Ultra-high resolution array painting facilitates breakpoint sequencing.

OBJECTIVE: To describe a considerably advanced method of array painting, which allows the rapid, ultra-high resolution mapping of translocation breakpoints such that rearrangement junction fragments can be amplified directly and sequenced. METHOD: Ultra-high resolution array painting involves the hybridisation of probes generated by the amplification of small numbers of flow-sorted derivative chromosomes to oligonucleotide arrays designed to tile breakpoint regions at extremely high resolution. RESULTS AND DISCUSSION: How ultra-high resolution array painting of four balanced translocation cases rapidly and efficiently maps breakpoints to a point where junction fragments can be amplified easily and sequenced is demonstrated. With this new development, breakpoints can be mapped using just two array experiments: the first using whole-genome array painting to tiling resolution large insert clone arrays, the second using ultra-high-resolution oligonucleotide arrays targeted to the breakpoint regions. In this way, breakpoints can be mapped and then sequenced in a few weeks.

Non-small-cell lung carcinoma. Current and future therapeutic management.

Non-small-cell lung cancer (NSCLC) is one of the most common malignancies in developed countries and accounts for millions of deaths worldwide. The incidence of NSCLC correlates with smoking tobacco and is likely to increase in those countries with increasing per capita consumption of tobacco. NSCLC is particularly associated with morbidity--in the US several published clinical studies involving a variety of intensive surgical, chemotherapeutic and radiotherapeutic interventions have produced only a 10% 5-year survival. New technologies and breakthroughs in treatment notwithstanding, the current emphasis in clinical trials is to refine and optimise available therapies to maximise patient disease-free and overall survival. These accruing multi-institutional clinical trials may offer surgical resection with or without neoadjuvant therapy, multiagent chemotherapy, and/or standard or hyperfractionated radiation therapy. Since most NSCLC patients have inoperable disease, the most active current trial design entails chemoradiotherapy, since previous trials appear to suggest superior results to radiotherapy alone in these patients.

Restrictive dermopathy: a report of three cases.

We report three infants with a rare syndrome of restrictive dermopathy, in which rigidity of the skin at birth is associated with characteristic facial anomalies, generalised arthrogryposis, bony abnormalities, and lung hypoplasia. The skin has a distinctive pathology with compaction of the dermal collagen and fibrosis of the subcutaneous tissue. The inheritance is likely to be autosomal recessive and the condition appears to be fatal in the early neonatal period.

Monocyte markers and the common acute lymphoblastic leukemia antigen on chronic lymphocytic leukemia cells.

The mononuclear cells from 42 patients with chronic lymphocytic leukemia (CLL) were analyzed for surface markers using monoclonal antibodies and flow cytofluorimetry. All 42 CLLs were Ig+HLA.DR+OKT3-OKT4-OKT6-OKT8- and monoclonal with respect to light chain type. Thirty of the 42 tumors expressed the common acute lymphocytic leukemia antigen, and cells from 18 leukemias stained with the antibodies OKM1 and 63D3, which are specific for different antigens characteristic of myeloid cells. Eight of the leukemias failed to stain with PI153, which has been reported to recognize all B-cell CLLs.
