Solution structure of microcin J25, the single macrocyclic antimicrobial peptide from Escherichia coli.

The three-dimensional solution structure of microcin J25, the single cyclic representative of the microcin antimicrobial peptide class produced by enteric bacteria, was determined using two-dimensional 1H NMR spectroscopy and molecular modeling. This hydrophobic 21-residue peptide exhibits potent activity directed to Gram-negative bacteria. Its primary structure, cyclo(-V1GIGTPISFY10GGGAGHVPEY20F-), has been determined previously [Blond, A., Péduzzi, J., Goulard, C., Chiuchiolo, M. J., Barthélémy, M., Prigent, Y., Salomón, R.A., Farías, R.N., Moreno, F. & Rebuffat, S. (1999) Eur. J. Biochem., 259, 747-755]. Conformational parameters (3JNHCalphaH coupling constants, quantitative nuclear Overhauser enhancement data, chemical shift deviations, temperature coefficients of amide protons, NH-ND exchange rates) were obtained in methanol solution. Structural restraints consisting of 190 interproton distances inferred from NOE data, 11 phi backbone dihedral angle and 9 chi1 angle restraints derived from the coupling constants and three hydrogen bonds in agreement with the amide exchange rates were used as input for simulated annealing calculations and energy minimization in the program XPLOR. Microcin J25 adopts a well-defined compact structure consisting of a distorted antiparallel beta sheet, which is twisted and folded back on itself, thus resulting in three loops. Residues 7-10 and 17-20 form the more regular part of the beta sheet. The region encompassing residues Gly11-His16 consists of a distorted beta hairpin, which divides into two small loops and is stabilized by an inverse gamma turn and a type I' beta turn. The reversal of the chain leading to the Phe21-Pro6 loop results from a mixed beta/gamma turn. A cavity, in which the hydrophilic Ser8 side-chain is confined, is delimited by two crab pincer-like regions that comprise residues 6-8 and 18-1.

[Detection of Helicobacter pylori by polymerase reaction in bile samples from gallbladder and bile stones].

Some papers report helicobacter pylori existence in bile from surgical specimens obtained during gallbladder or bile ducts surgery. The aim of this work was search by PCR, H. Pylori presence in bile specimens from patients suffering of gallbladder stones or by bile ducts stones. Bile samples were obtained by gallbladder punction during cholecystectomy in 26 patients, 19 of them with gallbladder stones and 7 also with gallbladder stones and bile duct stones. Age ranged from 22-69 years old, median 49.6 years old. Samples were sent to specialized biomolecular laboratory to perform PCR techniques. Two of 26 patients (7.6%) had positive reaction for the presence of DNA of H. Pylori in bile samples. Our research suggest that DNA of H. Pylori can be founded in bile samples patients with gallbladders and duct stones in Argentina.

gamma-Amino-beta-hydroxybutyric acid as add-on therapy in adult patients with severe focal epilepsy.

Total seizure frequency was evaluated among adult patients with severe partial epilepsy refractory to medical treatment. Clinical status was assessed before, and after a synthetic analog of gamma-amino-beta-hydroxybutyric acid (GABOB) was used as add-on therapy. Twenty-five patients with temporal and frontal lobe epilepsy were included in the trial. They received GABOB 250 mg twice daily during a 26-week period. Concentrations of concomitant antiepileptic medication were not modified. Twenty-five percent of the patients showed a 50% reduction of the total seizure frequency. There were no serious adverse effects related to medication.

[Esophageal involvement in progressive systemic sclerosis].

Systemic sclerosis is a collagen disease with high frequency of oesophageal affection, specially if we use manometry as an evaluation method that has a high rate of clinic complication in high rate of patients. The aim of the study was to evaluate the prevalence of the oesophageal affection through the clinical, radiological, endoscopical and manometrical studies and to compare the results with other authors. Between January 1978 and February 1993, we evaluated 101 patients with different clinical types of scleroderma 89 females and 12 males with ages between 16-83 (mean: 49, 19 years). In order to determine systemic sclerosis diagnosis following Masi Rodman criterion's we found from 101 cases 83 had dysphagia (82.2%) motor 66 (65%) organic 17 (34.7%) GER was seem in 41 cases (40.5%). According to Heitz's radiological classification: I: 26 patients (25%) II: 58 patients (52.4%) III: 19 patients (18.8%) IV: 17 patients (16.9%). In reference to Neschis manometric classification we found II 88 (76%) III 77 (76%) In accordance with endoscopical classification we found I: 16 (15.8%); II: 49 (48.5%); III: 19 (18.8%); IV: 17 (16.9%); Although this motor and organic oesophageal alterations have an evident diagnostic value, they have also been referred in another metabolic and collagen diseases but with lesser frequency than was found in the systemic sclerosis by us.

Association of triiodothyronine binding activity to soluble adenylate cyclase in testicular preparations.

Cytosolic adenylate cyclase activity from rat seminiferous tubules was purified by chromtography in DEAE-cellulose, hydroxylapatite and Bio-Gel A-0.5 m as well as by centrifugation in sucrose gradients. In all these purification steps, fractions with adenylate cyclase activity also contained binding activity for L-T3. Binding studies indicate the existence of two L-T3 receptor components associated to adenylate cyclase activity. The component exhibiting the highest hormone affinity has the lowest binding capacity.
