Uptake of indolmycin in gram-positive bacteria.

The antimicrobial activity of indolmycin correlates with the generation time of the investigated strains. Thus, in Staphylococcus aureus ATCC 13150 with a 37-min generation time, the minimal inhibitory concentration (MIC) was 0.6 microgram ml-1, and in Bacillus subtilis ATCC 27142 with a generation time of 23 min, the MIC reached 10.5 micrograms ml-1. Competition experiments in staphylococci and B. subtilis with aromatic amino acids demonstrated that indolmycin uses the uptake systems that are responsible for tryptophan. When the Ki values of indolmycin for the uptake of the aromatic amino acids in staphylococci were compared, there was a significantly higher influence on the uptake of tryptophan with respect to phenylalanine and tyrosine. In addition, indolmycin low resistant mutants of S. aureus ATCC 13150 showed a 10- to 100-fold decrease in Km value for the uptake of tryptophan and a 10-fold decrease for tyrosine uptake. The Km value for phenylalanine remained unchanged. A significant correlation existed between the Ki values of indolmycin for the uptake of tryptophan in the wild-type strains of S. aureus and B. subtilis and the MIC against the corresponding strain. Low Ki values corresponded to low MIC. These results imply that, in addition to improvement of the antibiotic structure for target affinity, the tryptophan uptake system can be used as a test model for the structural evaluation of indolmycin with respect to an increased transport activity into bacterial cells.

Enrichment of Auxotrophic Mutants in Streptomyces griseus.

A method for the isolation and enrichment of tryptophan auxotrophic mutants of the indolmycin-producing strain Streptomyces griseus ATCC 12648 was developed, using penicillin selection. With UV irradiation of 2.2 x 10 muW cm, a mutation rate to tryptophan auxotrophy of 5.6 x 10 was achieved. With 300 mug ml of penicillin G, an enrichment of tryptophan auxotrophs of about 1,000-fold was attained. Approximately 40% of all survivors were tryptophan auxotrophs.

Gunacin, a new quinone antibiotic from Ustilago sp.

In a screening program for antibiotics which were antagonized by cysteine, a strain, which was characterized as Ustilago sp., was found to produce a new quinone antibiotic, gunacin. The molecular weight M+ = 348.084 determined by mass spectroscopy, corresponds to a molecular formula of C17H16O8. Further spectroscopic data prove that gunacin is a new antibiotic. The antibiotic possesses a good inhibitory effect against mycoplasmas and Gram-positive bacteria including multi-resistant strains. It also possesses a weak activity against Gram-negative bacteria with the exception of Proteus vulgaris, which is more strongly inhibited. The main activity against fungi is found against Trichophyton mentagrophytes. Gunacin shows an inhibition of the DNA synthesis in vivo, is antagonized by mercapto compounds and possesses an acute toxicity of LD50 = 16 mg/kg i.p. and LD50 = 12 mg/kg i.v. in mice. Against HeLa-cell the antibiotic shows an ED50 = 12.11 microgram/ml. Thirty five microgram/ml of gunacin induces 1,063 interferon units.

Interaction of indolmycin in the metabolism of tryptophan in rat liver.

When compared with other tryptophan analogues, indolmycin is a potent inhibitor of tryptophan pyrrolase and tryptophan decarboxylase, both enzymes involved in tryptophan catabolism. Otherwise the decarboxylation of 5-hydroxytryptophan is only slightly affected by indolmycin. The indolmycin derivative, 2-methylamino-5-(1-naphthylmethyl)-oxazolidin-4-one (YX-CX 22 XX) demonstrates only a slightly weaker inhibitory effect in the tryptophan-tRNA-ligase system in E. coli but does not show any significant action on the tryptophan metabolism in the eukaryotic system.

Indolmycin inhibits prokaryotic tryptophanyl-tRNA ligase.

Indolmycin specifically prevents the formation of tryptophanyl-tRNA in a prokaryotic system in vitro using Escherichia coli enzymes. However, the drug has little effect in an eukaryotic system in vitro (rat liver enzymes). Analysis of the type of inhibition revealed that indolmycin competes with tryptophan as a pure competitive inhibitor of prokaryotic tryptophanyl-tRNA ligase.
