[LIM protein KyoT2 interacts with human tight junction protein ZO-2-i3].

It was reported that LIM protein KyoT2 negatively regulated transcription by association with the RBP-J DNA-binding protein. Using yeast two-hybrid system with LIM protein KyoT2 as a bait, we have isolated an alternatively spliced form of human tight junction protein 2--ZO-2-i3. Sequence analysis indicated that ZO-2-i3 is composed of 19 exons, and selected usage of exons led to an alteration in the region following the kinase domain as compared with the published sequence. To identify the interaction between LIM protein KyoT2 and ZO-2-i3, yeast two-hybrid system, purification of KyoT2 protein, and GST pull-down assay were performed in the experiments. After KyoT2 and ZO-2-i3 changed vectors, positive two-hybrid yeast was obtained. Using KyoT2 protein and antibody in GST pull-down assay positive result was also obtained. Therefore we conformed KyoT2 interacted ZO-2-i3 in vitro. Furthermore it was identified in yeast that KyoT2 associated with ZO-2-i3 through its LIM2 domain.

Expression of LIM Protein KyoT in Adult Mouse Testis.

The KyoT expression in the adult mouse was reported here for further investigation on the functions of KyoT in adult mouse. To study the expression of mRNA and protein of KyoT, Northern blot, RT-PCR, Immunohistochemical SABC methods and in situ hybridization methods were used in the experiments. Two kinds of KyoT were expressed at high levels in testis of adult mouse, and KyoT immunore activity was mainly located in Leydig's cells. The reactive substance was distributed in cytoplasm rather than in nuclei. KyoT mRNA hybridization signals were also detected in cytoplasm of Leydig's cells rather than in nuclei. The spermatogenic cells and negative controls showed negative results. These results suggest that KyoT was expressed in testis of adult mouse and mainly located in Leydig's cells.

Screening of Genes Encoding Nuclear Localization Signal in a Mouse Embryonic cDNA Library.

A genetic screening system was established previously for the cloning of gene fragments encoding nuclear localization signals (NLS). In the current study, the system was improved and its ability to distinguish nuclear and cytoplasm-localized proteins was verified by inserting a known NLS-encoding gene. A mouse 10.5-d pc embryonic cDNA library was then inserted into the cloning site of the screening vector and 22 out of 10(4) clones were shown to be positive after screening using the system. The cDNA inserts of some of these clones were sequenced, and typical and untypical NLS were identified. The nuclear localizing activity of the cDNA inserts was further verified using the GFP fusion protein system. So, the system is useful in the cDNA library screening of genes encoding NLS-bearing proteins.

Market reform: a challenge to public health--the case of schistosomiasis control in China.

This study examines how the provision of schistosomiasis control has adapted to increased exposure to market forces which has occurred in China over the past decades with the main emphasis on contemporary performance of the services. Financial and service data were collected and analysed from ten schistosomiasis stations in the Hunan province. A document and literature review, key informant interviews, as well as two focus group discussions were conducted to establish their context. The study found that the schistosomiasis control stations had shifted their emphasis from prevention to clinical services and that 62% of the stations' total income now comes from charging for individual clinical services, while 90% of the total costs was related to providing these services. The study found that revenue generation had become the primary motive, and that over-treatment and prescription had become an accepted practice for all the stations. The study concludes that a combination of lax supervision and accountability, and a greater reliance on user-payment and market mechanisms has severely compromised the provision of the public goods elements of the schistosomiasis control programme.
