Origin and phylogeographical structure of Chinese cattle.

Complete mitochondrial D-loop sequences of 231 samples were used to explore the origin and genetic diversity of Chinese cattle. Phylogenetical analysis of these sequences revealed both Bos taurus and Bos indicus mitochondrial types in Chinese cattle. Four of the previously identified mitochondrial DNA lineages (T1-T4) were identified in the Bos taurus type, including lineage T1, which was found for the first time in Chinese cattle. Two lineages (I1 and I2) were identified in the Bos indicus type. Our results support the suggestion that the Yunnan-Guizhou Plateau is the domestication site of Chinese zebu. We also found evidence that Tibetan cattle originated from taurine and zebu cattle. The distribution pattern of Chinese cattle breeds was closely related to the geographical and climatic background. It was possible to divide Chinese cattle in this study into two major groups: northern and southern cattle.

Formation of amorphous alloys by ion beam mixing and its multiscale theoretical modeling in the equilibrium immiscible Sc-W system.

Unique amorphous alloys are synthesized at the compositions of 25 and 40 atom % of W by ion beam mixing in the equilibrium immiscible Sc-W system characterized by a positive heat of formation of +14 kJ/mol. In thermodynamic modeling, a Gibbs free energy diagram is constructed based on Miedema's theory, and the diagram predicts a glass-forming range of the Sc-W system to be within 12-58 atom % of W. To develop an atomistic model, ab initio calculations are first conducted to assist the construction of an n-body Sc-W potential under the embedded atom method. The proven realistic potential is applied in molecular dynamic simulations to study the crystal-to-amorphous transition in the Sc-W solid solutions, thus determining the glass-forming ability of the system to be within 15-50 atom % of W. Apparently, both theoretical predicted glass-forming ranges cover the experimentally measured one, showing an excellent agreement. We report, in this paper, the experimental results from ion beam mixing and the multiscale theoretical modeling concerning the amorphous alloy formation in the Sc-W system together with a brief discussion of the structural transition mechanism.

Surgical repair of patients with tetralogy of Fallot and unilateral absence of pulmonary artery.

UNLABELLED: BACKGROUND; Patients with tetralogy of Fallot and unilateral absence of pulmonary artery are a high-risk group for whom there is no consensus on the correct approach to medical management. The purpose of this report is to review a 29-year experience in the treatment of those patients. METHODS: Between May 1966 and February 1995, 2,511 patients underwent correction of tetralogy of Fallot in our department, 24 of those patients with unilateral absence of pulmonary artery (20 had absence of the left pulmonary artery, 4 had absence of the right pulmonary artery). Valved conduits were used in 9 patients, right ventricular patches were used in 4 patients, and transannular patches with a monocusp that was made of the patient's pericardium were used in 11 patients. RESULTS: There were two operative deaths; both were in patients with hypoplasia of the left ventricle. All survivors had good early and late results. CONCLUSIONS: A right ventricular patch should be used in patients with tetralogy of Fallot and infundibular stenosis; a transannular patch with a monocusp should be used in patients with tetralogy of Fallot and stenosis of the left or right pulmonary artery's origin as well as the pulmonary trunk. A homograft valved conduit is suitable for patients with anomalous coronary artery or pulmonary atresia.

Evaluation of surgical effect on cardiovascular anomalies associated with corrected transposition of great arteries.

From September, 1985 to April, 1994, 33 patients underwent surgical repair of corrected transposition of the great arteries associated with other congenital heart anomalies. Of them, 31 patients were SLL type, the other 2 patients were attributed to IDD type. Operations were performed on ventricular septal defect in 28 patients, atrial septal defect in 15, pulmonary stenosis in 29 and two had valvuloplasty for left atrioventricular valve regurgitation, patent ductus arteriosus closure was performed on one and modified Fonton procedure on one. There were five peri-operative deaths being 15.7% of operative morbility; late death was only one patient. The following-up of the survival 27 patients for 2 months to 9 years showed a satisfactory results.

Micronucleus formation in peripheral-blood lymphocytes from smokers and the influence of alcohol- and tea-drinking habits.

In recent years, studies on the genotoxic effects of smoking and its modifying factors have been widely followed with interest. In this report, frequencies of micronuclei (MNF) in peripheral-blood lymphocytes in 220 healthy smokers have been detected by the micronucleus test by finger-skin puncture, which needs only 1 to 2 drops of peripheral blood. On the basis of analysis of matched-pair data, the modifying effects of alcohol and tea are discussed. Our results show that (I) smoking significantly increases MNF in lymphocytes as compared with healthy non-smokers (p less than 0.01); (2) alcohol significantly increases MNF in smokers (p less than 0.01); (3) tea decreases MNF induced by smoking (p less than 0.01). Therefore, the anti-carcinogenic role of tea merits further study.

Levels of direct-acting mutagens, total N-nitroso compounds in nitrosated fermented fish products, consumed in a high-risk area for gastric cancer in southern China.

A high gastric cancer mortality in Fujian province (Peoples Republic of China) has been associated with the consumption of certain salted fermented fish products such as fish sauce (FS). We have investigated the levels and nature of N-nitroso compounds (NOC) and genotoxins present, before and after nitrosation, in 49 FS samples collected from villages in this high-risk area, pooled into six samples. The concentrations of total NOC before nitrosation ranged from 0.2 to 16 mumoles/l, and after nitrosation at pH 2 and pH 7, they rose by up to 4800- and 100-fold, respectively. In nitrosated samples, 40-50% of total NOC was not extractable into organic solvents; volatile N nitrosamines accounted for 1-2% and N-nitrosamino acids for 8-16% of total NOC. None of the FS samples exhibited genotoxic activity, but after nitrosation all were weakly active in the SOS chromotest. The highest SOS-inducing potency was observed with nitrosated ethyl acetate extracts of most samples. The formation of methylating agents was measured by incubation of nitrosated FS with DNA and subsequent analysis of 7-methylguanine adduct. 2 of the 6 nitrosated FS samples caused a slight increase in DNA methylation. 1 pooled home-made FS sample (the only one tested) contained tumour promoter-like substances, as measured by expression of certain EBV genes in Raji cells. HPLC fractionation of ethyl acetate extracts of FS samples allowed identification of three UV-absorbing peaks that, upon nitrosation, produced direct-acting genotoxins. This genotoxicity was partly ascribed to the formation of nitrite-derived arene diazonium cations that were characterized by a coupling reaction with N-ethyl-1-naphthylamine and thin-layer chromatography.

[Operation for discrete subvalvular aortic stenosis].

33 patients with discrete subvalvular aortic stenosis, membranous in 14 cases, fibromuscular rige in 16 and tunnel in 3 were treated surgically in the past ten years. This spectrum of abnormalities can be differentiated by echocardiography and angiocardiography. Our policy is to excise the membrane or fibromuscular rige. When the outflow tract has remain narrow owing to concentric thickening of that portion of the septum and free wall proximal to aortic annulus, a septal myomectomy is indicated. Because the operation of subaortic stenosis is safe and effective, once the diagnosis is established, operation is a reasonable surgical treatment alternative. Operative mortality rate was 6%. Follow-up results of all survivors are satisfactory.

Role of nitrosamides in the high risk for gastric cancer in China.

Gastric cancer is the leading cause of death from cancer in China. Samples of fish sauce, a traditional seasoning, were collected in the high-risk area for gastric cancer in the Fuzhou area, Fujian Province. When fish sauce samples were nitrosated at pH 2.0, direct mutagenicity and high contents of N-nitrosamide were detected (30.9-78.0 microM); the N-nitrosamide content of three samples of fish sauce made in Guangdong and purchased from a market outside Fujian were low (2.1-6.0 microns). When the nitrosated fish sauce extract was given to newborn rats by gavage, dysplasia and adenocarcinoma were induced in the glandular stomach in the 4th and 16th experimental week, respectively. N-Nitrosamides were also found in fasting gastric juice from patients with chronic gastritis in the high-risk area of Putian. The mean concentration of total N-nitrosamides in the extracts correlated with the severity of gastritis in the stomach. These findings indicate that N-nitrosamides may play an important role in causing gastric cancer in China.

[The influence of castration on the induction of gastric adenocarcinoma by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Wistar rats].

Six-week old non-castrated and castrated Wistar rats of both sexes, 20 in each group, received a continuous dose of 83 micrograms/ml MNNG in drinking water for 32 weeks. All animals were autopsied at the 52nd week for pathological examination. The incidence of the gastric adenocarcinoma was 31.6-50.0%, and no significant differences were obtained statistically between different MNNG-treated groups. No tumor was found in the control group. Quantitative and qualitative studies of the intramucosal cyst (IC) showed that the ICs might be classified into simple and dysplastic. The former was subdivided into 5 groups and the commonest type was fundic cyst (81%). In MNNG-treated groups, the number of fundic cysts was decreased; nevertheless, the neck cell cysts were increased in number markedly. The number of dysplastic cysts in the MNNG-treated groups was significantly increased and this important precancerous lesion was closely related with the genesis of gastric adenocarcinoma.

[Studies on the gap junctional intercellular communication (GJIC) of human stomach carcinoma cells in comparison with normal cells and the effect of the tumor promoter, TPA].

This work was conducted by using a rapid and simple technique, scrape-loading and dye transfer (SLDT) to study GJIC of human stomach carcinoma MGC-803 cells in comparison with normal WB rat liver cells, Chinese hamster V79 cells and a primary culture of chicken embryonic myoblasts. Cells were plated and grown overnight to confluency in 35 mm plastic dishes in appropriate media. Monolayered cells, after rinsing in PBS, were immersed in the mixed 0.05% Lucifer Yellow (MW 457.2) and 0.05% Rhodamine-Dextran (MW. 10,000) in PBS. Scrape loading was performed by utilization of a sharp knife. Cells were incubated in dye solution for an additional 3 min. at room temperature before rinsing with PBS and observation under fluorescent microscope. Cells competent in GJIC showed transfer of Lucifer Yellow from the injured border to interior cells while the high MW. Rhodamine-Dextran dye stayed in situ in the loaded cells. Cells incompetent in GJIC did not show dye transfer; both Lucifer Yellow and Rhodamine-Dtranex were retained in the original loaded cells of the injured border. The background cell monolayer away from the scrape line was dark indicating that none of the dye molecules could permeate through cell membrane in the conditions described. It was found that human stomach carcinoma MGC-803 cells lack GJIC; Chinese hamster V79 cells showed modest GJIC; WB rat liver cells and chick myoblasts showed marked GJIC. The tumor promoter, TPA(1-100 ng/ml), inhibits GJIC of the normal cells efficiently. An inhibitor of calmodulin, Trifluoperazine (TFP) (5-20 microM), evidently increased the GJIC of stomach carcinoma MGC-803 cells. Noteworthy is that TFP in the dosage range used in SLDT experiments showed inhibitory effect on cell growth and DNA synthesis of MGC-803 cells documented in parallel experiments. These results indicate that the lack of GJIC in MGC-803 cells correlates with their uncontrolled cell proliferation; the improvement of GJIC correlates with the inhibition of tumor cell proliferation. TPA inhibition of GJIC in normal cells in this work confirmed previous reports. Interestingly, it was found that when V79 cells were treated with TFP and then shifted to medium containing both TFP and TPA, GJIC was blocked. It is likely that TPA overcomes the effect of TFP on GJIC of MGC-803 cells. These results provide further evidence for the role of GJIC in carcinogenesis, specially the tumor promotion phase.

[Mutagenicity of gastric juice in patients with chronic gastritis].

Mutagenicity of fasting gastric juice from 108 chronic gastritis patients in high- and low-risk areas of stomach cancer was assayed using in vitro SCE test in cultured human lymphocytes and micronucleus test in cultured Chinese hamster V79 cell. The results showed that mutagenicity could be found in this gastric juice. The positive rate of these two tests, SCE/cell, micronucleated cell rate and micronucleus rate were significantly higher in high-risk area than in low-risk area (P less than 0.01). There was a tendency that the mutagenic activity was higher in high pH than in low pH gastric juice and in males than in females. In high-risk area, the mutagenicity of gastric juice was obviously higher in younger than in elder aged group. Because no S9 was added in this test system, the results seem to suggest that the mutagenicity found in the gastric juice of patients with chronic gastritis might be, at least in part, related to direct-acting N-nitroso-compounds synthesized in the stomach.
