B and T cell abnormalities in patients with primary IgA nephropathy.

The in vitro function of B and T cells was studied in 16 patients with primary IgA nephropathy (PIgA-N). The distribution of OKT3+ cells (total peripheral T cells) and of regulatory T cell subsets (helper OKT4+ and cytotoxic/suppressor OKT8+ cells) was evaluated and a testing for 47 HLA-A, B, C, DR, and DQ antigens was carried out in the 16. B lymphocyte IgA production, after stimulation by pokeweed mitogen in the presence of T cells from normal donors treated with mitomycin C, was significantly greater in patients than in controls. T lymphocytes from patients with PIgA-N were more efficient than T cells from controls in providing IgA specific helper activity for normal B cells. The analysis of the individual data showed that the overactivity of B cells and the T cell operational dysfunction was present in about 50% of the patients and did not correlate. No numerical imbalance between T lymphocyte subsets nor any association between lymphocyte behavior, HLA antigen distribution, and a number of clinical, laboratory, and immunohistological findings was observed. Our data, therefore, suggest that PIgA-N is an immunologically heterogeneous disease and that an IgA-specific B cell overactivity and/or overall IgA-specific T cell helper activity may be present in some patients.

[Immunochemical determination of immunoglobulins and lysozyme in normal human saliva - principal assay technics].

We have studied a method to determine the concentrations of low level of immunoglobulins and lysozyme in saliva. The concentration of IgG, IgA and IgM in human whole saliva were measured by fluorimetric method and the concentration of lysozyme by turbidimetric method. The results of our study in 18 normal subjects (15 females and 3 males), have showed the geometric mean concentration (microgram/ml) IgG 5,1; IgA 177; IgM 2,6 and lysozyme 117.

[Fluorometric method for the assay of IgG produced in lymphocyte cultures].

We have studied the IgG secreted in a system of CML. Microquantity of IgG (30-3000 micrograms/ml) of culture was determined using a fluorometric method based on the determination of the fluorescence derived by immune-complex IgG-FITC labeled goat Anti-human antibody to IgG covalently coupled to hydrophilic beads. This techniques has showed a good sensibility and accuracy.

[Lipid variations in coronary disease].

Levels of total cholesterol (TC), triglyceride (Tg), high-density-lipoprotein-cholesterol (HDL-C), apolipoproteins A and B (ApoA and ApoB), were assayed in the serum of 63 patients suffering from coronary atherosclerosis, who survived myocardial infarction (M.I.) compared to healthy subjects. TC was assayed by an enzymatic-colorimetric technique: after precipitation with PEG 6000 the same method was used to determine HDL-C. Tg were assessed using total enzymatic method and ApoA and ApoB by RID. On the basis of TC, Tg, HDL-C, ApoA and ApoB values we have identified 6 main patterns, presenting a very characteristic lipaemic model. No significant difference was found between survivors and controls as regards TC and Tg; on the contrary, there was a statistical difference between the two groups for HDL-C, ApoA and ApoB.

[Urinary hydroxyproline in advanced breast carcinoma and response to treatment with medroxyprogesterone acetate (MAP) in high doses].

The Authors have studied total urinary hydroxyproline (TUH), together with other hematochemical analyses, in 104 patients suffering from advanced breast cancer with or without bone metastases. TUH escretion is higher in patients with osseous lesions and, if simultaneous pathologic values of TUH and serum alcaline phosphatase occur, the presence of bone metastases is statistically even more probable. Moreover TUH seems to vary in accordance with the clinical response of the disease at osseous level in 46 patients treated with Medroxyprogesterone acetate (MPA) at high doses, showing itself as a possible marker of not negligible clinical value.

[Variation of the TG lymphocyte subpopulation in the aged].

Peripheral blood from 20 healthy subjects of whom 10 were young (15 - 30) and 10 aging (73 - 85) was examined for the proportions of TG lymphocyte population by two different methods TG cells proportions were significantly (P 0.001) increased in aging subjects when compared with the young subjects.

[Thea value and limits to kinetic analysis of human serum Ig. Preliminary note].

We have considered accuracy and sensitivity of a new kinetic method called "Fixed time", in order to determine human Ig. The method based on velocity of the reaction, considers the increase in turbidness, which is due to immune-complex formation during a constant and predetermined period of time from the start of the reaction to when equilibrium is achieved. Experience with testing times demonstrates that the best results were obtained by introducing a variant to the method adopted up to now. According to that method, velocity reaction was usually estimated in a period of time comprised between the 1st and the 6th minute, while turbidness is now estimated after 15" (first time) and after 3'15" (final time) from the beginning of the reaction.

[Infrared spectroscopy in the quantitative determination of urinary calculi constituents (author's transl)].

Infrared spectroscopy of urinary calculi can be used for the quantitative determination of the major constituents in mixed stones; the method is simple and consists in area-measurements of specific absorption peaks of the spectrum of each compound. In the area-measurement method the average error is +/- 2,5% in calcium-oxalate-apatite and +/- 2% in ammonium magnesium phosphate-carboapatite mixtures; within these limits the calculus matrix do not affect significantly the value of areas. An investigation of 64 mixed renal stones shows that in 57% of the samples, apatite is present in the nucleus; uric acid and calcium-oxalate are the most frequent superficial compounds, while the external layer of ammonium magnesium phosphate and the mixture uric acid-calcium phosphate are rare events. In calcium-oxalate-apatite and ammonium magnesium phosphate-carboapatite calculi the quantitative composition of the nucleus and the external layer differ significantly. The proposed method of analysis of internal and external structures in renal calculi can be useful for the study of etiology, for the prevention and the treatment of urolithiasis.

[Analysis of urinary calculi. Study methods].

A comparative study of the composition of a hundred urinary gravel stones is carried out, made by chemical analysis and spectroscopy infra-red. This research has shown that the spectroscopic method is better than chemical analysis: the latter method is excessively sensitive, above all when considering magnesium, phosphates and uric acid. The marked chemical disclosure of these substances takes us to false conclusions regarding the composition of the gravel stone with percentages varying between 20 and 40% of the cases. On the other hand chemical analysis has not revealed the presence of oxalates in 24% of the gravel stones examined. The spectroscopic method has shown itself to be simpler and faster to execute (a part from being more sensitive and precise), and capable of furnishing precise information on the composition of gravel stones, even very small ones of a weight of 500 microgram.

One-step purification by high-performance liquid chromatography of retinal S-antigen.

A number of subsequent chromatographic techniques are commonly used, over a period of a few days, in order to obtain pure retinal S-antigen (S-Ag), a 50,000-dalton molecular weight protein with strong uveitopathogenic properties. A rapid high-performance liquid chromatographic technique employing a TSK-G3000 SWG gel-filtration column is described for the purification of bovine S-Ag. This single chromatographic step produced S-Ag preparations of high purity and recovery when compared to the conventionally isolated S-Ag.
