Consequences of detergent pollution of the sea: effects on regenerating sponge cubes of Geodia cydonium.

Regenerating cubes of the sponge Geodia cydonium cyconium were used as a model in the investigation of detergent pollution in the sea. The anionic detergent sodium dodecylsulphate (SDS) and a 1:1 mixture of Faks and Radion, two commercial laundry detergents, were used in the concentration range from 1 X 10(-9) g/ml (1 ppb) to 1 X 10(-5) g/ml. It is shown that SDS is taken up, weakly accumulated but not incorporated into the macromolecular fractions of the sponge. At concentrations of 0.1 ppm and above, SDS decreases the uptake of thymidine, uridine and phenylalanine into the acid-soluble sponge fraction. Their incorporation into the acid insoluble fractions, which have been isolated, was different from the controls at 10 ppb and higher levels. Faks and Radion were less active by a factor of 10. However, they showed similar effects. The chemical composition of the regenerating sponge cubes with respect to DNA, RNA and protein content has been evaluated. The alterations are less pronounced on detergent incubation than precursor uptake. The use of the cetyltrimethyl-ammoniumbromide-turbidity-dilution technique reveals drastic qualitative changes in the nucleic acid fractions. The relevant literature on biological effects of detergent is listed. It is shown that this investigation extends the scale of known effects far into the low and pollution-relevant concentration levels.

Mode of action of 9-beta-D-arabinofuranosyladenine on the synthesis of DNA, RNA, and protein in vivo and in vitro.

The influence of 9-beta-D-arabinofuranosyladenine (ara-A) and its 5'-triphosphate derivative on programmed synthesis was tested with an intact cell system as well as with isolated enzyme systems. The effect of ara-A was tested in mouse lymphoma cells (L5178Y). The compound reduces cell proliferation in low concentration by cytostasis; under high ara-A concentration of radioactive precursors into DNA, RNA, and protein showed that ara-A selectively inhibits DNA synthesis. Formation of a polysome complex is not affected by ara-A. [3H]ara-A is incorporated into DNA in an intact cell system; 1 molecule of ara-A is incorporated per 8000 molecules of deoxyadenosine. Most of the ara-A molecules appeared to be in internucleotide linkages. Incorporation of ara-A into RNA could not be detected. 9-BETA-D-Arabinofuranosyladenine 5'-triphosphate (ara-ATP) does not reduce the incorporation rate of the following enzymes, isolated from quail oviducts: DNA-dependent RNA polymerases I and II, polyadenylic acid polymerase, and poly(adenosine diphosphate ribose) polymerase. The compound was found to inhibit DNA synthesis catalyzed by DNA polymerases isolated from quail oviducts and from oncogenic RNA viruses (Rous sarcoma viruses). All the enzymes tested were inhibited by ara-ATP in a competitive way with respect to deoxyadenosine 5'-triphosphate. The highest affinity of ara-ATP, i.e., the highest inhibitory potency of the drug, was found in the assays with the eukaryotic low-molecular DNA-dependent DNA polymerase. The influence on the eukaryotic high-molecular DNA-dependent Dna polymerase was a litte less. Compared to the eukaryotic DNA polymerases, the viral enzymes (RNA-directed DNA polymerase and DNA-directed DNA polymerase) are affected to a smaller extent by ara-ATP. No effects of ara-A and ara-ATP are observed in a protein-synthesizing, cell-free system isolated from L5178Y cells.

[Dwarfism in pericentric inversion of the X-chromosome].

We report on a 9 2/12 year old girl with pericentric inversion of the X-chromosome. It was diagnosed accidentally by a chromosome-analysis led through because of small stature. She bears a strong phenotypic resemblance to her mother (normal chromosomes), carrier of the pericentric inversion however is the phenotypically normal father. The breakpoint of the X-chromosome not lying within the critical region suggests that the patient will neither suffer a gonadal dysfunction. Both short stature and phenotypic dysmorphic features have familial cause.
