Sex differences in systemic and local immune responses to Entamoeba histolytica after intraperitoneal and rectal immunization in Balb/c mice.

The antibody responses against Entamoeba histolytica in Peyer's patches and spleen after rectal, intraperitoneal and intragastric immunization with glutaraldehyde-fixed amebas (GFA) was compared between male and female mice by the use of a spot forming cell (SFC) assay. We found that female mice elicit significant higher anti-amebic SFC responses than male mice, in both spleen and Peyer's patches after rectal and intraperitoneal stimulation, whereas by intragastric route females show higher responses than males in spleen but not in Peyer's patches.

Active immunization in hamsters with live Entamoeba histolytica trophozoites of low virulence.

Live trophozoites of Entamoeba histolytica of low virulence (LV) were inoculated intraportally in hamsters. One week later they were challenged intrahepatically with highly virulent (HV) trophozoites of the same strain and sacrificed 1 week later. Significant protection manifested by the production of smaller lesions was observed in immunized animals. Antiamebic IgG levels were significantly higher in the immunized group. Histologically, livers of immunized animals presented small lesions containing foamy macrophages, epithelioid cells, fibroblasts and devoid of amebas. Lymphocyte and plasma cell infiltrates were seen periportally.

Kinetics of the anti-amebic antibody producing cells response in Peyer's patches and spleen after both local and systemic stimulation in Balb/c mice.

The kinetics of the anti-amebic antibody producing cell (APC) response was analyzed by a spot forming cell (SFC) assay, after a single dose of glutaraldehyde fixed amebas (GFA) by intragastric, rectal and intraperitoneal routes. It was found that mice elicited both local and systemic immune responses in Peyer's patches and spleen, by either local or systemic stimulation. The analysis of the isotype of the anti-amebic APC in mice given four times the GFA by the mentioned routes revealed that mice produced anti-amebic APC of the three major isotypes in both spleen and Peyer's patches.

Morphological characterization of experimental amebic liver lesions in gerbils.

Gerbils are susceptible to amebic liver abscess (ALA) production under experimental conditions. However, little is known about the histopathological changes that occur during the evolution of the lesion. We analyzed microscopically the sequence of cellular events of ALA in gerbils inoculated intraportally or intrahepatically with E. histolytica. At early stages trophozoites were associated with different degrees of inflammatory reaction. Amebas were also observed in areas devoid of inflammatory cells. Damaged hepatocytes were either related or not to inflammatory reaction. At late stages, lysis of macrophages and epithelioid cells were seen in relation to necrotic tissue and trophozoites. We conclude that amebic liver necrosis in gerbils at early stages may result from lysis of inflammatory cells as shown previously by us in hamsters, but also by direct contact and lytic activity of amebas to hepatocytes. The late destruction and expansion of hepatic tissue necrosis is apparently associated with damage of macrophages and epithelioid cells.

[Prevalence of dental caries and rampant caries in a preschool population].

The results from a study on prevalence of ordinary dental caries and rampant caries in a children population are presented. The study was carried out at Dentistry Out patient Service in the Children Hospital "San Juan de Aragón" of the Federal District Department. The DMF index was obtained and its was proposed another one, in order to classify the different degrees of rampant caries in function of dental damage. An interview was applied to the fathers of the children of the study group in order to determinate hygiene oral habits, eating and familiar antecedents that could influence in the process of the ordinary and rampant caries and to compare between them. Serum and saliva were analyzed to determinate levels of albumin and IgM, IgG and IgA; there were also others factors tested that could indicate caries development.

Polyclonal activation of B lymphocytes in patients with amoebic hepatic abscess.

In the course of tissue invasion by the amoeba, cellular immune responses are generated with the production of lymphokines by T lymphocytes that could induce activation of B cells. Therefore, it is possible that besides the induction of B lymphocytes specific for amoebic antigens, there could be activation of B cells with different specificities, that is, polyclonal in nature. IgM antibodies against some natural and synthetic antigens were quantified in both sera from amoebic hepatic abscess (AHA) patients and healthy controls by means of an ELISA. Titers in each case analyzed were significantly higher in AHA patients than in controls. Results indicate that there is polyclonal activation of B lymphocytes in this disease.

[Mast cells in Peyer's patches in the mouse].

Mast Cells participate in immediate and late hypersensitivity activities, immunoregulation and inflammation. Recently two groups of them have been detected on the basis to its morphology, content of granules and function: a) Those who are localized in connective tissue and b) Those of the intestinal mucosa. Those of the intestinal mucosa are T dependent and proliferate in parasitic as well as in intestinal hypersensitivity conditions to diverse antigens, in turn, Peyer's Patches (PP) are considered antigen catcher and initiators of intestinal immune responses; these are subject to the influence of diverse substances, several of which are within mast cells (for example: histamine, prostaglandins, etc); for which in this work we investigate the morphology relation between PP and mast cells. Balb/c mice small intestine segments with muscular layer and PP of proximal, middle and distal levels where studied; were histologically processed, toluidine blues stained and mast cells counted in different intestinal layers (PP underlying zone, rest of the muscular, corium and submucosa. Data were analyzed by the paired double T test for mean differences. A greater Quantity of mast cells were observed at the marginal zone of the PP in comparison to the rest of the muscular layer, submucosa and corium. The abundance of mast cells in relation to the PP possibly indicates its modulatory influence on the function of lymphoid cells of the PP.

Induction of humoral immune response to Salmonella typhimurium in mouse Peyer's patches.

The mechanisms of immune response generation and regulation at the intestinal level are not well known, mainly due to the lack of suitable and reproducible methods to measure local immune responses. The Cunningham direct and indirect hemolytic plaque assay for the quantification of antibody producing cells against Salmonella in Peyer's patches of mice orally infected with Salmonella typhimurium was used. After infection IgM and IgG producing cells were determined on days 6, 9, 12 and 19. Specific antibody producing cells appeared after bacterial invasion of Peyer's patches, nine days after infection. At this time, there were more antibody producing cells in the distal part of the intestine, which correlated with a higher infection of these Peyer's patches as detected by bacterial culture. After day nine, the number of plaque forming cells was similar in both parts of the intestine. The peak of response was on day 12 and diminished on day 19. The number of IgM and IgG producing cells was similar in all days analyzed. Histological analysis of Peyer's patches of infected mice showed inflammation with disorganization and tumefaction.
