The proteome of the late Middle Pleistocene Harbin individual.

Denisovans are a hominin group primarily known through genomes or proteins, but their precise morphological features remain elusive because of the fragmentary nature of the discovered fossils. Here, we report 95 endogenous proteins retrieved from a nearly complete cranium from Harbin, China, dating to at least 146,000 years ago and previously assigned to a new species, Homo longi. This individual has three Denisovan-derived amino acid variants and clusters with Denisova 3, suggesting that the Harbin individual belongs to a Denisovan population. This study fills the gap between morphological and molecular evidence, enhancing our understanding of Denisovans' spatiotemporal dispersal and evolutionary history.

Spatio-temporal context for robust multitarget tracking.

In multitarget tracking, the main challenge is to maintain the correct identity of targets even under occlusions or when differences between the targets are small. The paper proposes a new approach to this problem by incorporating the context information. The context of a target in an image sequence has two components: the spatial context including the local background and nearby targets and the temporal context including all appearances of the targets that have been seen previously. The paper considers both aspects. We propose a new model for multitarget tracking based on the classification of each target against its spatial context. The tracker searches a region similar to the target while avoiding nearby targets. The temporal context is included by integrating the entire history of target appearance based on probabilistic principal component analysis (PPCA). We have developed a new incremental scheme that can learn the full set of PPCA parameters accurately online. The experiments show robust tracking performance under the condition of severe clutter, occlusions, and pose changes.

A nearly modern amphibious bird from the Early Cretaceous of northwestern China.

Three-dimensional specimens of the volant fossil bird Gansus yumenensis from the Early Cretaceous Xiagou Formation of northwestern China demonstrate that this taxon possesses advanced anatomical features previously known only in Late Cretaceous and Cenozoic ornithuran birds. Phylogenetic analysis recovers Gansus within the Ornithurae, making it the oldest known member of the clade. The Xiagou Formation preserves the oldest known ornithuromorph-dominated avian assemblage. The anatomy of Gansus, like that of other non-neornithean (nonmodern) ornithuran birds, indicates specialization for an amphibious life-style, supporting the hypothesis that modern birds originated in aquatic or littoral niches.

A swimming mammaliaform from the Middle Jurassic and ecomorphological diversification of early mammals.

A docodontan mammaliaform from the Middle Jurassic of China possesses swimming and burrowing skeletal adaptations and some dental features for aquatic feeding. It is the most primitive taxon in the mammalian lineage known to have fur and has a broad, flattened, partly scaly tail analogous to that of modern beavers. We infer that docodontans were semiaquatic, convergent to the modern platypus and many Cenozoic placentals. This fossil demonstrates that some mammaliaforms, or proximal relatives to modern mammals, developed diverse locomotory and feeding adaptations and were ecomorphologically different from the majority of generalized small terrestrial Mesozoic mammalian insectivores.

Preparation of massive anti-infective reconstituted bone xenograft and related studies.

The purpose of this study was to develop a new type of anti-infective bone transplantation material. A massive anti-infective reconstituted bone xenograft with calcium phosphate drug core (CPC-MARBX) was prepared; a drug delivery profile and capability of repairing large segmental infected bony defect were characterized with drug delivery tests and in the rabbit model with large segmental infected bony defect. CPC-MARBX was produced with relatively simple procedures. The duration of the drug delivery was about 25 days in vitro and 30 days in vivo. The infected bony defect was successfully repaired with good healing. CPC-MARBX is readily available and can be applied in repairing the large segmental infected bony defect.

Automated extraction of fine features of kinetochore microtubules and plus-ends from electron tomography volume.

Kinetochore microtubules (KMTs) and the associated plus-ends have been areas of intense investigation in both cell biology and molecular medicine. Though electron tomography opens up new possibilities in understanding their function by imaging their high-resolution structures, the interpretation of the acquired data remains an obstacle because of the complex and cluttered cellular environment. As a result, practical segmentation of the electron tomography data has been dominated by manual operation, which is time consuming and subjective. In this paper, we propose a model-based automated approach to extracting KMTs and the associated plus-ends with a coarse-to-fine scale scheme consisting of volume preprocessing, microtubule segmentation and plus-end tracing. In volume preprocessing, we first apply an anisotropic invariant wavelet transform and a tube-enhancing filter to enhance the microtubules at coarse level for localization. This is followed with a surface-enhancing filter to accentuate the fine microtubule boundary features. The microtubule body is then segmented using a modified active shape model method. Starting from the segmented microtubule body, the plus-ends are extracted with a probabilistic tracing method improved with rectangular window based feature detection and the integration of multiple cues. Experimental results demonstrate that our automated method produces results comparable to manual segmentation but using only a fraction of the manual segmentation time.

Model-based automated extraction of microtubules from electron tomography volume.

We propose a model-based automated approach to extracting microtubules from noisy electron tomography volume. Our approach consists of volume enhancement, microtubule localization, and boundary segmentation to exploit the unique geometric and photometric properties of microtubules. The enhancement starts with an anisotropic invariant wavelet transform to enhance the microtubules globally, followed by a three-dimensional (3-D) tube-enhancing filter based on Weingarten matrix to further accentuate the tubular structures locally. The enhancement ends with a modified coherence-enhancing diffusion to complete the interruptions along the microtubules. The microtubules are then localized with a centerline extraction algorithm adapted for tubular objects. To perform segmentation, we novelly modify and extend active shape model method. We first use 3-D local surface enhancement to characterize the microtubule boundary and improve shape searching by relating the boundary strength with the weight matrix of the searching error. We then integrate the active shape model with Kalman filtering to utilize the longitudinal smoothness along the microtubules. The segmentation improved in this way is robust against missing boundaries and outliers that are often present in the tomography volume. Experimental results demonstrate that our automated method produces results close to those by manual process and uses only a fraction of the time of the latter.

Active and dynamic information fusion for multisensor systems with dynamic Bayesian networks.

Many information fusion applications are often characterized by a high degree of complexity because: (1) data are often acquired from sensors of different modalities and with different degrees of uncertainty; (2) decisions must be made efficiently; and (3) the world situation evolves over time. To address these issues, we propose an information fusion framework based on dynamic Bayesian networks to provide active, dynamic, purposive and sufficing information fusion in order to arrive at a reliable conclusion with reasonable time and limited resources. The proposed framework is suited to applications where the decision must be made efficiently from dynamically available information of diverse and disparate sources.

[Forensic application of 9 Y-STRs fluorescence-labeled multiplex amplification system].

OBJECTIVE: In order to increase significantly the discriminatory potential of Y-STR systems available to the forensic community, we have developed a system capable of simultaneously amplifying 9 Y-STR loci by fluorescence-labeled multiplex PCR technique. METHODS: Primers of STR loci DYS434, GATA-A10, DYS438 and DYS439 were labeled with 6-FAM, primers of STR loci DYS531, DYS557, DYS448 were labeled with HEX, and primers of STR loci DYS456, DYS444 were labeled with TAMRA, respectively. PCR products were analyzed using capillary electrophoresis and GeneScan Software on the ABI Prism310 DNA Analyzer. Series experiments were carried out to evaluate the useful value in forensic application such as the sensitivity, male specificity and genotyping DNA different tissues of the same individual. RESULTS: 9 Y-STR loci were exactly determined following optimization of the polymerase chain reaction. In a sample of 120 males, a total of 105 different haplotypes was identified, 97 of them being unique. Overall, haplotype diversity was 0.996 8. It was proved that genotyping of these 9 Y-STR loci in some sexual crime should be prior to that of automosomal STR. CONCLUSION: The results suggest that the newly constructed 9-plex will be very powerful for establishing Y-STR database, population genetic studies and mixture stains identification.

Active and dynamic information fusion for facial expression understanding from image sequences.

This paper explores the use of multisensory information fusion technique with Dynamic Bayesian networks (DBNs) for modeling and understanding the temporal behaviors of facial expressions in image sequences. Our facial feature detection and tracking based on active IR illumination provides reliable visual information under variable lighting and head motion. Our approach to facial expression recognition lies in the proposed dynamic and probabilistic framework based on combining DBNs with Ekman's Facial Action Coding System (FACS) for systematically modeling the dynamic and stochastic behaviors of spontaneous facial expressions. The framework not only provides a coherent and unified hierarchical probabilistic framework to represent spatial and temporal information related to facial expressions, but also allows us to actively select the most informative visual cues from the available information sources to minimize the ambiguity in recognition. The recognition of facial expressions is accomplished by fusing not only from the current visual observations, but also from the previous visual evidences. Consequently, the recognition becomes more robust and accurate through explicitly modeling temporal behavior of facial expression. In this paper, we present the theoretical foundation underlying the proposed probabilistic and dynamic framework for facial expression modeling and understanding. Experimental results demonstrate that our approach can accurately and robustly recognize spontaneous facial expressions from an image sequence under different conditions.

Palaeontology: pterosaur egg with a leathery shell.

The recent discovery of a pterosaur egg with embryonic skeleton and soft tissues from the Yixian Formation confirmed that the flying pterosaurs were oviparous. Here we describe another pterosaur egg whose exquisite preservation indicates that the shell structure was soft and leathery.

Triassic marine reptiles gave birth to live young.

Sauropterygians form the largest and most diverse group of ancient marine reptiles that lived throughout nearly the entire Mesozoic era (from 250 to 65 million years ago). Although thousands of specimens of this group have been collected around the world since the description of the first plesiosaur in 1821 (ref. 3), no direct evidence has been found to determine whether any sauropterygians came on shore to lay eggs (oviparity) like sea turtles, or gave birth in the water to live young (viviparity) as ichthyosaurs and mosasauroids (marine lizards) did. Viviparity has been proposed for plesiosaur, pachypleurosaur and nothosaur sauropterygians, but until now no concrete evidence has been advanced. Here we report two gravid specimens of Keichousaurus hui Young from the Middle Triassic of China. These exquisitely preserved specimens not only provide the first unequivocal evidence of reproductive mode and sexual dimorphism in sauropterygians, but also indicate that viviparity could have been expedited by the evolution of a movable pelvis in pachypleurosaurs. By extension, this has implications for the reproductive pattern of other sauropterygians and Mesozoic marine reptiles that possessed a movable pelvis.

[A study of quadriplex PCR with chimeric primers for short tandem repeats loci].

OBJECTIVE: To establish a simplified method of multiplex PCR based on chimeric primers for STR loci. METHODS: A set of chimeric primers and universal primers were designed to carry out a multiplex PCR for amplifying four short tandem repeats(STR) loci. Then the amplified STR loci were detected by polyacrylamide gel electrophoresis and the gels were sliverstained. RESULTS: A quadriplex STR system was developed on the basis of both chimeric and universal primers. CONCLUSION: The STR multiplex PCR based on both chimeric and universal primers hss been achieved readily and reproducibly by simple adjustment of the individual primer concentrations. The use of chimeric primers provides a method for primer design that eliminates the multiple optimization steps involved in developing the multiplex PCR.

An Early Cretaceous tribosphenic mammal and metatherian evolution.

Derived features of a new boreosphenidan mammal from the Lower Cretaceous Yixian Formation of China suggest that it has a closer relationship to metatherians (including extant marsupials) than to eutherians (including extant placentals). This fossil dates to 125 million years ago and extends the record of marsupial relatives with skeletal remains by 50 million years. It also has many foot structures known only from climbing and tree-living extant mammals, suggesting that early crown therians exploited diverse niches. New data from this fossil support the view that Asia was likely the center for the diversification of the earliest metatherians and eutherians during the Early Cretaceous.

Surface aerosol radiative forcing derived from collocated ground-based radiometric observations during PRIDE, SAFARI, and ACE-Asia.

An approach is presented to estimate the surface aerosol radiative forcing by use of collocated cloud-screened narrowband spectral and thermal-offset-corrected radiometric observations during the Puerto Rico Dust Experiment 2000, South African Fire Atmosphere Research Initiative (SAFARI) 2000, and Aerosol Characterization Experiment-Asia 2001. We show that aerosol optical depths from the Multiple-Filter Rotating Shadowband Radiometer data match closely with those from the Cimel sunphotometer data for two SAFARI-2000 dates. The observed aerosol radiative forcings were interpreted on the basis of results from the Fu-Liou radiative transfer model, and, in some cases, cross checked with satellite-derived forcing parameters. Values of the aerosol radiative forcing and forcing efficiency, which quantifies the sensitivity of the surface fluxes to the aerosol optical depth, were generated on the basis of a differential technique for all three campaigns, and their scientific significance is discussed.

Archaefructaceae, a new basal angiosperm family.

Archaefructaceae is proposed as a new basal angiosperm family of herbaceous aquatic plants. This family consists of the fossils Archaefructus liaoningensis and A. sinensis sp. nov. Complete plants from roots to fertile shoots are known. Their age is a minimum of 124.6 million years from the Yixian Formation, Liaoning, China. They are a sister clade to all angiosperms when their characters are included in a combined three-gene molecular and morphological analysis. Their reproductive axes lack petals and sepals and bear stamens in pairs below conduplicate carpels.

The earliest known eutherian mammal.

The skeleton of a eutherian (placental) mammal has been discovered from the Lower Cretaceous Yixian Formation of northeastern China. We estimate its age to be about 125 million years (Myr), extending the date of the oldest eutherian records with skull and skeleton by about 40 50 Myr. Our analyses place the new fossil at the root of the eutherian tree and among the four other known Early Cretaceous eutherians, and suggest an earlier and greater diversification of stem eutherians that occurred well before the molecular estimate for the diversification of extant placental superorders (104 64 Myr). The new eutherian has limb and foot features that are known only from scansorial (climbing) and arboreal (tree-living) extant mammals, in contrast to the terrestrial or cursorial (running) features of other Cretaceous eutherians. This suggests that the earliest eutherian lineages developed different locomotory adaptations, facilitating their spread to diverse niches in the Cretaceous.

Palaeontology: 'modern' feathers on a non-avian dinosaur.

Discoveries of integumentary coverings on non-avian theropod dinosaurs are becoming commonplace. But the only definitive evidence so far that any of these animals had feathers as we know them today has come from the oviraptorosaur Caudipteryx and the enigmatic coleurosaur Protarchaeopteryx, both of which are considered by some to be secondarily flightless birds. Here we describe the occurrence of pinnate feathers, which clearly feature a rachis and barbs, on a small, non-avian dromaeosaur from northern China. This finding indicates that feathers of modern aspect evolved in dinosaurs before the emergence of birds and flight.
