The incorporation of [1-14C] acetate into the methyl ketones that occur in steam-distillates of bovine milk fat.

1. The (14)C-labelling of the fatty acids and the methyl ketones in steam-distillates of milk fat from a lactating cow that had been injected intravenously with [1-(14)C]acetate was determined. 2. The labelling patterns of the C(6)-C(16) fatty acids and the corresponding methyl ketones with one fewer carbon atoms were similar, particularly so for the C(5)-C(10) compounds at 9 and 22hr. after the injection of [1-(14)C]acetate. The isolation of (14)C-labelled methyl ketones in the range C(3)-C(15) is evidence that the beta-oxo acid precursors, which are glyceride-bound in the milk fat, are synthesized in the mammary gland from acetate. The absence of heptadecan-2-one in steam-distillates and the extremely low specific radioactivity of stearic acid are further evidence for this biosynthetic pathway. 3. The specific radioactivities of the C(5)-C(15) methyl ketones were higher (with the exception of C(9) methyl ketone in the second milking) than the specific activities of the corresponding fatty acids with one more carbon atom. This is consistent with the methyl ketone precursors' being formed during the biosynthesis of fatty acids rather than being products of beta-oxidation of fatty acids.

Determination of size distribution of fat globules in intravenous fat emulsions by photon correlation spectroscopy.

A photon correlation spectroscopy method has been developed to characterize the size distribution of fat globules in intravenous fat emulsions (IFE) in terms of mean diameter, standard deviation of the distribution, and percentage of large particles outside the distribution. Mean fat globule diameters of samples of all IFE products available in Canada were about 0.3 microns, similar to values reported in the literature. The methodology is sufficiently sensitive to detect the presence of 5% by weight of 2 microns polystyrene microspheres in an intravenous fat emulsion. The effect of changes in instrument settings and variables on the results has been evaluated.

Liquid chromatographic method for identity, assay, and content uniformity of five tricyclic drugs.

A liquid chromatographic (LC) procedure has been developed for the assay, content uniformity, and identification of single active ingredient solid and liquid formulations of amitriptyline, chlorpromazine, imipramine, thioridazine, and trifluoperazine. The drugs are extracted from their formulations with methanol or dilute hydrochloric acid, and identified by comparison of retention times with those of known standards; drugs are quantitated against these standards with dl-norephedrine hydrochloride as the internal standard. The precision of replicate injections is better than 2.5% for peak area and better than 1% for peak height. The precision of triplicate determinations of tablet composites is better than 2.2%.

Titrimetric determination of nonesterified fatty acids in intravenous fat emulsions.

A titrimetric method, suitable for use at a limit of 5 mEq/L, has been developed for the determination of total nonesterified fatty acids in intravenous fat emulsion preparations. The method differentiates titrant consumed by the nonesterified fatty acids from that consumed by egg yolk phospholipids, usually present as an emulsifying agent. The total nonesterified fatty acids in 8 products from 4 manufacturers were in the range from 0.4 to 3.8 mEq/L. The mean standard deviation of the method is 0.09 mEq/L.

Analysis of selected drug formulations for volatile nitrosamines.

A number of selected drug formulations on the Canadian market were analyzed for N-nitrosodimethylamine (NDMA) and N-nitrosodiethylamine (NDEA) content by using a gas chromatograph interfaced to a thermal energy analyzer (GC/TEA). Of the 34 formulations analyzed, NDMA was found in 21 at levels up to 12 ppb and NDEA was found in 11 at levels up to 0.6 ppb. In many cases, the drug entity in the formulation did not contain the precursors for the observed nitrosamines.

Analysis of piperazine drug formulations for N-nitrosamines.

A quantitative method is described for the measurement of N-mononitrosopiperazine (NPIP) and N,N'-dinitrosopiperazine (DNPIP) in drug formulations containing piperazine, using a gas chromatograph interfaced to a thermal energy analyzer (GC/TEA). The method has detection limits of 20 ppb for NPIP and 12 ppb for DNPIP. In a survey of 6 products available on the Canadian market, all contained NPIP at levels of 0.38-15.3 micrograms NPIP/g piperazine and none contained any detectable amount of DNPIP.
