In the centrosymmetric title polymer, catena-poly[[bis[aqua(1,10-phenanthroline-kappa2N,N')lead(II)]-di-mu3-5-carboxy-3-sulfonatobenzoato-1:2:1'kappa4O3:O1,O1':O1;2':1:2kappa4O1:O1,O1':O3] dihydrate], {[Pb(C8H4O7S)(C12H8N2)(H2O)].H2O}n, each seven-coordinate lead(II) ion is bound by five O atoms from one water molecule and three 5-sulfoisophthalate (sip) anions, and by two N atoms from a 1,10-phenanthroline (phen) ligand. The sip sulfonate group is monodentate. One O atom of the sip carboxylate group is chelated to one Pb2+ cation, with the other also bridging an adjacent Pb2+ cation. The carboxyl group is uncoordinated. This unusual coordination results in a chain structure along the b axis, which is linked by strong intermolecular hydrogen bonds into a three-dimensional network.
BACKGROUND & OBJECTIVE: Chemotherapy, which includes selecting appropriate drugs and suitable dose, is necessary to advanced ovarian carcinoma. Whether dose delivery affects prognosis of ovarian cancer is controversial. This study was to investigate the impact of dose delivery of postoperative first-line chemotherapy on prognosis of stage III C epithelial ovarian carcinoma. METHODS: Clinical data of 47 stage III C epithelial ovarian carcinoma patients, treated with cisplatin-based adjuvant first-line chemotherapy, were retrospectively analyzed. The relative cumulative dose (CD) and dose intensity (DI) were calculated for each drug and for all drugs combined (namely overall CD and overall DI). The impact of dose delivery on progression-free survival (PFS) and overall survival (OS) was analyzed. RESULTS: Median overall CD was 0.76, and median overall DI was 0.53; the median CD of cisplatin was 0.78, and the median DI of cisplatin was 0.57; the median CD of paclitaxel was 0.81, and the median DI of paclitaxel was 0.51. Dose delivery had no impact on PFS and OS of the patients. However, in non-paclitaxel chemotherapy subgroup, the higher overall CD, or CD for cisplatin, or DI for cisplatin, the longer PFS (P=0.01, P=0.03, P=0.01) and OS (P=0.01, P=0.04, P=0.01); in paclitaxel chemotherapy subgroup, dose delivery had no impact on PFS and OS. In suboptimal surgery subgroup (residual disease of > 2 cm), the higher DI for cisplatin, the longer PFS; the higher overall CD, or DI for cisplatin, the longer OS. In optimal surgery subgroup (residual disease of < or =2 cm), dose delivery had no impact on PFS and OS. Residual disease was the only independent prognostic factor of epithelial ovarian carcinoma. CONCLUSIONS: The dose delivery below standard level may influence survival of the patients with residual disease of > 2 cm or treated by non-paclitaxel chemotherapy. The diameter of residual disease is the independent prognostic factor of epithelial ovarian carcinoma.

OBJECTIVE: To study the potential anti-tumor effect of telomerase inhibitors. METHODS: Human oral squamous carcinoma cell line KB was selected as target cell. The effects of 3'-azido-3'-deoxythymidine (AZT) and human antisense phosphorothioate (AS-ONS) for telomerase template on KB cell line were investigated. The cytotoxic effect of AZT and AS-ONS on tumor cells was quantified using MTT colorimetric assay. Assay of 3H-TdR incorporation was undertaken to measure the cell proliferation. The changes of telomerase activity after treatment was detected and quantified by telomeric repeat amplification protocol (TRAP) semi-quantitative analysis. The Flow Cytometry was used to detect apoptosis and measure cell cycle. RESULTS: Both AZT and AS-ONS inhibited the growth of KB cell line in a certain range of concentrations, and meanwhile the telomerase activity was reduced after treatment. In addition, both AZT and AS-ONS can induce apoptosis and arrest G1 phase of cell cycle. CONCLUSION: The results obtained above indicated that AZT and AS-ONS could be potentially used as an anti-oral carcinoma agent or an auxiliary treatment for cancer. Those inhibitory effects might be partially due to the induction of apoptosis and the prolongation of cell cycle.
To study the frequencies of numerical and structural aberrations for chromosome in sperm of benzene exposed workers, the multi-color FISH was used. Four DNA probes(one for chromosome 1 centromere and one for 1 p terminal, and two for chromosome 18 centromere) were hybridized with interphase sperms, and the frequencies of numerical aberrations for chromosome 1, 18 and structural aberrations of chromosome 1 were detected simultaneously. The time weighted average concentration (TWA) of benzene in workplace (42.29 mg/m3) was higher than that of our national maximum allowable concentration (6 mg/m3). The geometric concentration of urinary trans, trans-muconic acid(tt-MA) in exposed group was significantly higher than that of control group. A total of 144,282 sperm of 15 benzene-exposed workers and 135,937 sperm in 14 controls were scored. The frequency of hybridization efficiency was 99.85%. The mean frequencies of disomic sperms for chromosome 1 and 18 in exposed group(0.088% +/- 0.041%, and 0.087% +/- 0.049%, respectively) were statistically increased over that of the control group(0.045% +/- 0.024%, and 0.035% +/- 0.028%), and the mean frequencies of nullisomic sperms for chromosome 1 and 18(0.11% +/- 0.059%, 0.075% +/- 0.035%) in exposed group were statistically increased over that of control group too (0.048% +/- 0.018%; 0.045% +/- 0.024%). The frequencies of diploidy sperm were no difference in both exposed and control groups. The mean frequencies of terminal duplication and terminal deletion for chromosome 1 p(0.16% +/- 0.037%; 0.14% +/- 0.053%, respectively) were significantly increased over that of control group(0.082% +/- 0.023%; 0.069% +/- 0.028%, respectively). The mean frequencies of centromeric duplication and centromeric deletion for chromosome 1(0.10% +/- 0.035%; 0.10% +/- 0.041%, respectively) were significantly increased over that of control group(0.075% +/- 0.023%; 0.060% +/- 0.029%). Our experiments showed that exposed to benzene at higher concentration(42.29 mg/m3) may induce increases in frequencies not only of numerical aberrations for chromosome 1 and 18, but also of structural aberrations for chromosome 1 of sperms in exposed workers.
OBJECTIVES: To assess the effect of freezing on the sperm DNA. METHODS: To assess the sperm DNA preserved at -80 degrees C by using the single cell gel electrophoresis (SCGE). RESULTS: There was no statistical difference on the time factor by analysis of variance (ANOVA). CONCLUSIONS: Integrity of sperm DNA could not be devastated in frozen states.
