Imbalanced deoxyribonucleoside triphosphate pools and spontaneous mutation rates determined during dCMP deaminase-defective bacteriophage T4 infections.

DNA precursor imbalances are known to be mutagenic in both eukaryotic and prokaryotic systems. Almost certainly, such mutagenesis involves competition between correctly and incorrectly base-paired precursors at replication sites. Since other factors may be involved, it is important to identify specific mutations induced by specific pool imbalances. Using bacteriophage T4, we have developed a system for such analysis. We prepare double mutants of T4; one mutation affects a phage-coded enzyme of deoxyribonucleoside triphosphate (dNTP) metabolism, while the second is an rII mutation known to revert along a specific pathway. We determine dNTP pools in infection by such a mutant and measure both the spontaneous reversion rate of the rII mutation and, in some cases, the nucleotide sequence at the mutant site. In this paper we analyze mutations induced by a deficiency of T4-encoded deoxycytidylate deaminase. This causes pools of 5-hydroxymethyl-dCTP to expand some 30-fold, while dTTP pools contract. This specifically stimulates AT-to-GC reversion. One of the four AT-to-GC reverters tested, rIIUV215, increases its reversion rate at least 1000-fold under these pool-imbalance conditions, while the other mutants tested show increases of only about 10-fold. Therefore, factors other than dNTP competition, including local DNA sequence environment, must be invoked to fully explain mechanisms of dNTP pool imbalance-induced mutagenesis. We discuss models for this, and we also report unexpected effects of the dCMP deaminase deficiency upon pools of ribonucleoside triphosphates.

Parasitic infection among residents of an institution for mentally retarded persons.

Data on occurrence of intestinal parasites among resident and entering populations in a South Carolina institution for mentally retarded persons were examined. Following an eradication effort in 1973 for Trichuris trichiura, the prevalence of this parasite remained below 5 percent for the resident population, with the exception of 1974 and 1975, when small numbers of stool examinations occurred. Significantly higher proportions of the entering population, compared to the residents, harbored T. trichiura, Ascaris lumbricoides, Giardia lamblia, and Entamoeba histolytica. We concluded that annual examination and treatment can control trichuriasis; however, complete eradication may require examination following home visits by the resident population.

Changes in intraocular pressure during acute exercise.

The effect of vigorous physical activity on intraocular pressure has been well documented. Other studies have only focused on pretest and posttest measurements of intraocular pressure. This study presents a continuous evaluation of intraocular pressure during exercise and correlates changes in intraocular pressure with changes in heart rate through time. A total of 17 adult male and female subjects were submaximally tested on a bicycle ergometer using the Astrand-Rhyming protocol. The results of this test were used to design an individualized four-stage workload that would proportionally take each subject to 85% of a predicted maximum heart rate. During this test, intraocular pressure was measured at the end of each stage and through 15 min of recovery with an American Optical non-contact tonometer. A statistically significant decrease (p less than 0.05) in intraocular pressure was noted at the end of the fourth stage and during 1, 5, and 10 min of recovery; however, no trend was found to indicate a significant correlation between heart rate and intraocular pressure.

Physical fitness and intraocular pressure.

The ability of moderate to intense aerobic exercise to reduce intraocular pressure (IOP) has been reported. Previous studies have focused on a single postexercise recovery period not considering the long-term effect of improved physical fitness on IOP. A total of 32 adult subjects, all having IOP's of greater than 18 mmm Hg in each eye as determined by an American Optical Corporation Non-Contact Tonometer, were selected for the study. After a complete medical and visual evaluation, participants were randomly assigned to either an intervention group (N = 18) receiving a 6-month supervised exercise program to improve physical fitness or into a control group (N = 14). After 6 months, each group was reevaluated to observe changes in physical fitness and IOP. Pre- and poststudy measurements of maximum oxygen uptake (ml/kg/min) made with a Beckman O2 analyzer showed significant improvement in physical fitness in the intervention group (p less than 0.05). There was a significant decrease (p less than 0.05) in IOP, 2.0 mmm Hg in the right eye and 1.3 mm Hg in the left eye, in the intervention group; however, decreases in IOP were comparable and significant (p less than 0.05) in the control group. Results from multiple regression analysis for each individual group and for combined groups indicated that changes in IOP were not dependent upon changes in physical fitness.

Suppressors of mutations in the bacteriophage T4 gene coding for both RNA ligase and tail fiber attachment activities.

The protein product of T4 gene 63 catalyzes both the attachment of tail fibers to fiberless phage particles and the ligation of single-stranded RNA (Snopek at al., Proc. Natl. Acad. Sci. U.S.A. 74:3355-3359, 1977). To investigate whether the gene 63 product has a role in nucleotide metabolism, we isolated false revertants of amM69 in gene 63. We screened for revertants that could grow at 30 degrees C but not at 43 degrees C on Escherichia coli OK305 when nucleotides were limiting. These false revertants contained the original mutation in gene 63 and new suppressor mutations. Some of these suppressor mutations caused temperature sensitivity by themselves, allowing single mutants carrying the suppressor to be recognized and isolated. The results of mapping and complementation studies indicated that most of these ts suppressors were in the t gene (lysis), one was in gene 5 (baseplate), and one was in gene 18 (sheath). The mutation in gene 18, tsDH638, suppressed three different amber mutations in gene 63 but did not suppress amber mutations in several other genes. None of the suppressors that were characterized were in genes with known functions in nucleotide metabolism. However, an intriguing property of these false revertants was that they were very sensitive to hydroxyurea, an inhibitor of nucleotide metabolism.

A clinical evaluation of the efficacy of mebendazole in the treatment of trichuriasis.

In a single-blind clinical study of mebendazole, a new, broadspectrum anthelmintic, 70 patients with light to severe infections of Trichuris trichiura were treated. Ten subjects received placebos, and 60 received 100 mg mebendazole twice a day for three and four days. The drug was well tolerated by the patients, and no toxic side effects were observed. Egg count comparisons using the McMaster method on feces collected before and one month after treatment indicated a cure rate for trichuriasis of 65.6% and 89.3% and a total egg reduction of 69.5% and 95.9% for three- and four-day regimens, respectively. Patients not cleared showed an egg reduction rate of 31.6% and 76.8% on the three- and four-day regimens, respectively.

The relationship between knowledge of cardiovascular dietary risk and food shopping behaviors.

A 44-item knowledge and attitude questionnaire was administered to 176 randomly selected shoppers to determine their cardiovascular and nutrition knowledge and attitude toward the diet-heart relationship. An analysis of descriptive sales slips issued by the supermarket determined food purchasing behavior. Food items purchased were categorized according to their polyunsaturated-saturated (P/S) ratio less than 1 (PS0), P/S ratio greater than 1 (PS1), and high cholesterol (greater than 100 mg per serving) (HCHO). We also categorized purchases by cholesterol content per serving: high, moderate, low, and very low levels. We observed a significant (r = -.25, P = .0008) negative correlation between purchasing behavior of PS0 and knowledge. A partial correlation revealed a similar association when controlling for income (r = -.26, P = .0002), age (r = -.26, P = .0005) and education (r = -.22, P = .0038). Correlation for whites was significant and negative (r = -.29, P = .0003) and for blacks nonsignificant and positive (r = .02, P = .9069). Regression analysis found a significant negative slope (b = -.57, P = .0001) of PS0 on knowledge, a marginally significant positive slope (b = -.21, P = .0527) of PS1 on knowledge, and a significant negative slope (b = -.86, P = .0171) of HCHO on education. The interaction of race and knowledge was significant for PS0 (P = .0005).
