[Quantitative fluorogenic real-time PCR assay for respiratory syncytial virus detection].

OBJECTIVE: To Establish a rapid and objective quantitative fluorogenic real-time PCR assay for early detection of human respiratory syncytial virus (hRSV). METHODS: Two pairs of primers and one TaqMan Fluorogenic probe that are specific for the recognition of the most conservative N gene of hRSV for virus detection with LighCycler PCR in 93 nasopharyngeal secretion specimens collected from infants and young children. The assay was compared with virus isolation, routine PCR, nested PCR, and enzyme-linked immunosorbent assay (ELISA). RESULTS: This TaqMan assay had a sensitivity of 1 x 10(2) cDNA copies/microl with a dynamic range between 1 x 10(2) and 1 x 10(7) cDNA copies/microl, which was the same as that of nested PCR, but 10 times more sensitive than routine PCR. The specificity of the assay was evaluated by comparing hRSV with polivirus type 1, coxsackie virus type 2, influenza A, influenza B and adenovirus type 7. A PCR product of the expected size (195 bp) was produced and fluorescence signal detected for hRSV, but not for any of the other viruses. The results in LightCycler and Rotor-Gene instrument were consistent. Forty-four specimens (43.9%) were hRSV-positive with this assay and 4 (4/93,4.3%) were hRSV-positive with ELISA, showing rather low correlation between the two methods. No visible relation was found between the concentration of hRSV RNA and severity of the disease. CONCLUSION: This assay is rapid, sensitive, specific and quantitative, and has the potential of wide application for early diagnosis of hRSV infection and evaluation of the therapeutic effect.

[Study on applying chromatographic fingerprints to establishing quality criterion of processed tuber of Pinellia pedatisecta].

OBJECTIVE: To explore the utility of principal component analysis (PCA) in conjunction with the window preprocessing scheme on chromatographic data for quality control. METHOD: Chromatographic fingerprints of processed tuber of Pinellia pedatisecta were determined by HPLC, and the Window Preprocessing and PCA were used for data processing. RESULT: The quantitative differences among different growing areas and different process batches were found with this method. CONCLUSION: The method can be used in quality control for monitoring between-batch products of traditional Chinese pharmaceutical process.

[Study on traditional Chinese pharmaceutical processing of radix Pinellia pedatisecta].

OBJECTIVE: To study the effects of concentration, volume and temperature of aluminium water on the change of chromatoghraphic data of processed Rhizoina Arisaematis. METHOD: Chromatographic fingerprints of processed Radix Pinellia pedatisecta were determined by HPLC, and multivariate analysis was made for data processing. RESULT AND CONCLUSION: No change on the whole chromatoghraphic data was observed at different levels of concentrition and volume of aluminium water, while there was a great change at different levels of temperature.

[Study on extraction conditions for minute powder of processed rhizome of Corydalis with vinegar].

OBJECTIVE: To examine the factors in extracting the minute powder of processed rhizome of Corydalis with vinegar for its application. METHOD: The one-way variation analysis was used for experimental design. Tetrahydropalmatine was determined with HPLC method. The samples were separated on a Diamonsil C18 column with mobile phase of acetonitrile-TEA phosphate buffer (pH 7.0) (50:50) and detection wavelength was 280 mm. RESULT: The extraction conditions were established as follows: extraction temperature is 60 degrees C, water-added amount is 20 times the weight of the powder, and extraction time is 30 minutes. CONCLUSION: The extraction rate of the tetrahydropalmatine in the minute power is about 10% higher than that of the processed rhizome with vinegar. But less water amount, less time, and lower temperature were needed for extraction. The application of minute powder can reduce energy consumption and raise economic benefit in industrial production.
