A T7-expression system under temperature control could create temperature-sensitive phenotype of target gene in Escherichia coli.

Temperature-sensitive (TS) mutants of a gene are ones of which the activity or phenotype is very similar to that of wild type only at certain temperature and they provide extremely powerful tool for studying protein function in vivo. Here we report a novel strategy to generate TS phenotype of the interest gene in Escherichia coli based on a temperature-sensitive T7-expression system. A TS T7-RNA polymerase is generated by interrupting it with a TS intein from Saccharomyces cerevisiae vacuolar ATPase subunit (VMA), resulting that the gene flanked by T7-promoter and T7-terminator will be transcribed only at the permissive temperature (18 degrees C), not at the restrictive temperature (37 degrees C). The feasibility to create TS phenotype of this strategy was detected using lacZ as target. Reverse transcriptase polymerase chain reaction (PCR) indicated that at 18 degrees C, transcripts of T7-promoter controlled lacZ were at least 85 times more than those at 37 degrees C. Western blot analysis and enzymatic assay showed that large amounts of active His6-tagged LacZ produced at 18 degrees C but little at 37 degrees C. This strategy appears more promising than other TS creation methods because the target is pre-designed, no modification is introduced, and only simple DNA manipulation is required.

Influence of stress on the transmission characteristics of multilayer thin films.

The relationship between multilayer film strains and film thickness changes was analyzed based on the stress formula and the small flexibility principle of elasticity. A theoretical model of the relationship between changes in substrate curvature and film thickness was established, and the formula of every layer thickness with uneven changes of multilayer films was obtained for the spectra of the before-thinning and after-thinning filter. Both the simulated result for a narrow bandpass filter and experimental results showed that nonuniformities in the film thickness caused by substrate curvature changes are one of the main reasons for spectrum degradation in narrow bandpass filters.

Effects of the 1,320-nm Nd:YAG laser on transepidermal water loss, histological changes, and collagen remodeling in skin.

The goal of the study is to investigate the effects of the 1,320-nm Nd:YAG laser on skin barrier function and dermal remodeling. Forty-eight female mice were irradiated with the 1,320-nm laser four times at an interval of 1 week (350 micros, 10 mm, single pass 10% overlap). Three doses were used: 18, 22, and 26 J/cm2. Transepidermal water loss (TEWL) was measured at day 0, 1, 7, 21, 30, and 60 after treatments. Skin biopsies were taken to observe the changes in dermal structures and quantify the increases in the number of fibroblasts and collagen content. The dose of 22 J/cm2 induced the lowest ratio of TEWL (0.58+/-0.06, p < 0.05), highest degree of fibroblast proliferation (45.68+/-5.70, p < 0.05), and the most increase in type I collagen fibers (49.8%, p < 0.05) and hydroxyproline content (85.7%, p < 0.05). The dose of 26 J/cm2 caused marked inflammatory infiltration and the most increase in type III collagen fibers. The increase of type I collagen fibers was greater than type III collagen fibers for all three doses (p < 0.05). Our results demonstrated that 22 J/cm2 was most effective in enhancing skin barrier function and stimulating new collagen synthesis. Dermal remodeling after the 1,320-nm laser was mainly through the synthesis and deposition of collagen type I. Inflammatory reactions were in favor of the formation of type III collagen.

Simultaneous three-photon-excited violet upconversion luminescence of Ce3+:Lu2Si2O7 single crystals by femtosecond laser irradiation.

We found that Ce3+:Lu2Si2O7 single crystals could be excited at 800 nm by using a femtosecond Ti:sapphire laser. The emission spectra of Ce3+:Lu2Si2O7 crystals were the same for one-photon excitation at 267 nm as for excitation at 800 nm. The emission intensity of Ce3+:Lu2Si2O7 crystals was found to depend on the cube of the laser power at 800 nm, consistent with simultaneous absorption of three 800 nm photons. The measured value of the three-photon absorption cross section is sigma'3=2.44x10(-77) cm6 s2.

Expression modification of uncoupling proteins and MnSOD in retinal endothelial cells and pericytes induced by high glucose: the role of reactive oxygen species in diabetic retinopathy.

Uncoupling proteins (UCPs) are mitochondrial transporters present in the inner membrane of mitochondria. They belong to the family of anion mitochondrial carriers. UCPs could act as proton carriers activated by metabolites and create a shunt between complexes of the respiratory chain and ATP synthase. The increased leakiness of the mitochondrial inner membrane to protons may be to minimize superoxide production by limiting the maximum Deltamu(H+). The purpose of this study was to detect UCP expression in retinal capillary cells and their modification in high levels of glucose. The role of reactive oxygen species (ROS) of mitochondria and UCPs in pathogenesis of diabetic retinopathy was investigated. Bovine retinal capillary endothelial cells and pericytes were cultured with selective culture media, respectively. Passage cells were cultured in three different glucose concentrations (5, 23, 30 mM) until passage four. ROS changes in mitochondria of these cells in different glucose concentrations were detected with scanning laser confocal microscopy (SLCM). The mitochondria membrane potential (Deltapsi), cell death rate and apoptosis rate were measured with flowing cytometry. UCP expression in retinal capillary cells was detected by immunocytochemistry. Expression and modification of MnSOD and uncoupling proteins (UCPs) in different concentrations of glucose were detected by means of semi-quantitative RT-PCR. ROS in mitochondria of both endothelial cells and pericytes increased as the glucose concentration of media increased. Deltapsi and cell death rate of endothelial cells increased also. ROS was correlated to Deltapsi and cell death rate positively in endothelial cells. No difference in Deltapsi and cell death rate among different glucose levels was found in pericytes. Apoptosis rate of endothelial cells and pericytes in high glucose levels was higher than that in lower glucose levels. UCP1 and UCP2 were expressed in cultured retinal capillary cells whereas UCP3 was not. At high levels of glucose, expression of UCP1, UCP2 and MnSOD increased to accommodate ROS production compensatively. The compensative mechanism disappeared when glucose concentration was too high (30 mM). The results of this study showed that increasing mitochondrial ROS could be induced by high glucose concentration. Those proteins related to antioxidation mechanism, such as MnSOD and UCPs, could exert compensative action to a certain extent. This compensative action was insufficient when the glucose concentration was too high.

Fabrication of protein-conjugated silver sulfide nanorods in the bovine serum albumin solution.

Highly ordered silver sulfide nanorods conjugated with the Bovine Serum Albumin (BSA) protein have been successfully achieved at ambient temperature. Such a process is very simple and controllable, directly using silver nitrate and thioacetamide (TAA) as the reactants in the aqueous solution of BSA. The products have been characterized by XRD, HRTEM-SAED, SEM-EDS, TG-DTA, FT-IR, and CD spectroscopy. The results of the research show that the as-prepared Ag2S nanorods are monodispersed with sizes about 40 nm in diameter and 220 nm in length, and exhibit a high degree of crystallinity and good photoluminescence. Furthermore, an interesting mechanism is discussed for the formation of the Ag2S nanorods.

Film from peroxidation of an amino phospholipid and its biocompatibility.

A new kind of phospholipid product with fluorescence was derived from autoxidation of an aminophospholipid, L-alpha-phosphatidylethanolamine dilinoleoyl in solid film state. Atomic force microscopy showed that the film product was composed of round and elliptical vesicles with diameters of about 20-45 nm. The product was difficult to dissolve in water and most organic solvents. Fourier transform infrared spectroscopy, solid-state (13)C NMR, and solid-state (31)P NMR indicated that the unsaturated double carbon bond decreased with the reaction, and polymerization within/between phospholipid molecules occurred. Endothelial cells from human vein seeded onto the film showed the highest proliferating activity compared with seeding onto Corning culture plate, glass plate, or phospholipid film with phosphorylcholine head group, as evaluated by 3-(4,5-timethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay (p < 0.001). The hemocompatibility of the film was also assessed by platelet adhesion and protein adsorption. The results suggest that the product has potential applications as a new biomaterial coating.

Comparison of biophysical properties of skin measured by using non-invasive techniques in the KM mice following 595 nm pulsed dye, 1064 nm Q-Switched Nd:YAG and 1320 nm Nd:YAG laser non-ablative rejuvenation.

BACKGROUNDS/AIMS: The aim of the study was to compare the changes of the biophysical properties and to objectify the effects of 595 nm pulsed dye, 1064 nm Q-switched Nd:YAG and 1320 nm Nd:YAG lasers non-ablative rejuvenation by non-invasive techniques. METHODS: KM mice were used for the study. The 595 nm pulsed dye, 1064 nm Q-switched Nd:YAG and 1320 nm Nd:YAG laser treatments were evaluated with biophysical parameter measurements including skin elasticity, skin color, skin trans-epidermal water loss (TEWL) and skin hydration. RESULTS: All three lasers improved the biophysical properties in the skin of KM mice. In skin elasticity measurements, the 1064 nm laser treatment showed the lowest ratio (0.61+/-0.09) while the 1320 nm laser showed the highest one (0.76+/-0.07) on day 60. For erythema values, a significant increase was observed immediately after the 1064 nm laser treatment (196.67+/-19.17), but the lowest values occurred with the 1320 nm laser treatment (189.83+/-16.54). None of the three lasers resulted in obvious changes of skin melanin. TEWL increased immediately after laser irradiation, then began to recover and decreased 60 days after the 595 and 1064 nm laser treatments. With the 1320 nm laser treatment the TEWL began to decrease from day 7 and obtained the lowest mean values (5.23+/-1.13). The water-holding capacity increased initially for the 595 and 1320 nm laser irradiation, while decreased for the 1064 nm laser. At day 60 of the experiment, skin hydration values in all animals were superior to those of the controls. The 1320 nm laser treatment caused the highest ratio (1.29+/-0.26). Both the values of TEWL and skin hydration for the 1320 nm laser treated areas differed significantly from the other two lasers. CONCLUSIONS: Our data showed the 1064 nm Q-switched Nd:YAG laser treatment was most effective in improving the skins' mechanical properties, while the 1320 nm Nd:YAG laser can enhance greatly the skin barrier function and the water-holding capacity. Moreover, we demonstrated the biophysical properties differed considerably between different areas.

Biophysical, histological and biochemical changes after non-ablative treatments with the 595 and 1320 nm lasers: a comparative study.

BACKGROUND/PURPOSE: The objective was to compare the efficiencies of the 595 nm pulsed dye and the 1320 nm Nd : YAG laser non-ablative rejuvenation. METHODS: KM mice were irradiated with the 595 nm pulsed dye and the 1320 nm Nd : YAG lasers. Histological changes were evaluated immediately, 1, 7, 21, 30 and 60 days after the two laser treatments. Skin hydration and hydroxyproline content were measured to quantify the degree of improvement of the skin's water-holding capacity and the rate of hydroxyproline synthesis. RESULTS: Although not statistically significant, the 1320 nm Nd : YAG laser treatment induced 9.7% greater improvement of skin hydration than the 595 nm laser while the 595 nm pulsed dye laser treatment led to a thicker dermis and 8.7% greater increase of hydroxyproline than the 1320 nm laser. More than 50% increase of collagen type I was observed in 75% of 595 nm laser-treated sites and 42% of 1320 nm laser-treated sites, and more than 25% increase of collagen type III was observed in 75% of 595 nm laser-treated sites and 50% of 1320 nm laser-treated sites. The 595 nm laser treatment was better in increasing the amount of collagen fibers, especially collagen type I (P < 0.05). CONCLUSION: Our results demonstrated that the 595 nm laser appeared to be more effective in increasing new collagen formation, while the 1320 nm laser was superior to the 595 nm laser in improving the skin's water-holding capacity.
