Salivary flow in borderline hypertension.

Salivary flow was studied in normotensives and in subjects with hypertension. Salivary flow rate was lower in borderline hypertensives than in normotensives. Propranolol, phentolamine and noradrenaline had no influence on salivary flow. Atropine decreased this parameter. The stimulating effect of neostigmine on saliva production was more pronounced in normotensives than in borderline hypertensives. The data support the assumption that in subjects with borderline hypertension the parasympathetic influence on the salivary glands is reduced.

[Diagnosis of avian encephalomyelitis].

The diagnostic value of laboratory examination methods was assessed in the diagnosis of avian encephalomyelitis in seven to twenty-two days old chickens. Histological examination and immunofluorescent test are expedient methods meeting the requirements for timely application of infection-control measures. The agreement of histological examination with the over-all laboratory examination is 70 to 81% and the agreement of the immunofluorescence test is 76%. Bioassay on chickens followed by a histological examination appears to be the best method, providing results with the highest significance. However, it is lengthy, applicable to the necessary demonstration of a virus or to cases when the histological or immunofluorescence examinations are dubious. With the over-all laboratory evaluation the method shows agreement in 95 to 100% of cases. The diagnostic value of the method of virus isolation in chick embryos is reduced by the fact that intercurrent viruses (adenoviruses, IB and others) may be passaged at the same time.

[Effect of the addition of DMSO and EDTA on the activity of formaldehyde and hydrogen peroxide against Bacillus cereus spores].

The question was investigated if DMSO in concentrations between 1 and 6% and 0.1% EDTA could improve the effect of the sporocidal disinfectants formaldehyde and hydrogen-peroxide. In suspension-tests with spores of Bac. cereus it was found, that the sporocidal activity of a 1.2 and 3% formaldehyde solution did not increase, in contrary it decreased significantly if EDTA or DMSO were present in the solution. The same effect could be found with 0.8-5% hydrogenperoxide in combination with the above mentioned substances. These results are not in agreement with the observations of other authors working with vegetative bacteria. It is supposed that DMSO rather reactivates injured spores than that it acts as a carrier for the respective disinfectant. This effect seems to mask a relatively faint increase of the permeability for the disinfectants. Concerning EDTA it could be possible, that it did not succeed in removing the Ca++ from the cortex, in contrary it seems, that it binds itself to the calciumdipicolinate thus stabilizing the spore structure.

[Incidence of Meckel's diverticulum].

A Meckel's diverticulum (M.d.) was seen in 70 children. Among 3233 appendectomies 58 M.d. (1.79%) were observed, in 12 cases the laparotomy was necessary for other reasons. 6 of 70 children (8.5%) with M.d. had complications due to the diverticulum. Ectopic tissue was found in 4 cases. Postoperative complications were not observed at the own patients.

[The ultra-sound diagnosis of Meckel-syndrome (author's transl)].

A case is reported of an antenatal completely sonographic diagnosis of a case of Meckel-Syndrome. During the second ultra-sound screening at 33 weeks gestation the suspicion of hydrocephalus was diagnosed. An extended ultra-sound diagnosis at 34 gestation showed the following: 1. internal hydrocephalus 2. encephalocele or meningomyelocele 3. bilateral polycystic kidneys 4. suspicious of aplasia of the diaphragm on the left 5. suspicious of hepatic cysts. The infant died a few hours after delivery. The autopsy report corroborated the essential features of the ultra-sound diagnosis. In summary these were interpreted as meckel-syndrome. This case shows that a complete antenatal diagnosis of meckel-syndrome by ultra-sound only is possible. The case also shows that diagnosis prior to 24 weeks gestation is desirable in order to carry out the necessary treatment.

Structure-activity relationship in nonsteroidal antiinflammatory agents, including QSAR in fenamate derivatives.

First, it is shown that it is not yet possible to outline a generally valid receptor model for the non-steroidal anti-inflammatory agents. This is demonstrated by three examples of receptor models, namely by the model according to Shen, by the receptor concept of Appleton and Brown, and by the concept concerning chelate complex formation between NSA and heme iron of the cyclooxygenase complex according to Peterson et al. This means that the "custom-made" synthesis of NSA with, perhaps, quantitatively and qualitatively better properties is not yet possible. On the basis of this statement, QSAR investigations may be justified in order to optimize known NSA. In this paper, QSAR calculations on 21 fenamate derivatives are reported. For this, both the multivariate and the univariate Hansch Analysis were used. Altogether, 16 approaches to QSAR has been performed using 1 - 4 biological and 3 - 19 physico-chemical parameters, respectively, with 7 - 21 objects. In all cases a dominating influence of pi has been indicated. Other physico-chemical parameters seemed to be of less importance, e. g., sigma, Es, log xi MR, MV, parachor, and Verloop's steric constants. Possible new fenamate structures of higher biological activity are briefly discussed.

[Investigations on disinfection and sterilization of surfaces by ultraviolet radiation (author's transl)].

Some model investigations have been made in order to find out if disinfection or sterilization in an ultraviolet radiation cabinet (fig. 1) is effective. The reduction rate of spores of Bacillus anthracis dried to germ carriers of aluminium, ceramics or wood was used for indicating the germicidal effect. It was found, that a 7.5 times higher dose of radiation was necessary to reach a similar reduction rate of spores on germ carriers of ceramics as it was found for the ones of aluminium (tab. 1 and 2). Very high dosage of radiation was necessary to get a safe reduction of spores on each germ carrier (fig. 2 and 3). On wooden germ carriers an average logarithmical reduction (R) of no more than 0.67 was found after 30 h of exposure (tab. 3). Unreliable results have been found in the sterilization experiments with spore bearing germ carriers of aluminium and ceramics. In a few rare cases sterility could be obtained by an ultraviolet radiation dose of 2.000.000 microW s/cm2 (tab. 4). But generally a dosage of even 222.600.000 microW s/cm2 was not sufficient to sterilize the copntaminated germ carriers. The ultraviolet radiation cabinet cannot be recommended for disinfection or sterilization.

[Survival of anthrax spores on surfaces (author's transl)].

The survival of spores of Bac. anthracis on the surfaces of wood, resopal, aluminum, teflon, PVC, lacquered sheet metal and ceramics was investigated during a period of 987 days. Two figures are showing the logarithmical reduction rates of germ counts on these materials. The lowest reduction rates were found on wood, the highest on ceramics. On dark surfaces the reduction rate was not as high as on light ones. The increase of relative humidity from 35% to 45% seems to enforce th inactivation of the spores of Bac. anthracis.

[Disinfectant testing with Bacillus spores (author's transl)].

This paper deals with special conditions for testing the sporocidal effect of disinfectants. A method for the production of spore suspensions (Fig. 1) and the negative influence of heating to its resistance against disinfectants was described (Fig. 2). It was shown that spores of a non-virulent strain of Bac. anthracis could be used for this procedure (Fig. 3). In performing the suspension--and the germ-carrier test it was necessary to watch the effect of postinhibition and of inactivators. The application of 10 microliter disposible loops in the suspension-test was recommended. Since the test for the evaluation of the influence of organic matter on the results of disinfection was unreliable (Fig. 4), it was proposed to replace it by invention of a safety-factor. The germ-carrier test should be done with constant amounts of spores, dried to the surface under defined conditions. Exposition of germ-carriers to disinfectants should be done in a chamber with constant temperature and rel. humidity, since the results are strongly influenced by these factors (Fig. 5).

[The differentiation of clostridial microcultures on nuclepore-filters by fluorescent antibodies as a rapid screening test for spores. I. Method of producing microcultures (author's transl)].

Spores of 32 strains of 11 different clostridial species (Table 1) were allowed to germinate as microcultures on Nuclepore membrane-filters to ascertain whether they could be differentiated according to their patterns of growth. The questions dealt with in the preliminary studies, investigated which pore-size of filters would be best for this purpose and which time necessary for each species to produce microcolonies of 10 to 20 cells from a single spore. Nuclepore-filters with a pore-diameter of 1 mu gave the best results and the times for producing microcolonies of the desired size ranged from 4 hours with Cl. septicum up to 10 hours with Cl. tetani (Table 3). The final result of the study was, that the differences in the patterns of growth were not great enough to allow identification. Only Cl. perfringens formed specific and distinctive compact colonies (Tables 4 and 5).

[The differentiation of clostridial microcultures on nuclepore-filters by fluorescent antibodies as a rapid screening test for spores. II. Application of fluorescent antibodies (author's transl)].

The study deals with the question of whether a combination of microcolony-cultivation on polycarbonate-membrane-filters with the fluorescent-antibody-method will lead to a rapid-diagnosis method for clostridial-spores. Formaldehyd-inactivated vegetative cells of eight different clostridial-strains (Table 1) were used for the immunisation of rabbits according to the method published by Böhm and Strauch (5). The results of the immunisation procedure, developed for aerobic-sporeformers, showed, that satisfactory results could not be obtained in all cases (Table 2). The eight conjugates were tested against microcolonies of 32 strains of different clostridial-species on membrane-filters, produced by the technique described in the first communication. The results showed (Table 4) that differentiation was always possible when homologous antisera were used. In most cases, in order to differentiate between the various species, conjugates of several serotypes had to be employed together. However due to their uniform antigenic nature, Cl. feseri and Cl. tetani could both be identified by using one strain for each to immunize. With clostridial-spores the species could be identified within a period of 6 to 12 hours by using the membrane-filter-fluorescent-antibody technique.
