Use of monoclonal antibodies for radioimmunoassay of water buffalo milk progesterone.

In order to standardize a radioimmunoassay of milk progesterone as a routine method for confirmation of oestrus and diagnosis of pregnancy in water buffalo, monoclonal antibodies against progesterone were produced. Hybridomas were prepared by fusing spleen cells from a Balb/c mouse immunized with progesterone 11 alpha-hemisuccinate-bovine serum albumin conjugate with the mouse myeloma cell line NS-1. Thirty wells out of 94 secreted anti-progesterone antibodies. Of the ten independent hybridomas derived, one (AF65) was suitable for the quantification of milk progesterone by radioimmunoassay. The tracer used in the assay was progesterone-11 alpha-hemisuccinate [( 2-125I]iodohistamine). The sensitivity of the assay was 50 pg/tube. The mean progesterone concentration at oestrus was 0.8 +/- 0.2 ng/ml increasing to 8.5 +/- 0.8 ng/ml 24 d later in pregnant animals.

Visual evoked potentials and CSF IgG at different stages of multiple sclerosis: a possible correlation.

Visual Evoked Potentials (VEP) have been investigated in 102 patients affected by Multiple Sclerosis (MS), classified both by disease stage and by optic pathway status. VEP abnormalities were significantly correlated with the clinical stage and increased CSF IgG levels. The delay in the VEP latencies and the CSF IgG values proved to be increased in parallel in the worsening phases of MS. The diagnostic value of these findings is also discussed.

Immunogenetics of the McB1 macroglobulin allotype in cattle.

The paper describes a cattle serum antigen (McB1) which is controlled by a dominant gene (McB1), independent from those controlling the two macroglobulin markers McA1 and McA2 as well as from that controlling the low-density lipoprotein marker Ld1A1. McB1 is located on a high-molecular-weight serum protein which, very likely, is an alpha2-macroglobulin.
