Distinguishable transformation-defective phenotypes among temperature-sensitive mutants of Rous sarcoma virus.

Eight transformation-defective, temperature-sensitive (ts) mutants of the Prague strain of Rous sarcoma virus, subgroup A, have been isolated after mutagenesis with 5-bromodeoxyuridine followed by selection on the basis of focus tests. Five of these mutants, ts GI201, GI202, GI203, GI204, and GI205, exhibit properties like most previously reported isolates in that they show a temperature-sensitive response to each of a variety of transformation-specific parameters tested. Interestingly, GI201, in addition to the temperature-sensitive defect, carries a lesion that was observed as a nonconditional loss of expression of plasminogen activator protease. Three mutants, ts GI251, GI252, and GI253 have been disignated partial transformation-defective (PTD) mutants since they behave as ts mutants according to some tests for transformation and as wild type according to others. These three mutants fail to form foci at the nonpermissive temperature (41 degrees C) and art nontumorigenic in 3-week-old chickens (body temperature, 42 degrees C). The agglutinability by concanavalin A of cells infected with these mutants shows a definite temperature sensitivity, as do the rate of 2-deoxyglucose uptake and the disappearance of the 250, 000-dalton normal cell glycoprotein (large, external, transformation sensitive [LETS]). Although the PTD mutant-infected cells, unlike cells infected with other transformation mutants, exhibit a cell-bound plasminogen activator protease at the nonpermissive temperature, this activator is not detectable as a free protease in the medium, as it is with wild-type, virus-infected cells. The PTD mutants behave like the wild-type parent in their ability to induce transformed growth properties in the infected cells, i.e., growth beyond normal cell saturation density with or without serum-supplemented medium and growth leading to colony formation in soft-agar- or methyl cellulose-containing suspension media.

Life dissatisfactions, job dissatisfaction and illness of married men over time.

This study examines the degree to which job and marital satisfaction and participation in leisure activities could predict concurrent and future health status relative to ethnicity, class and occupational factors, and mental health. Data were obtained from a longitudinal study of a random sample of 1034 Manhattan families with at least one child 6 to 18 years of age. The wives in these families were interviewed twice at 5-year intervals. Multiple regression analyses, using variables abstracted from the records of 640 wives of working husbands, indicated that husbands' health status at Time I (R = .33) and Time II, 5 years later (R = .28) was significantly predicted by the Time I assessments on 12 independent variables. The largest single unique contributor, i.e. net of all other independent variables, to concurrent husband's health was job dissatisfaction (partial r = .15), followed by husband's institutionalization for mental illness (r = .14), being Spanish (r = .10), and the factor of Unhappy Marriage (r = .09). This preliminary investigation indicated that uncertain life satisfactions were important predictors for either concurrent or future health status which operated across class, ethnic, occupational and mental health factors. These findings tend to support models which postulate stressful psychosocial conditions as potential etiologic agents in the development of illness.

Further evidence for the existence of a viral envelope protein defect in the Bryan high-titer strain of Rous sarcoma virus.

Electron microscopy observations of purified Bryan high-titer Rous sarcoma virus (BH RSV) using the freeze-drying technique showed that progeny made in the absence of a helper virus lacked visible surface projections or spikes. Phenotypic mixing experiments employing BH RSV and a thermolabile mutant of vesicular stomatitis virus, tl 17, yielded no evidence of pseudotype formation. Since tl 17 is known to be defective for an envelope glycoprotein, the lack of successful phenotypic mixing with BH RSV is consistent with the observed absence of viral spikes.

Patients' preferences for resident physician dress style.

A survey study was conducted to determine patients' preferences for resident physician attire as well as the relationship of physicians' attire to patient satisfaction and perception of the quality of care. Questionnaire responses were obtained from 200 consecutive subjects equally divided among four clinical settings (a hospital-based internal medicine clinic, an emergency room, an internal medicine ward, and a community-based internal medicine clinic) which are maintained by the University of California, Irvine, Medical Center. The questionnaires were administered only to English-speaking patients who had seen a physician on a previous visit. A majority of the patients at the four settings indicated that they had no preference regarding attire (70.5% for male doctors, 66.5% for female doctors): however, 44% of the patients said that neatness of dress was moderately to very important. The aggregate findings were not greatly altered when adjusted for demographic variables such as age, sex, race, occupational status, and education.
