Photobleaching of 1,3-diphenylisobenzofuran by novel phthalocyanine dye derivatives.

As part of a wider programme to identify novel photosensitizers for photodynamic therapy, the ability of a number of phthalocyanine dyes, including some novel copper phthalocyanine derivatives with a range of water solubilities, to produce potentially cytotoxic species in solution was examined. The experiments were performed in dimethylformamide using 1,3-diphenylisobenzofuran (DPIBF) as the scavenger. The study revealed that all the dyes tested produced DPIBF photobleaching on illumination in vitro, but with widely different (greater than 12x) rates. The possible correlation of DPIBF photobleaching rates with a number of the dyes' properties is discussed.

Coloured complexes of all-trans-retinal with benzocaine and other local anaesthetics.

Coloured Schiff base complexes with lambda max values of 500 nm or above were formed between the visual pigment all-trans-retinal and the local anesthetics procaine (lambda max = 533 nm), benzocaine (4-amino-benzoic acid ethyl ester, lambda max = 535 nm), 3-amino-benzoic acid ethyl ester (lambda max = 500 nm) and 2-amino-benzoic acid ethyl ester (lambda max = 509 nm) in methanol acidified with HCl. The anaesthetics lidocaine and urethane failed to form coloured compounds with lambda max values greater than 500 nm under the same conditions. The relevance of these observations to the effect of anaesthetics on the visual pigments is discussed.

The photokilling of bladder carcinoma cells in vitro by phenothiazine dyes.

The potential photodynamic therapy photosensitizers Methylene Blue, Azure C, Methylene Violet, Thionine, Methylene Green, Haematoporphyrin, Nile Blue A, chloroaluminium phthalocyanine and bis-aluminium phthalocyanine were examined for their photoeffects and dark toxicity against a human superficial bladder carcinoma cell-line. By examination of [3H]thymidine uptake into dye-treated cells after irradiation with a copper-vapour pumped dye laser, it was found that Methylene Blue was the most phototoxic and dark toxic of all the dyes tested, suggesting that the dye might be of some use as a topically applied photodrug for use in photodynamic therapy of superficial or early-recurring carcinomas.
