[Prevalence of hemorrhagic fever with renal syndrome virus in domestic pigs: an epidemiological investigation in Shandong province].

OBJECTIVE: To investigate the epidemiological significance of hemorrhagic fever with renal syndrome virus (HFRSV) infection in domestic pigs in Shandong province, and study the role of domestic pigs in the prevalence of HFRS. METHODS: Epidemiological investigation was performed in 4 cities of Shandong province. Reversed passive hemagglutination assay (RPHA), reversed passive hemagglutination inhibition (RPHI), HPR-SPA, immunofluorescent antibody (IFA), and reverse transcriptional PCR (RT-PCR) were used to detect antigen and antibody of HFRSV. RESULTS: HFRSV antigen and antibody were detected in the heart, liver, lung, spleen, kidney, blood, urine, and stool of domestic pigs as well as in the sewage of the pigpen facilities. The positivity rate of HFRSV antigen ranged from 3.33% to 5.00% in the organ of pigs, and HFRSV positivity rate in the blood, urine, stool, and sewage was 3.67%, 7.04%, 2.51%, and 5.56%, respectively, with a total serum antibody positivity rate of 1.96%. The virus was isolated from the HFRSV antigen-positive samples, and could infect many organs after artificial infection of the pigs. HFRSV antigen can be detected in suckling rat brain and histopathological examination suggested transient pathological changes in such organs as the liver, lung and kidney. HFRSV may proliferate in pigs and was discharged through multiple routes. CONCLUSION: Domestic pigs can be the host of HFRSV for the viral transmission.

Coordinate regulation of lymphocyte-endothelial interactions by pregnancy-associated hormones.

Precursors of uterine NK cells home to the uterus during early pregnancy from multiple lymphohemopoietic sources. In mouse uterine tissue, pregnancy markedly up-regulates both L-selectin- and alpha(4) integrin-dependent adhesion pathways for circulating human CD56(bright) cells, the phenotype of human uterine NK cells. Based on roles for these adhesion molecules in lymphocyte homing, we examined effects of pregnancy or the steroid hormones 17beta-estradiol or progesterone on lymphocyte-endothelial interactions in secondary lymphoid tissues and in uterus. From preimplantation gestation day 3, specialized high endothelial venules in peripheral lymph nodes and Peyer's patches supported elevated L-selectin and alpha(4)beta(7) integrin-dependent lymphocyte adhesion under shear throughout pregnancy, as compared with high endothelial venules of virgin or postpartum donors. Squamous endothelium from nonlymphoid tissue was not affected. Pregnancy-equivalent endothelial responses were observed in lymph nodes and Peyer's patches from ovariectomized mice receiving 17beta-estradiol and/or progesterone replacement therapy. Adhesion of human CD56(bright) cells to uteri from pregnant or hormone-treated ovariectomized mice was enhanced through L-selectin- and alpha(4) integrin-dependent mechanisms and involved multiple vascular adhesion molecules including mucosal addressin cell adhesion molecule-1, VCAM-1, and peripheral lymph node addressin. Analysis of Tie2-green fluorescence protein transgenic mice demonstrated that CD56(bright) cells adhered primarily to vascular endothelium within the decidua basalis. Microdomain localization of adhesion involving large clusters of lymphocytes was induced on uteri from natural matings, but not pseudopregnancy. Steroid hormones also had independent effects on L-selectin function in splenic lymphocytes that mimicked physiological stimulation induced by pregnancy or fever-range temperatures. These results provide the first evidence for coordinated, organ-specific, steroid hormone-induced changes in lymphocyte homing mechanisms that could contribute to local and systemic immune responses during pregnancy.

Prognostic factors for children with Hodgkin's disease treated with combined-modality therapy.

PURPOSE: Evaluation of pretreatment factors to identify children at high risk for relapse after combined-modality therapy for Hodgkin's disease. PATIENTS AND METHODS: From 1990 to 2000, 328 pediatric patients with clinical stage I to IV Hodgkin's disease were treated with chemotherapy and low-dose involved-field radiotherapy on prospective, collaborative, risk-adapted protocols at three institutions. Pretreatment factors were analyzed by univariate and multivariate analysis for prognostic significance for 5-year disease-free survival (DFS) and overall survival (OS). RESULTS: With a median follow-up of 59 months (range, 8 to 125 months), the 5-year DFS and OS for all patients were 83% and 93%, respectively. Several factors were associated with inferior DFS and OS by univariate analysis. By multivariate analysis, male sex; stage IIB, IIIB, or IV disease; bulky mediastinal disease; WBC more than 13.5 x 10(3)/mm3; and hemoglobin less than 11.0 g/dL were significant for inferior DFS. A prognostic index was developed incorporating the five significant factors from the multivariate analysis, assigning each a score of 1. The 5-year DFS and OS for children with a prognostic score of 0 to 1 were 94% and 99%; score 2, 85% and 96%; score 3, 71% and 92%; and score 4 or 5, 49% and 72%, respectively. There was a significant difference in DFS among each of these groups, with significantly worse OS in those with a score of 4 to 5. CONCLUSION: A prognostic index that was based on five pretreatment factors correlated with inferior DFS by multivariate analysis stratified patients by outcome; this may be useful in assigning children with Hodgkin's disease to risk-adapted therapy.

Bone morphogenic protein-7 (BMP-7), a novel therapy for diabetic nephropathy.

BACKGROUND: Bone morphogenic protein-7 (BMP-7), an essential developmental renal morphogen, is a secreted differentiation factor of the adult collecting duct. It activates receptors in the collecting duct, distal nephron, proximal tubule, and glomerulus. BMP-7 is therapeutic in tubulointerstitial nephritis raising the question of broader efficacy in chronic kidney disease (CKD). METHODS: Diabetes was induced in 200 g rats by a single dose of streptozotocin. After 16 weeks, glomerular hypertrophy and proteinuria were established, and therapy with BMP-7 (10, 30, or 100 microg/kg intravenously twice a week), enalapril (20 mg/kg), or vehicle was begun and continued until 32 weeks. Kidney weight, glomerular filtration rate (GFR), urine albumin excretion, blood pressure, pathology, and BMP-7 expression were measured. RESULTS: Diabetic vehicle-treated rats developed renal insufficiency by 32 weeks (GFR, 0.34 +/- 0.02 mL/min/100 g body weight vs. 0.55 +/- 0.02 in normal). In the diabetic BMP-7 high-dose-treated rats, GFR was preserved (0.70 +/- 0.08, P < 0.01 vs. vehicle), and higher than diabetic enalapril-treated rats (0.58 +/- 0.06). Kidney weights of vehicle-treated animals were not affected, but were reduced in all of the treatment groups (P < 0.001). Proteinuria was reversed to normal by BMP-7 in a dose-dependent manner. The reduction in proteinuria by the intermediate dose of BMP-7 was similar to the effect of enalapril therapy. Glomerular area and interstitial volume were significantly decreased in the BMP-7 and enalapril-treated animals. Glomerular sclerosis was prevented by BMP-7 therapy more effectively than by enalapril. Enalapril controlled hypertension throughout the course of therapy while BMP-7 did not affect blood pressure until the final 4 weeks of therapy. Diabetic vehicle-treated rats lost BMP-7 expression in the kidney. BMP-7 and enalapril therapy restored BMP-7 expression at high levels. CONCLUSION: BMP-7 partially reversed diabetic-induced kidney hypertrophy, restoring GFR, urine albumin excretion, and glomerular histology toward normal. Restoration of BMP-7 expression was associated with a successful repair reaction and a reversal of the ill-fated injury response.

Pharmacokinetics and interactions of a novel antagonist of chemokine receptor 5 (CCR5) with ritonavir in rats and monkeys: role of CYP3A and P-glycoprotein.

The mechanisms of pharmacokinetic interactions of a novel anti-human immunodeficiency virus (anti-HIV-1) antagonist of chemokine receptor 5 (CCR5) [2-(R)-[N-methyl-N-(1-(R)-3-(S)-((4-(3-benzyl-1-ethyl-(1H)-pyrazol-5-yl)piperidin-1-yl)methyl)-4-(S)-(3-fluorophenyl)cyclopent-1-yl)amino]-3-methylbutanoic acid (MRK-1)] with ritonavir were evaluated in rats and monkeys. MRK-1 was a good substrate for the human (MDR1) and mouse (Mdr1a) multidrug resistance protein transporters and was metabolized by CYP3A isozymes in rat, monkey, and human liver microsomes. Both the in vitro MDR1-mediated transport and oxidative metabolism of MRK-1 were inhibited by ritonavir. Although the systemic pharmacokinetics of MRK-1 in rats and monkeys were linear, the oral bioavailability increased with an increase in dose from 2 to 10 mg/kg. The area under the plasma concentration-time curve (AUC) of MRK-1 was increased 4- to 6-fold when a 2 or 10 mg/kg dose was orally coadministered with 10 mg/kg ritonavir. Further pharmacokinetic studies in rats indicated that P-glycoprotein (P-gp) inhibition by ritonavir increased the intestinal absorption of 2 mg/kg MRK-1 maximally by approximately 30 to 40%, and a major component of the interaction likely resulted from its reduced systemic clearance via the inhibition of CYP3A isozymes. Oral coadministration of quinidine (10 and 30 mg/kg) increased both the extent and the first-order rate of absorption of MRK-1 (2 mg/kg) by approximately 40 to 50% and approximately 100 to 300%, respectively, in rats, thus further substantiating the role of P-gp in modulating the intestinal absorption of MRK-1 in this species. At the 10 mg/kg MRK-1 dose, however, the entire increase in its AUC upon coadministration with ritonavir or quinidine could be attributed to a reduced systemic clearance, and no effects on intestinal absorption were apparent. In contrast to rats, the effects of P-gp in determining the intestinal absorption of MRK-1 appeared less significant in rhesus monkeys at either dose.

[Preparation of egg yolk immunoglobulin (IgY) against p53 protein expressed in E.coli].

OBJECTIVE: To isolate immunoglobulin (IgY) from egg yolk of p53 protein-immunized hens and to study its reactivity to the antigens. METHOD: Immunization of egg-laying hens was performed for 3 times at the interval of 14 days with purified p53 protein that had been expressed in E.coli. The eggs laid by these immunized hens were then collected during the whole period of the experiment to prepare IgY from egg yolk by ammonium sulfate precipitation. Final purification and identification of the IgY were performed using SDS-PAGE, enzyme-linked immunosorbent assay and Western blotting. RESULT: All immunized hens developed specific antibodies to p53 protein in contrast to the control ones. The highest titers of the IgY occurred 4 weeks after the first immunization and reached 1:10(6), which remained stable for up to 3 months. CONCLUSION: IgY technology is less costly, non-invasive, fast, simple and highly efficient to generate polyclonal antibodies.

[Construction of recombinant adeno-associated virus carrying hepatitis B surface antigen gene and preliminary study of the gene expression and function].

OBJECTIVE: To construct recombinant adeno-associated virus (rAAV) carrying hepatitis B surface antigen (HBsAg) gene and study the function of the expressed HBsAg. METHODS: HBsAg gene (subtype ayw) was amplified from PTHBV-1 by PCR and cloned into the adeno-associated virus vector pSNAV to form the recombinant pSNAV-HBsAg, which was transfected into BHK-21 cells by means of lipofectamine. Using G418 selection, a mixed cell line, BHK-HBsAg, was isolated, which was capable of HBsAg expression and was subsequently infected with HSV-1-HSV1-rc/Delta UL2 that was able to package the rAAV. After purification, rAAV-HBsAg was obtained. The expression of HBsAg in BHK-21 cells and 293 cells infected with rAAV-HBsAg were detected by enzyme-linked immunosorbent assay (ELISA), and the HBsAg antibodies in the sera of rAAV-HBsAg-immunized BalB/C mice were assayed by radio immunoassay. RESULTS: As detected by ELISA, the expressed HBsAg in mixed cell line mounted to 28.6+/-6.72 ng per 5 x 10(6) cells. The BHK-21 cells and 293 cells infected with rAAV-HBsAg were both capable of HBsAg expression, the amount of which augmented with the increase of multiplicity of infection (MOI). BalB/C mice immunized with rAAV-HBsAg produced anti-HBsAg antibodies. CONCLUSIONS: rAAV-HBsAg can induce humoral immune response against HBsAg and therefore can be a promising candidate hepatitis B vaccine, and in addition, it may serve the purpose of exploring possible immunotherapy for chronic HBV infection.

[Quantitative study of bacteriological infections in dogs undergoing gunshot wound in seawater].

OBJECTIVE: To investigate the changes in bacterial proliferation and the time of infection occurrence in dogs wounded by gunshot in seawater. METHOD: Fourteen canine models of gunshot wound in seawater were established, with another 2 dogs receiving the wound without seawater immersion serving as control. Gross observation and quantitative bacterial analysis were performed at 0, 6, 12 and 24 h after the injury respectively, and two dynamic models for the bacterial growth were set up on the basis of statistical analysis of the results. RESULTS: Bacterial quantity in both of the two groups tended to increase with time, but at each time point after the injury, the seawater group had higher bacterial count than the other group. Within 6 h after the injury, the bacterial count reached the critical point for clinical infection in seawater group. The two bacterial growth dynamics models were (1) Y=9.12 x 10(3+0.247X) and (2) Y=1.35 x 10(3+0.227X) for the seawater group and land group respectively, with the variant X representing time after gunshot injury, and Y the bacterial count per gram tissue. Using the dynamic models, we found that to reach the same bacteria count, the land group needed 4 h more than the seawater group(4.2 h). CONCLUSIONS: Bacterial proliferation is accelerated in the wounds in seawater to result in earlier infection onset with more severity, suggesting that early debridement within 4 h after the injury, complete washing of the wound and early administration of the antibiotics are necessary for the wound management.

[Experimental study of repairing segmental bone defect with reconstituted freeze-dried bone allograft].

OBJECTIVE: To study the effect of basic fibroblast growth factor (bFGF) and hyaluronic acid gel (HAG) combined with freeze-dried bone allograft in repairing segmental bone defect and to explore their mechanism. METHODS: The 15 mm segmental bone/periosteum defects were created on bilateral radius in 50 New Zealand rabbits and were treated with four different kinds of implants on 25 radius respectively (group A: bFGF and HAG combined with freeze-dried bone; group B: bFGF combined with freeze-dried bone; group C: HAG combined with freeze-dried bone; group D: simple freeze-dried bone as a control). The repair of defect was observed radiologically and histologically and were analyzed by radionuclide bone imaging and measurement of calcium contents at different periods. RESULTS: The new bone formation, bone metabolic activity and calcium contents of defects were higher in group A than in group B (P < 0.05), and were higher in group B than in groups C and D (P < 0.05). There were no significant difference between groups C and D. The bone defects healed in the 8th week in group A, in the 10th week in group B, but did not healed in the 10th week in groups C and D. CONCLUSION: As an osteogenetic factor, bFGF promotes the new bone formation; as a slow-release carrier, HAG enhances the effectiveness of bFGF. The combination of bFGF, HAG and freeze-dried bone allograft can repair the segmental bone defect more effectively.

[Pathogenic mechanisms of enterotoxigenic Escherichia coli in guinea pigs].

OBJECTIVE: To explore the pathogenic mechanisms of enterotoxigenic Escherichia coli (ETEC) in guinea pigs. METHODS: Pathological examination of the intestines was performed in ETEC-infected guinea pigs, and the intracellular free Ca2+ concentration, cytoplasmic pH, cell membrane potential and mitochondria membrane potential were determined immunohistochemically and by means of flow cytometry. RESULTS: The guinea pigs were sensitive to the human-origin ETEC, which caused pathological changes in the small intestines such as edema, hyperemia and lymphocyte infiltration, but no bacteria invasion into the epithelial cells was identified. Under transmission electron microscope, the ileal epithelial cells were shown with vacuolar degeneration and evidence of mitochondrial proliferation; heat-labile (LT) and heat-stable (ST) toxins pervaded in the ileal tissue, resulting in significant increase of intracellular free Ca2+ concentration, cytoplasmic pH value and cell membrane potential, with concomitance of significant decrease in mitochondrial membrane potential. CONCLUSIONS: ETEC disturbs the absorption and secretion functions of the intestinal epithelial cells, and induces extensive inflammation in the small intestines of guinea pigs. Besides their action on intestinal epithelial cells, LT and ST also affect the myocytes in the muscularis, and their action sites and mechanism might be similar. ETEC-induced diarrhea is correlated with excessive water excretion from the cells.

[Study on serum p53 protein in cops in Guangzhou city].

OBJECTIVE: Serum p53 protein overexpression was detected in population exposed to traffic exhaust gas to study the relation between traffic exhaust gas and the increased risk in p53 gene mutation. METHODS: Serum p53 protein expression was measured by enzyme-linked immunosorbent assay. Relationship between different types of job and serum p53 protein overexpression were studied by pearson Chi-square tests. RESULTS: Results on serum p53 protein overexpression on jobs outside of office (5.74%) were not significantly higher than jobs inside the office. However, it suggested that traffic police men (12.12%) working outside of office, with whose length of service longer than 30 years had a significant overexpression of serum p53 protein than the others (5.36%) whose length of service was less than 30 years (P < 0.05, OR = 2.43, 95% CI: 1.11 - 5.33). Overexpression rate of p53 protein appeared to be 6.89% in the group whose average weekly exposure hours were more than 40 hours, which was significant higher than the group whose exposed hours were less than 40 hours (P < 0.05, OR = 1.71, 95% CI: 1.03 - 2.81). CONCLUSION: The result suggested that traffic exhaust gas was likely to cause mutation of p53 gene and increasing the incidence of lung cancer.

[Levels and distribution of 15 rare earth elements in tumor and normal lung tissue from the patients with lung cancer].

Ten tumor tissue and normal tissue around tumors were collected from the patients with lung cancer. After digested by mixed acids(HNO3:HClO4 = 10:1), 15 rare-earth elements (REEs) in the tissue were measured directly by ELAN 5000 ICP-MS. Tissue levels and distribution of 15 REEs were analyzed and compared with those in healthy human lung tissue measured in the same condition. Re (rhenium) was used as an internal standard element to compensate for matrit inhibition and sensitivity drift. Recoveries of standard addition for REEs was 92.9%-111.3% and the precision was 0.96%-3.07%. Results showed that levels of 15 REEs in normal tissues around the tumor was as 2.07-2.51 times high as those in the tumor tissue, while the REE distribution curves of them was similar. There were positive abnormalities of Ce and negative abnormalities of Eu in REE curves of both tissue after chondrite-normalized. Compared REE level and distribution pattern in normal tissue with those in tumor tissue from patients with lung cancer, it was found that contents of light REE in lung tissue of health human were much higher while the contents of media and heavy REE were much lower. LREE/HREE in the tumor tissue and normal tissue with lung cancer and health human lung tissue were 12.99, 13.72 and 44.11 respectively. This study suggested that there might be light REEs accumulation in human lung tissue compared with REE distribution in the air particulates and soil. Light REEs accumulation in the lung of healthy people was more obviously than that in patients with lung cancer.

[Effects of acrylamide on DNA damage in human keratinocytes].

OBJECTIVE: To investigate the toxic and DNA damaging effect of acrylamide (AA) on human keratinocytes and its mechanism. METHODS: (1) After the keratinocyte cell line HaCaT cells were exposed to AA with different concentrations for 44 hours, cell survival rate was detected by MTT method. (2) The effects of DNA damage of exposed cells were detected by comet assay. (3) After treating the cells with 2.00 mmol/L of AA plus 0.50 mmol/L of 1-aminobenzotriazole (1-ABT), an inhibitor of cytochrome P-450 enzymes (CYP-450), for 4 hours, the relationship between DNA damage and CYP-450 was studied. RESULTS: (1) Cytotoxicity measurement of AA showed that cell survival rate decreased significantly after 44-hour treatment. (2) Cytotoxicity was not detected after 4-hour AA treatment, but significant DNA damage was observed in all treatment groups, and the degree of damage increased with the concentration of AA. Moreover, the tail lengths of comet cells were in dose-effect relationship. As for cells treated by 1-ABT with 2 mmol/L AA, comet rate and tail length were 15.4% and (8.2 +/- 2.0) micro m respectively, which were decreased significantly (P < 0.01) when compared with 2 mmol/L AA treatment group [80.6% and (44.3 +/- 4.0) micro m]. CONCLUSIONS: Acrylamide has significant cytotoxicity and genotoxicity on HaCaT cells. AA-induced DNA damage may be related to the oxidative metabolite(s) of AA through CYP-450.

[A cohort study on the relationship between blood pressure levels and the mortality of cerebro-cardiovascular diseases in Guangzhou workers].

OBJECTIVE: To study the relative risk (RR) of mortalities of cerebro-cardiovascular diseases (CVD) in Guangzhou workers with hypertension. METHOD: Prospective cohort study was conducted in 78,379 workers, aged >or= 35, from 399 factories. Cox regression model were mainly used for data analysis. RESULTS: (1) There were 48,705 male and 29,674 female workers in the cohort. All workers were followed-up for 8 years. 363 CVD deaths (male 305, female 58) mainly died of stroke, coronary heart disease and hypertension. The crude mortalities were 78.58/100,000 person years in male and 24.55/100,000 person years in female. (2) Compared with the optimal or normal blood pressure (ONBP), the RR (95%) of CVD deaths with high blood pressure (HBP) were 6.19 (4.85 - 7.91) in male and 2.78 (1.59 - 4.85) in female. In male, compared with ONBP, the RR of CVD deaths without illness but with 1st-grade HBP at baseline, and of those suffered non-CVD but with 1st-grade HBP at baseline, and of those suffered CVD with 2nd-grade HBP at baseline were 3.98, 3.25 and 3.15 respectively (P < 0.01). (3) After stratifying of age, smoking, drinking, educational levels and occupational exposure, the RR of CVD deaths was higher in those who were younger, or ever-smoking, non-drinking, higher educational level, exposed to occupational hazards and with hypertension. CONCLUSION: There is relationship between BP levels and CVD mortality. High BP may affect CVD deaths at younger age. Comprehensive measures should be used to reduce the risk of CVD deaths.

[Photocatalytic degradation of low level toluene in the gas phase].

Photocatalytic degradation of low level toluene in the gas phase at low retention time (17s-83s) was studied. The effects of the inlet concentration, retention time, relative humidity, ultraviolet wavelength and carrier of the catalyst were investigated, as well as the change of the photocatalytic degradation capability with the load. It was showed that the degradation rate of toluene at low retention time decreased with increase of inlet concentration, and it decreased more significantly at shorter retention time. Within a range of retention time, the degradation of toluene followed with first order kinetics approximately, and the reaction rate constant increased with inlet concentration. The relative humidity affected the degradation rate a little when the inlet concentration was low. The photocatalytic degradation rate when irradiated with low-pressure mercury lamp (254 nm) was significantly higher than that irradiated with black light lamp having same power. The photocatalyst coated on the stainless steel mesh showed higher activity while it was on the mesh with higher number, however it was not as good as the photocatalyst coated on the aluminum plate.

[Pulmonary function in first generation children of chronic obstructive pulmonary disease (COPD) patients: assessment of related predicting factors].

OBJECTIVE: To study whether familial aggregation exists in pulmonary function impairment and COPD. METHODS: Pulmonary function, smoking habits, history of occupational dust exposure and clinical manifestation among 117 children of 59 smokers with COPD and 55 children of 28 control smokers without COPD. A multivariate linear regression analysis was carried out to study FEV1 among the children. Logistic regression analysis was conducted to study the factors influencing the FEV1 < 70% pred. RESULTS: Height and male gender were positively correlated to children's FEV1 (ml); whereas age, clinical symptoms, history of occupational dust exposure and being children of diseased probands were negatively correlated to children's FEV1 (ml). With other relevant factors adjusted, the FEV1 (ml) was lower among those being children of COPD patients; the more severe the COPD conditions of the parents, the lower the FEV1 of the children. Cigarette consumption having clinical symptoms, and being children of COPD probands showed increased risk of FEV1 below 70% predicted (OR = 1.987). Height, age, and history of occupational dust exposure were not correlated to FEV1 < 70% pred among children. CONCLUSION: Being children of COPD parents is the independent risk factor for lower FEV1 (ml) and FEV1 below 70% pred, which demonstrates the presence of familial aggregation of COPD and pulmonary function impairment.
