributors to the decomposition of soil organic matter, mineralization of plant nutrients and nutrient cycling. Some evidences indicate that soil fauna of the detritus food webs appears to show positive, neutral or negative responses to global change, especially atmospheric CO2 enrichment across different studies. Soil nematodes are representative of a large portion of this fauna, since they are abundant and trophically diverse in most soils. The free-air CO2 enrichment (FACE) technology was used to expose three plots of rice (Oryza sativa) under elevated (ambient + 200 mumol.mol-1) atmospheric CO2, while three control plots were outfitted with FACE apparatus under ambient CO2. Nematode trophic groups were monitored in this study at depths of 0-5 cm and 5-10 cm during rice jointing and ripening stages in Wuxi site, China. Significant differences were found between depths and sampling dates in the total numbers of nematodes, bacterivores, plant parasites and omnivores-predators during the study period. The total numbers of nematodes and bacterivores at the 5-10 cm depth were higher in treatment plots than in control plots across all sampling dates. At 0-5 cm depth, the numbers of bacterivores were higher in treatment plots than in control plots, while those of omnivores-predators exhibited an inverse trend during the study period. Significant differences were also observed between treatment plots and control plots in the numbers of fungivores in our experiment.

[Current situation and prospect of elevated atmospheric CO2 effects on soil nematodes].

The elevated atmospheric CO2 can make effects on the ecosystem, these effects will be mediated by the soil fauna. In this paper, we firstly discussed the response of the soil nematodes to the elevated atmospheric CO2 in different soil pattern and in different ecosystems. Some evidences indicate that soil fauna of the detritus food webs appear to show positive, neutral or negative responses to the atmospheric CO2 enrichment across different studies. The research prospect in the future was put forward. This paper can help us understand some ecological processes of farmland ecosystems according to the effect of elevated CO2 on soil nematodes and provide the foundation for managing farmland ecosystems effectively.

Rapid increase in colorectal cancer rates in urban Shanghai, 1972-97, in relation to dietary changes.

BACKGROUND: In urban Shanghai, the largest industrial and commercial city in China, the age-adjusted (to world standard) incidence rates for colorectal cancer increased from 14.8 to 24.1 per 10(5) man-years and from 11.7 to 20.7 per 10(5) woman-years between 1972-73 and 1996-97. These changes were even more pronounced for colon cancer. The reasons for the rapid increases in cancer rates are not fully understood, but may involve dietary habits that have changed substantially overthe past two decades. METHODS: Based on incidence data on 37000 colorectal cancers from 1972-1997 and dietary information during the past 20 years, an ecologic correlation analysis was performed. RESULTS: Available data indicate that per capita food consumption in Shanghai of vegetable oil, poultry, eggs, and pork rose rapidly during the period 1978-97, whereas consumption of seafood, grain, and fresh vegetables changed little or showed little consistent trends. Statistically significant positive associations were observed between colon cancer rates and per capita consumption of vegetable oil, poultry, fresh eggs and pork. DISCUSSION: These findings suggest that increases in dietary fat and certain protein consumption may play a role in the rising colon cancer rates in Shanghai.

[A long-term observation of 48 cases of tracheoesophageal shunt phonation by the anastomosis of the membranous portion of the tracheal section with the anterior wall of esophagus after total laryngectomy].

OBJECTIVE: To study the long-term efficacy of tracheoesophageal (TE) shunt phonation by the anastomosis of the membranous portion of the tracheal section with the anterior wall of esophagus after total laryngectomy. METHODS: A questionnaire was designed to follow 48 patients with rehabilitated speech and swallow functions by the above approach. The qualities of phonation and speech, and the degree of aspiration evaluated, together with the survival rate and complications statistically analyzed. RESULTS: The speech level of 35 laryngectomees was similar to the normal laryngeal speech level. Thirty-five cases had longer maximal phonation time and hearing distance and higher speech intelligibility. Other 5 laryngectomees had less effective phonation, but higher speech intelligibility. The total effective rate was 83.3% (40/48). The speech fluency in 40 laryngectomees was not as good as that in the normal people. Forty percent laryngectomees(16/40) had slight liquid food aspiration which did not influence normal eating. Eight patients(8/48) failed to speak and no liquid aspiration occurred after the operation. The effective rate was affected obviously by neck infection and pharyngeal fistula formation. The survival rate was similar to those with single total laryngectomy. CONCLUSIONS: The TE shunt phonation had the advantages of simple, one-stage operation and high success rate. Some laryngectomees had slight liquid food aspiration, but would not influence normal eating. So this TE shunt phonation operation may be useful during laryngeal surgery.

[Long-term results of one stage tracheoesophageal end-side anastomosis voice restoration after total laryngectomy].

OBJECTIVE: The purpose of this study is to discuss long-term results of one different kind of voice restoration after total laryngectomy with 10 years experience. METHOD: One stage tracheoesophageal end-side anastomosis after total laryngectomy was performed in 36 patients from 1991-1996. The key technique of this operation is to make a tongue-ship flap(1.5 cm x 1.5 cm-1.8 cm x 1.5 cm) with tracheal membranous part and dissect it from the esophagus to 1 cm below the upper incisal margin of trachea. Then a 0.8 cm middle vertical incision was made on the lowest anterior wall of the separated esophagus. Folding the tongue-ship flap posteriorly to approximate the esophagus incision and make an end-side anastomosis. Close the upper incisal margin of trachea with left to right direction suture. The follow up time is 5 - 10 year. One lost follow 2.5 years later and was judged as dead. RESULT: The short-term and long-term success rate were 69% (25/36) and 83% (30/36) respectively. The 3-, 5- and 10 year survival rate are 72%, 69%, 69%, respectively. CONCLUSION: This technique does not change the pattern of total laryngectomy and various neck dissections do not affect voice rehabilitation. Advantages of the operation: easy to operate and learn; the special form of tracheoesophageal shunt (high in the tracheal side and low in esophageal side) prevents the aspiration; high successful rate, excellent speech, fluent language, good acceptability and lower aspiration.

[Determination of andrographolide and dehydroandrographolide in Andrographis paniculata nees materials and related patent medicines by reversed-phase high performance liquid chromatography].

A simple and accurate method for the determination of andrographolide and dehydroandrographolide in andrographis paniculata Nees materials and patent medicines with high performance liquid chromatography (HPLC) has been developed. The two components were extracted from powdered samples by shaking with methanol. The resultant extracts were separated within 15 min on a BECKMAN C18 column (4.6 mm i. d. x 250 mm, 5 microm) and with a gradient elution of acetonitrile-water at a flow rate of 0.5 mL/min. The detection wavelength was 225 nm and the injection volume was 20 microL. In gradient elution program the volume fraction of acetonitrile in mobile phase was as follows: 0 min - 1 min, 40%; 1 min - 5 min, 40% - 50%; 5 min - 15 min, 50% - 70%. Both andrographolide and dehydroandrographolide have good linearity in the range of 10 mg/L to 100 mg/L with the correlation coefficients of 0.997 6 and 0.998 6 respectively. This method has been successfully applied for the analysis of andrographis paniculata Nees materials and related patent medicines.

Evaluation of myocardial viability with myocardial contrast echocardiography: a preliminary clinical study.

OBJECTIVE: To evaluate the efficacy of myocardial contrast echocardiography (MCE) in the assessment of myocardial viability, with positron emission tomography (PET) as the golden standard. METHODS: Eleven patients with anterior wall Q wave myocardial infarction were enrolled in this study, who received successful percutaneous transluminal coronary angioplasty (PTCA) 3 to 19 months prior to the examinations by PET and MCE that were completed within 2 d. RESULTS: MCE score for the segments of necrotic myocardium, viable myocardium and normal myocardium was mostly 0, 0.5 and 1 respectively, and there was a significant difference between the grades of MCE score in identifying myocardial viability. In terms of diagnosing myocardium survival, MCE result was closely correlated with that of PET (with the correlation index rp of 0.78). CONCLUSION: Myocardial perfusion evaluation can be effectively accomplished by MCE, which might serve as a new approach to assess myocardial viability in clinical practice.

Changes in pulmonary vascular endothelial cells after acute lung injury induced by bone marrow extract injection in rabbits.

OBJECTIVE: To investigate the changes of the markers of pulmonary vascular endothelial cells (PVECs) after acute lung injury (ALI) induced by bone marrow extract (BME) injection in rabbits. METHODS: Thirty-one rabbits were randomized into control (CG, n=10) and experimental groups (EG, n=21). The rabbits in EG were injected with homogeneous bone marrow extract (0.35 ml/kg, 2 ml/h) at a slow and continuous rate through the jugular vein to establish the model of ALI. At 6 h after the injection, the number of circulating endothelial cells (CECs) in the blood, contents of granule membrane protein-140 (GMP-140), angiotensin converting enzyme (ACE) and endothelin-1 (ET-1) in the plasma and the content of GMP-140 in the pulmonary tissue were determined at various time intervals. Then the animals were killed and routine pathological examination and electron microscopy were performed to observe the changes in the pulmonary tissue. RESULTS: The levels of plasma GMP-140, ACE, ET-1 and CECs were significantly increased in the early stage (0.5 h) and remained higher for 6 h. The marked increase of plasma GMP-140 (3.25 times) in the early stage was negatively correlated to PaO(2), but positively to other parameters. IHC-staining showed that the GMP-140 on the surface of PVECs became weak. CONCLUSIONS: BME injection at slow and continuous rate can establish an acceptable model of ALI. Determination of plasma GMP-140 might be an important measure for the early surveillance and the evaluation of prognosis of ALI in clinical management of serious traffic accidents.

Inheritance of microsatellite DNA markers in the Pacific abalone Haliotis discus hannai.

Microsatellite markers have been developed for a variety of abalones, and locus-specific homozygote excesses at population level have been recorded for microsatellite loci. To ascertain whether null alleles exist at microsatellite loci in the Pacific abalone, we studied the mode of inheritance of 7 microsatellite loci in 4 families with a reciprocal cross of 2 females x 2 males. All loci segregated codominantly, but only 3 loci ( Hdh1321, Hdh78, and Hdd108C) conformed to Mendelian segregation and can be used for parental analysis and population genetic studies. When null alleles were considered, 2 loci (Hdh1761 and Hdh1457) confirmed Mendelian expectations in all families, while the remaining 2 loci (Hdd114B and Hdd229) showed deviation from Mendelian segregation in at least one family even though null alleles were considered. These results indicated the need to test the inheritance pattern for microsatellite markers in abalones before using them for population genetic of parentage analysis.

Enhancement of antiangiogenic effects of human canstatin with a hypoxia-regulated transgene vector in lung cancer model.

UNLABELLED: Canstatin, a newly identified antiangiogenesis protein, has a potent inhibitory effect on the proliferation and growth of endothelial cells. To enhance the expression and antiangiogenic effects of canstatin in solid tumors, we constructed a eukaryotic expression vector that encodes human canstatin cDNA downstream from nine copies of the hypoxia-response element. METHODS: Canstatin complementary DNA from adult liver tissues was cloned into the mammalian expression vector pCMV-Script. Nine copies of the hypoxia-response element were ligated upstream from the canstatin gene near the cytomegalovirus promoter. The recombinant vector, pCMV9Cans, was transformed into A549 cells by cationic liposomes. The transformed cells were cultured under oxic and anoxic conditions. We detected canstatin messenger ribonucleic acid and protein expression in transformed cells by TaqMan polymerase chain reaction and Western blot analysis, respectively. Human umbilical vein endothelial cells were cocultured with recombinant vector transformed A549 cells using Transwell plates under oxic and anoxic conditions. The proliferation and apoptosis of the cocultured endothelial cells were evaluated with 3H-thymidine incorporation and terminal deoxynucleotidyl-mediated biotinylated deoxyuridine triphosphate nick end-labeling methods (TUNEL), respectively. A canstatin-encoding vector with no hypoxia-response element, pCMVCans, was used as the positive control, and naked plasmid-transformed and singly cultured parental cells were used as negative controls. The biologic activity of the vector in tumor tissues of lung cancer-bearing nude mice was evaluated by microvessel counts. Canstatin protein expression was assessed by Western blot analysis in tumor tissues. pCMVCans and empty vector were used as controls in the in vivo assays. RESULTS: Canstatin messenger RNA and protein were detected in both pCMV9Cans- and pCMVCans-transformed A549 clones. Under oxic conditions, canstatin expression was not significantly different in clones stably transformed with pCMV9Cans or pCMVCans. However, under anoxic conditions canstatin expression was significantly higher in pCMV9Cans-transformed cells than in pCMVCans-transformed cells. Moreover, the 3H-thymidine uptake rate of the human umbilical vein endothelial cells was markedly lower than that of the pCMVCans-transformed cells, and many endothelial cells underwent apoptosis when cocultured with pCMV9Cans-transformed A549 cells, especially under anoxic conditions. We detected canstatin expression in tumor tissues; the expression level in pCMV9Cans-transformed tumors was significantly higher than that in pCMVCans-transformed tumors. An in vivo assay showed that tumors transformed with pCMV9Cans remained small, and microvessels in those tumors were much fewer than those in pCMVCans-transformed tumors. CONCLUSION: The hypoxia-regulated vector pCMV9Cans increases the expression of canstatin, thereby enhancing its biological effects. We believe that the hypoxia-inducible canstatin-expressing vector is a promising gene therapy tool for antiangiogenesis research.
