A new prodrug of paclitaxel: synthesis of Protaxel.

2' and 7 Polyol carbonates of paclitaxel were synthesized and screened as potential paclitaxel prodrugs. Paclitaxel is released from 7-(2",3"-dihydroxypropylcarbonato) paclitaxel (Protaxel) at rates inversely proportional to pH, by an intramolecular cyclization. Compared to paclitaxel, maximum tolerated i.v. or i.p. doses (MTD) of Protaxel are about 2.5- to 3-fold higher; its efficacy is substantially higher in human cancer line xenografts in athymic mice, especially in prostate PC-3, breast MDA-MB 468 and ovary OVCAR-1.

Histological and electron microscopic analysis of tissue response to synthetic composite bone graft in the canine.

A synthetic composite bone graft material, composed of fibrillar collagen, hydroxyapatite, and tricalcium phosphate, was evaluated in canine femoral shaft defects and compared to ungrafted defects and defects grafted with autogenous bone. The results of mechanical testing of the grafted femora in torsion at 1 year postoperatively were previously reported. This report details the histological and microscopic features of the graft sites. The results of this study suggest that the healing of the graft site is not yet complete at 1 year and that healing may be more complete in defects that received the synthetic graft as compared to ungrafted sites and autogenous bone graft. The bone tissue appears to respond to the graft particles as if they were bone particles.

Effect of race and gender on neurological level in myelomeningocele.

The association of race and gender with different neurological levels of myelomeningocele was studied in 251 patients. Over-all, the white to black ratio was 3.6 and the male to female ratio was 0.86. However, the proportions of whites and females were significantly increased in thoracic-level patients (white to black ratio 13.6, male to female ratio 0.43), whereas the lumbar-level patients had sex and white to black ratios equivalent to the area population. This supports the concept that thoracic-level myelomeningocele has a different pathogenesis from lumbar-level.

Determination of niacin metabolites 1-methyl-5-carboxylamide-2-pyridone and N-1-methylnicotinamide in urine by high-performance liquid chromatography.

A test suitable for detecting latent niacin deficiency was developed. It measures the 24-h urinary output of the two major metabolites of niacin, 1-methyl-5-carboxylamide-2-pyridone and N-1-methylnicotinamide. The two metabolites were isolated from urine using separate ion-exchange extractions. They and their two internal standards were quantitated simultaneously by high-performance liquid chromatography using a reversed-phase ion pairing separation. Detection was by absorbance at 254 nm.
