Examination of the rate of peptide biosynthesis in neuroendocrine cell lines using a stable isotopic label and mass spectrometry.

The biosynthesis of neuroendocrine peptides is typically examined by following the rate of appearance of a radioactive amino acid into mature forms of peptides. In the present study, we labeled cell lines with L-leucine containing 10 deuterium residues (d(10)-Leu) and used mass spectrometry to measure the biosynthetic rate of gamma-lipotropin in the AtT-20 cell line and insulin in the INS-1 cell line. After 3 h of labeling, both peptides show detectable levels of the d-labeled form in the cells and media. The relative levels of the d-labeled forms are greater in the media than in the cells, consistent with previous studies that found that newly synthesized peptides are secreted at a higher rate than older peptides under basal conditions. When AtT-20 cells were stimulated with KCl or forskolin, the ratio of d- to H-labeled gamma-lipotropin in the medium decreased, suggesting that the older peptide was in a compartment that could be released upon the appropriate stimulation. Overexpression of proSAAS in AtT-20 cells reduced the ratio of d- to H-labeled gamma-lipotropin, consistent with the proposed role of proSAAS as an endogenous inhibitor of prohormone convertase-1. Labeling with d10-Leu was also used to test whether altering the pH of the secretory pathway with chloroquine affected the rate of peptide biosynthesis. In AtT-20 cells, 30 microm chloroquine for 3 or 6 h significantly reduced the rate of formation of gamma-lipotropin in both cells and media. Similarly, INS-1 cells treated with 10, 30, or 60 microm chloroquine for 6 h showed a significant decrease in the rate of formation of insulin in both cells and media. These results are consistent with the acidic pH optima for peptide processing enzymes. Stable isotopic labeling with d10-Leu provides a sensitive method to examine the rate of peptide formation in neuroendocrine cell lines.

[Analysis of differentially expressed lung metastasis-associated proteins in adenoid cystic carcinoma cell lines].

OBJECTIVE: To analyze differentially expressed metastasis-associated proteins in Adenoid cystic carcinoma cell lines of human salivary gland by proteomics. METHODS: Protein expression alterations of ACC-2 and ACC-M cells were described by 2-D gels. After image analysis by software, proteins of interest were excised from the gels and identified by matrix assisted laser desorption ionization time-of-flight mass spectrometer. RESULTS: 12 protein spots showed significantly differential expression patterns between two cell lines. In the identified protein candidates, transketolase, modulator recognition factor 2, Dim1p homolog, splicing factor (arginine/serine-rich 9) and v-Ha-ras l oncogene were all lowly expressed in the poorly metastatic ACC-2 cell and significantly upregulated in highly metastatic ACC-M cell, while type I collagen pro alpha and tumor necrosis factor (ligand) superfamily member 4 showed a high expression in ACC-2 cells and a low expression in ACC-M cells. Pirin (spot 6) just appears in ACC-2 cell and was not detectable in ACC-M cell, while retinal homeobox protein was just detected in ACC-M cell and did not appear in ACC-2 cell. CONCLUSIONS: The proteins may be involved in the adenoid cystic carcinoma lung metastasis through different mechanisms. Our work may contribute to discover diagnostic markers and therapeutic targets.

[Treatment of cervical radiculopathy by laminoplasty and foraminotomy].

OBJECTIVE: To evaluate the results of laminoplasty and foraminotomy in treatment of cervical radiculopathy. METHODS: Of 29 patients, there were 16 males and 13 females, aged 38 to 72 years with an average of 59 years. The reasons of intervertebral foramen stenosis were prolapse of intervertebral disc, osteophyte formation of Luschka joint, spinal canal stenosis combined with thickness of flavum ligmentum and facet joint hypertrophy. The most frequently affected intervertebral foramen were C5.6 and C6.7. The most significant symptoms after impairment of nerve root were reduced sensation, muscle weakness and diminished reflexes. On the basis of laminoplasty, the foraminotomy was performed on the stenotic foramen, including grade I decompression on 13 occasions, degree II on 21 occasions; and double level decompressions were performed on 5 patients. RESULTS: After operation, reduced sensation was recovered most significantly and quickly, and the recovery of muscle weakness followed, while the recovery of diminished reflexes was the slowest and worst. In the followed-up patients, the percentage of excellent and good results was 97%. CONCLUSION: In the cervical spondylotic patients who also have foraminar stenosis, performing laminoplasty with foraminotomy can get good results. If the indication are chosen properly, it can be used widely in clinic.

[The absorption kinetics of silymarin microemulsion in rat intestine].

AIM: To survey the morphology and size distribution of silymarin microemulsion, to investigate the absorption of silymarin microemulsion in rat intestine compared with the absorption of silymarin micelle in rat Jejunum. METHODS: The intestine in rats was canulated for in situ recirculation. RESULTS: The absorption rate constants (Ka) of silymarin microemulsion at the entire intestine, ileum jejunum, duodenum and colon were 6.22 x 10(-2), 2.27 x 10(-2), 1.9 x 10(-2), 1.9 x 10(-2), 1.05 x 10(-2) and 0.43 x 10(-2) h(-1), respectively. The absorption rate constants of two kinds of silymarin micelle at jejunum were 0.36 x 10(-2), 0.65 x 10(-2) h(-1). CONCLUSION: Silymarin microemulsion was well absorbed at the middle and lower segments of intestine in rats. The absorption was a first-order process with the passive diffusion mechanism.

Biopotentials of marine sponges from China oceans: past and future.

An extensive literature survey of over 17 Journals was carried out on Chinese sponges and their natural products in the period from 1980 to 2001. This review is thus intended to provide the first thorough overview of research on marine sponges from China Ocean territories. Information is provided about the rather-limited taxonomic study of Chinese marine sponges, with an analysis on their distribution and diversity. Research findings on the natural products and their bioactivity screening from Chinese sponges are summarized. The weaknesses, gaps and problems in the past R&D program of Chinese sponges are identified, which point to the future opportunities in exploiting these huge untapped sponge resources. The report is expected to serve as an entry point for understanding Chinese sponges and for furthering R&D on their bioactive compounds for new drug development.

Model studies on protein side chain modification by 4-oxo-2-nonenal.

trans-4-Oxo-2-nonenal (ONE) has recently been demonstrated to be a direct product of lipid peroxidation. In earlier studies to elucidate the structure of the trans-4-hydroxy-2-nonenal (HNE)-derived fluorescent Lys-Lys cross-link, we showed that ONE was capable of both oxidative and nonoxidative cross-linking of amines. A more comprehensive study on nonoxidative modification of protein nucleophiles by ONE is described here, focusing on the initial Michael addition of imidazole, thiol, and amine groups to C2 or C3 to give 4-keto aldehydes that can then condense with amines to form nucleophile-substituted pyrroles. 2,3-Substituted pyrroles (major) and 2,4-substituted pyrroles (minor) were distinguished by 2D NMR techniques, and N(tau)-substitution is preferred over N(pi)-substitution in the Michael addition of histidine. Mechanisms of both nonoxidative and oxidative side chain reactions of ONE are discussed, as is the relative propensity (ONE > HNE) to induce cross-linking of the model proteins ribonuclease A and beta-lactoglobulin.

Multi-strips on one gel method to improve the reproducibility, resolution power and high-throughput of two-dimensional electrophoresis.

Two-dimensional polyacrylamide gel electrophoresis is one of the most key separation tools which can reveal hundreds or even thousands of proteins at a time in proteomic research. In this paper, we report a new IPG strip application, called multi-strips on one gel (MSOG) method. By comparing the 2-DE patterns of the same sample, the different state samples and the same sample in the different second dimensional SDS running systems (large size and medium size gels), we found this new method can not only improve the reproducibility and resolution power of 2-DE pattern, but also achieve high throughput and economical format which is helpful to automatic proteomic research.

New Frontiers in the Proteome Research Quantitative Proteomics.

In the past years, proteome research has achieved very exciting advances. The automation of 2DE-MS approach and the use of multidimensional liquid chromatography (LC) and tandem mass spectrometry (MS/MS) have overcome many disadvantages of two-dimensional gel electrophoresis. The stable isotope labeling and the isotope-coded affinity tags (ICAT) strategy provided an ideal method for accurately quantitating the low copy number proteins which play a key role in cells or tissues. At the same time, the research of the quantitative proteomics has been improved greatly with the development of the protein chip technology. Recent advances of the research in the quantitative proteomics and the related technologies are reviewed.
