Salt-induced inhibition of the precipitin reaction of concanavalin A with polysaccharides and glycoprotein.

The time course of the precipitin reactions of concanavalin A with glycogen, dextran and ovalbumin was investigated by a light-scattering method near 30 degrees C in 10 mM-Tris/HCl buffer, pH 7.4, containing neutral salts, i.e. NaCl, KCl, NaBr, KI and NaClO4. With 0.8 microM-lectin and 0.36 mg of glycogen/ml, the half-life, t 1/2, of the precipitin reaction was independent of salt concentration between 0.1 M and 1.5 M, and was the same (175s) in the presence of NaCl, KCl, NaBr and KI but was significantly (27%) higher in NaClO4. In contrast, the five salts caused significant to marked enhancement in t 1/2 for the reactions of concanavalin A with dextran and ovalbumin. Likewise, whereas the turbidity produced in 1 h as a result of lectin-glycogen precipitation remained unchanged, those measured for the binding of dextran and ovalbumin were decreased in the presence of three salts. The increase in t 1/2 and decrease in turbidity were found to be higher with NaClO4, followed by KI; NaBr produced moderate and NaCl (or KCl) small but generally significant inhibition of the precipitin reactions with dextran and ovalbumin. The results showed that the lectin-ligand precipitin reactions involve salt-sensitive polar interactions that are less pronounced with compactly folded ligands such as glycogen.

Study of food habits, food practices and taboos in Bangladesh: their implications in nutrition education.

The paper reports the findings of a retrospective study conducted to determine the prevailing food habits, food taboos and practices among 381 mothers selected randomly from twelve villages situated in 4 administrative divisions of Bangladesh. Undesirable food taboos and practices, maldistribution of food, lack of understanding of the nutritional needs of the vulnerable groups and bad cooking practices were found to be widespread in rural Bangladesh. Nutritional and health implications of these are discussed and remedial measures suggested.

Serum-dependent perinuclear accumulation of Cdc42 in mammalian cells.

Cdc42 is a member of the Rho family of GTPase. Cdc42 has been implicated to be involved in the movement, multiplication and transformation of mammalian cells by controlling the rearrangement of actin cytoskeleton and gene expression. But the mechanism of Cdc42 function has not yet been discovered. In this report we present data showing a perinuclear accumulation of Cdc42 in response to fetal bovine serum (FBS). There was no change in the amount of Cdc42 in response to FBS in the cell. It was found that protein component(s) of serum plays a major role in the perinuclear accumulation of Cdc42. Epidermal growth factor has also been found to stimulate the perinuclear accumulation of Cdc42 while NGF has no effect. Kinase inhibitors, quercetin and NDGA were found to block signals for the perinuclear accumulation of Cdc42. This suggests that phosphorylation of cellular proteins is essential for transducing signals generated from the serum component(s) to induce the perinuclear accumulation of Cdc42. These results indicate that redistribution of Cdc42 might be an important step in alteration of gene expression for controlling various functions of the cell including cell division.

[Effect of the neuropeptide tuftsin and its derivatives on metabolism in various structures of the brain].

A study was made of the effect of the neuropeptide tuftsin and its derivatives on some indicators of the metabolism in limbic structures and cortex of the brain in 200 rats. It was shown that tuftsin and its derivatives raise the content of nicotinamide coenzymes at the expense of the oxidized forms of NAD, activate cytochrome-c-oxidase and dehydrogenases largely in the limbic structures. Thus the peptides under study were found to increase the intensity of cell respiration and to enhance intracellular redox processes, i. e. to largely activate the vital activity of the cells of the brain emotiogenous structures.
