Hybrid atomistic-coarse-grained treatment of multiscale processes in heterogeneous materials: a self-consistent-field approach.

A treatment of multiscale quasistatic processes that combines an atomistic description of microscopic heterogeneous ("near") regions of a material with a coarse-grained (quasicontinuum) description of macroscopic homogeneous ("far") regions is presented. The hybrid description yields a reduced system consisting of the original atoms of the near regions plus pseudoatoms (nodes of the coarse-graining mesh) of the far regions, which interact through an effective many-body potential energy V(eff) that depends on the thermodynamic state. The approximate nature of V(eff) gives rise to "ghost forces," which are reflected in spurious heterogeneities close to interfaces between near and far regions. The impact of ghost forces, which afflict all previous hybrid schemes, is greatly diminished by a self-consistent-field hybrid atomistic-coarse-grained (SCF-HACG) methodology. Tests of the SCF-HACG technique on a fully three-dimensional prototypal model [Lennard-Jones (12,6) crystal] yield thermomechanical properties (e.g., local stress) in good agreement with "exact" properties computed in the fully atomistic limit. The SCF-HACG method is also successfully used to characterize the grain boundary in a Lennard-Jones bicrystal.

Influence of quantum size effects on island coarsening.

Surface x-ray scattering and scanning-tunneling microscopy experiments reveal novel coarsening behavior of Pb nanocrystals grown on Si(111)-(7 x 7). It is found that quantum size effects lead to the breakdown of the classical Gibbs-Thomson analysis. This is manifested by the lack of scaling of the island densities. In addition, island decay times tau are orders of magnitude faster than expected from the classical analysis and have an unusual dependence on the growth flux F (i.e., tau is approximately 1/F). As a result, a highly monodispersed 7-layer island height distribution is found after coarsening if the islands are grown at high rather than low flux rates. These results have important implications, especially at low temperatures, for the controlled growth and self-organization of nanostructures.

Coarse-grained free-energy-functional treatment of quasistatic multiscale processes in heterogeneous materials.

A new treatment of quasistatic (reversible) multiscale processes in heterogeneous materials at nonzero temperature is presented. The system is coarse grained by means of a finite-element mesh. The coarse-grained free-energy functional (of the positions of the nodes of the mesh) appropriate to the thermodynamic-state variables controlled in the relevant process is minimized. Tests of the new procedure on a Lennard-Jonesium crystal yield thermomechanical properties in good agreement with the "exact" atomistic results.

Hybrid atomistic-coarse-grained treatment of thin-film lubrication. I.

A technique that melds an atomistic description of the interfacial region with a coarse-grained description of the far regions of the solid substrates is presented and applied to a two-dimensional model contact consisting of planar solid substrates separated by a monolayer fluid film. The hybrid method yields results in excellent agreement with the "exact" (i.e., fully atomistic) results. The importance of a proper accounting for the elastic response of the substrates, which is reliably and efficiently accomplished through coarse-graining of the far regions, is demonstrated.

Deficient neurogenesis in forebrain-specific presenilin-1 knockout mice is associated with reduced clearance of hippocampal memory traces.

To examine the in vivo function of presenilin-1 (PS1), we selectively deleted the PS1 gene in excitatory neurons of the adult mouse forebrain. These conditional knockout mice were viable and grew normally, but they exhibited a pronounced deficiency in enrichment-induced neurogenesis in the dentate gyrus. This reduction in neurogenesis did not result in appreciable learning deficits, indicating that addition of new neurons is not required for memory formation. However, our postlearning enrichment experiments lead us to postulate that adult dentate neurogenesis may play a role in the periodic clearance of outdated hippocampal memory traces after cortical memory consolidation, thereby ensuring that the hippocampus is continuously available to process new memories. A chronic, abnormal clearance process in the hippocampus may conceivably lead to memory disorders in the mammalian brain.

A new murine oxidative stress model associated with senescence.

A murine oxidative stress model was established via ozone inhalation, which was identified by detection of the response of antioxidant defense system, levels of oxidative products and effects of natural antioxidants on this model. Male BALB/c mice were exposed to 1.2 mg/m(3) ozone for 10 h per day. The control group was exposed to flowing air. From inhaling ozone, mice were killed at day 5, 10, 15 and 20, respectively. Exposure to ozone made mice show the increase of malondialdehyde (MDA) contents in heart, kidney and liver, as well as 8-OHdG levels in urine, and resulted in cytological nuclear concentration in brain neurons or thymocytes. Ozone exposure also impaired antioxidative capacity such as the decrease of total antioxidation capacity (TAC) in sera, reduced glutathione (GSH) in sera or thymus and glutathione-S-transferase (GST) activity in spleen or thymus but not in liver. Correlation analysis showed the significant inverse correlation (r=-0.894, P<0.05) between thymus weight index and inhalation doses of ozone. Meanwhile, thymocyte in model mice proliferated more poorly than normal controls. Catechin and clove extract could reverse parts of changes above induced by ozone inhalation. These results suggest that exposure to ozone can result in an increased production of reactive oxygen species in vivo, which causes oxidative stress. The mice under oxidative stress showed senescence-related alterations in physiological parameters as well. Taken together, our data demonstrates that an oxidative stress model in mice has been successfully established by ozone inhalation, which would be helpful to probe the relationship between oxidative stress and senescence and evaluate effects of antioxidants.

Toxicity studies of Rhizoma Polygonati Odorati.

The toxicity of the water-soluble extract of Rhizoma Polygonati Odorati was studied in the present investigation, which included its acute toxicity, chronic toxicity, and genetic toxicity. The aqueous extract of Rhizoma Polygonati Odorati did not cause seriously abnormal signs or death to animals in the acute toxicity test and in the 6-month chronic toxicity test. Neither was genetic toxicity found in the Ames test, the micronucleus test of bone marrow and the sperm malformation test in mice.

Hypoglycemic effects of aqueous extract of Rhizoma Polygonati Odorati in mice and rats.

Water soluble extract of Rhizoma Polygonati Odorati (RPO) was studied for its hypoglycemic effect in diabetic mice and rats. Results showed that RPO significantly lowered hyperglycemia caused by starch loading in both normal and diabetic mice. Four week's administration with RPO reduced fasting blood glucose, decreased glycosylated hemoglobin (GHb), and improved the glucose tolerance in diabetic mice. In diabetic rats complicated with hyperlipemia, RPO prevented and reduced both hyperglycemia and hypertriglyceridemia. The results support the view that RPO may influence glucose or carbohydrate metabolism of diabetic animals in many ways including inhibiting the activity of alpha-glucosidase in digestive canal, and improving the metabolism of glucose and triglyceride.

Differential effects of enrichment on learning and memory function in NR2B transgenic mice.

It has been known that environmental enrichment leads to better learning and memory in mice. However, the molecular mechanisms are not known. In this study, we used the 10th-12th of the NR2B transgenic (Tg) lines, in which the NMDA receptor function is enhanced via the NR2B subunit transgene in neurons of the forebrain, to test the hypothesis of the involvement of NMDA receptor function in enrichment-induced better learning and memory. Consistent with our previous results, both larger long-term potentiation (LTP) in the hippocampus and superior learning and memory were observed in naive NR2B Tg mice even after the 10th-12th generation of breeding. After enrichment, wild-type mice exhibited overall improvement in their performances in contextual and cued conditioning, fear extinctions, and novel object recognition tasks. Interestingly, the same enrichment procedures could not further increase the performance of NR2B Tg mice in contextual conditioning, cued conditioning, or fear extinction, thereby indicating that enhanced NMDA receptor function can occlude these enrichment effects. However, we found that in the novel object recognition task enriched NR2B Tg mice exhibited much longer recognition memory (up to 1 week), compared to that (up to 3 days) in naive NR2B Tg mice. Furthermore, our biochemical experiments showed that enrichment significantly increased protein levels of GluR1, NR2B, and NR2A subunits of glutamate receptors in both wild-type and NR2B Tg mice. Therefore, our results suggest an interactive nature of molecular pathways involved in both environmental and genetic NMDA receptor manipulations for enhancing learning and memory.

Watermarking medical signals for telemedicine.

Modern telecommunication infrastructure supports the possibility of delivering quality health care without the physical presence of medical experts. The integrity of biomedical signals being transmitted through communication channels must be established before their utilization. This paper investigates three digital watermarking techniques for signal integrity verification in an electroencephalogram (EEG) monitoring application for brain injury detection. The techniques studied are patchwork, least significant bit, and quantization watermarking methods. The three techniques are evaluated and compared in the following areas: sensitivity to noise contamination, robustness to EEG signal characteristic changes due to brain injury, and consistency under various communication channel models. The patchwork method performs best for noise contamination rejection among the three methods. The noise contamination detection rates of all three methods remain relatively stable across a wide range of EEG characteristics.

Cell-free entry of human T-cell leukemia virus type 1 to mouse cells.

Human T-cell leukemia virus type 1 (HTLV-1) is the etiologic agent for adult T-cell leukemia and HTLV-1-associated myelopathy / tropical spastic paraparesis. Recently we infected newborn mice by inoculating HTLV-1-producing human cells, and found that T-cells, B-cells and granulocytes were infected in vivo. To understand the mechanism of viral-cell interaction and the pathogenesis of HTLV-1 using the mouse model, it is important to clarify the cellular tropism using a cell-free HTLV-1 transmission system. We employed a highly transmissible cell-free HTLV-1 produced by a feline kidney cell line, c77, and studied the susceptibility of 9 kinds of mouse cell lines, EL4, RLm1, CTLL-2, J774.1, DA-1, Ba / F3, WEHI-3, NIH3T3 and B1, and two kinds of human cell lines, Molt-4 and Hut78. HTLV-1 proviral sequence was found by PCR in all 9 mouse cell lines as well as in 2 human cell lines and viral entry was blocked with sera from an HTLV-1 carrier and an adult T-cell leukemia patient. Unexpectedly, mouse cell lines EL4 and RLm1 and human cell lines Molt-4 and Hut78 showed similar efficiency for viral entry. These results suggest a wide distribution of HTLV-1 receptor in mouse cells.

[No evidence for association between the alpha 2-macroglobulin polymorphism and Alzheimer's disease in the Han Chinese].

OBJECTIVE: To determine the effect of the pentanucleotide deletion polymorphism adjacent to a consensus splice site in intron 17 of alpha 2-macroglobulin(A2M) gene on the development of late-onset Alzheimer's disease (LOAD) in the Han Chinese in Guangzhou area. METHODS: A2M and apolipoprotein E genotypes were examined by polymerase chain reaction(PCR) and restriction fragment length polymorphism (RFLP) techniques in the patients(32 men and 65 women) with LOAD and age-matched healthy elderly Chinese subjects (57 men and 54 women). RESULTS: The A2M gene deletion allele frequency was 2.6% in the cases with LOAD and 2.7% in the controls. In the LOAD cases, the frequency of apoE-epsilon4 increased significantly (Z=3.32, P<0.01). ApoE-epsilon4 was associated with LOAD (RR=2.67, chi(2)=10.71, P<0.01) while the A2M deletion polymorphism was not associated with LOAD even the samples were stratified according to the presence of the apoE-epsilon4 allele. CONCLUSION: The above data demonstrate that there is no evidence for the association of A2M polymorphism with the development of LOAD in the Han Chinese population in Gaungzhou.

[The millipore rapid screening of hydantoinase production strains].

OBJECTIVE: To find a strain with a higher hydantoinase activity. METHODS: The millipore rapid screening method was established to select the strains. The effects of several solvents on the rate of biotransformation were compared. RESULTS: A hydantoinase production strain has been selected by the millipore rapid screening method. Tween-80 (1%) is found to be a suitable solvent to dissolve the substrate. CONCLUSION: A new method is provided for rapid selection of hydantoinase production strains from large-scale samples. The difficulty in dissolving the substrate has been resolved to some extent.

[Autologous bone marrow transplantation for patients with chronic myelogenous leukemia after in vitro purging of the graft with bcr/abl antisense oligodeoxynucleotides].

OBJECTIVE: To investigate the efficacy of autologous bone marrow transplantation (ABMT) for patients with chronic myelogenous leukemia (CML) after in vitro purging of the graft with bcr/abl antisense oligodeoxynucleotides (AS-ODN). METHODS: Five CML patients, 2 in chronic phase (CP), 1 in accelerated phase (AP) and 2 in blast crisis (BC), all confirmed the presence of b3a2 bcr/abl mRNA by RT-PCR (reverse transcriptase-polymerase chain reaction). Patients had received 2 courses of intensive chemotherapy for "in vivo purging" before bone marrow harvesting. The autologous bone marrow cells were concentrated by CS3000plus and then incubated with 18-mer phosphorothioate bcr/abl AS-ODN (40 - 60 microg/ml, for 48 - 60 h). Conditioning regimens were TBI + Cy (total body irradiation + cyclophosphamide) or MAC + CCNU. RESULTS: After intensive chemotherapy, ph(+) cells in bone marrow reduced to 34% (24% - 46%) and the numbers of bcr/abl mRNA (+) CFU-GM to 45.6% (33% - 58%). After bcr/abl AS-ODN in vitro purging, two patients were bcr/abl mRNA negative and three positive with the number of bcr/abl mRNA (+) CFU-GM markedly reduced. Delayed hematopoietic reconstitutions were observed in all patients after ABMT. During a more than 2 years follow-up period, three patients obtained major cytogenetic remission (MCR) and lasted for 9 - 12 months and the durations of CP after ABMT were longer than that before ABMT. One of this three in BC before ABMT being survived disease-freely for 37 months with bcr/abl mRNA (-) after ABMT. Another patient in BC had a short duration of MCR and relapsed at +7 months. One patient died from severe infection and bleeding at day +74 because of delayed reconstitution. CONCLUSION: ABMT with bcr/abl AS-ODN purged graft can result in quite long duration of MCR and prolonged CP in some patients with CML.

[Genetic diversity of rhizobia of Medicago edgeworthii by AFLP and RFLP analysis].

Two hundred and ninety one isolates of rhizobia sampled from root nodules of Medicago edgeworthii which came from Yun Nan Province in china were analyzed by amplified restriction fragment polymorphism(AFLP) technique. According to the AFLP banding patterns, the results showed most of these isolates were genetically diversity. Ninety isolates were selected as representing the diversity among the 291 isolates. The 90 isolates were clustered into three groups at the level of similarity of 79% by computer analysis of the data. Additional representative isolates and reference strains were identified by restriction fragment length polymorphism (RFLP) analysis of PCR-Amplified 16S rDNA. Two different 16S rDNA PCR-RFLP types were found, which indicated that the isoltes were phylogentically closely related to Rhizobium mongolense.

[Experimental studies on antioxidation of extracts from several plants used as both medicines and foods in vitro].

OBJECTIVE: The scavenging effects of 8 extracts from several plants used as both medicines and foods on oxygen free radicals were studied in vitro. METHODS: Chemiluminescence (CL) method and ultra-weak luminescence technique were used, meanwhile, macrophage respiratiory burst was also carried out to evaluate the antioxidation reaction. RESULTS: The scavenging activity on superoxide anions (O2-.) differed in the following order: extract of clove (CE) > black tea (BTE) > green tea(GTE) > grape peel/seed (GE) > or = oologn tea (OTE) > > tumeric (TE). In Fenton reaction for hydroxyl radicals (.OH), the order as follows: BTE > CE > or = GTE > GE > OTE > > TE. Most extracts have stronger eliminating efficiency on .OH than on O2-. except CE, which may scavenge the O2-. formation more efficiently. After macrophages (5 x 10(6) cells/ml) were stimulated with phorbol myristate acetate, BTE showed stronger inhibiting action than CE on respiratory burst in macrophages. The results of ultra-weak luminescence measurement indicated that the extracts could delay and depress the DNA damage in CuSO4-Phen-Vc-H2O2-DNA chemiluminescence system. CONCLUSIONS: It suggests that the tested antioxidants could decrase the formation of O2-. or .OH, some have chain-breaking function and preventative function during DNA damage.

Human T-cell leukemia virus type 1 can infect a wide variety of cells in mice.

Analysis of human T-cell leukemia virus type 1 (HTLV-1)-infected cell types and the interplay of these infected cells in vivo should provide valuable information to elucidate the pathogenesis of HTLV-1-associated diseases in humans and in animal models. In this study, HTLV-1-infected cell types were identified in HTLV-1-infected C3H/HeJ mice. Pan T, CD4+, CD8+, granulocyte and pan B cell fractions in the splenocytes of MT-2 cell-inoculated mice were sorted by use of their cell surface high-density expression of CD3e, CD4, CD8, Gr-1 and B220 antigens, respectively, with a fluorescence-activated cell sorter. The pX sequence of HTLV-1 provirus in the lysate of each fraction was amplified by polymerase chain reaction and detected by Southern hybridization. Interestingly, in addition to the CD4+ cell fraction, the pX sequence was also found in CD8+ cell, B cell and granulocyte fractions. The broad cell spectrum of HTLV-1 infection in mice is consistent with the situation in humans. Our finding indicate that HTLV-1 receptor or coreceptor is widely distributed among different cell types in mice.

[Simultaneous observation of parameter of intraventricular pressure and energy metabolism in rat hearts using nuclear magnetic resonance].

A technique for simultaneous observation of parameters of intraventricular pressure and energy metabolic status in isovolumic rat hearts was described. The parameters of intraventricular pressure were collected and processed using a computerized monitoring system for heart function, and myocardial energy metabolism was investigated by collecting the 31P NMR spectra. The preparation was stable metabolically and functionally for 2 h under the NMR experimental conditions.
