The glyoxalase system catalyses the conversion of methylglyoxal (and other 2-ketoaldehydes) to D-lactic acid via the intermediate S-D-lactoylglutathione. It comprises two enzymes, glyoxalases I and glyoxalases II, and catalytic amount to reduced glutathione. Methylglyoxal inhibits cell growth, while the glyoxalase system by breaking down methylglyoxal may act as a promoter of cell growth. Inhibitors of glyoxalases may serve as possible therapeutic agents against cancer by virtue of their ability to elevate the level of methylglyoxal in the body.
Dysfunction and/or injury of endothelial cells play the important role in atherogenesis. Vascular endothelium has many functions, for example regulation of tonus, coagulation and fibrinolysis. These functions endothelial cells are doing through the secretion of several substances, which are presented in this article.

The characteristics of the human neutrophil collagenase and intracellular and extracellular mechanism regulating this enzyme were given. There were presented molecular basis collagenase latent state , activation of latent enzyme form through the system dependent on free oxygen radicals and the role of glutathione peroxidase in the regulation of the activity of the human leukocyte collagenase.
In rabbits with experimental atherosclerosis induced by a cholesterol-rich diet, alpha 1-antitrypsin concentration was decreased as compared with control, by 34%, whereas alpha 2-macroglobulin concentration was increased by 86%. In animals fed a methionine-rich diet changes in concentration of both inhibitors involved in elastase metabolism were but slight, if any.

BACKGROUND: Water immersion stress (WIS) in rats causes decrease in deformability of red blood cells (RBC). We hypothesize that this phenomenon is mediated by reactive oxygen species. Since tirilazad mesylate (U74006F) is a potent scavenger of free oxygen radicals, the aim of this study was to investigate possible influence of this drug on WIS induced decrease in RBC deformability in rats. MATERIAL AND METHODS: The rats were injected i.p. with tirilazad mesylate in a dose of 10 mg per kg body weight or vehiculum at the beginning of the experiment and immersed in water at 23 degrees C to the depth of xyphoid for 5 h. Control rats were kept in normal environment. After 6 h blood samples were collected for examination. An elongation index of RBC measured by Rheodyn SSD (Myrenne GmbH, Germany) was used as a parameter of deformability. Lipid peroxidation products have been assayed in RBC by BIOXYTECH LPO-586 kit. RESULTS: Tirilazad mesylate prevents a decrease in RBC deformability, but does not prevent an increase in concentration of lipid peroxidation products in erythrocytes during WIS. CONCLUSION: Positive effect of tirilazad on stress induced decrease in RBC deformability is not related to inhibition of lipid peroxidation within the erythrocytes.
The study was aimed at verification of previously found, in animals with experimentally induced atherosclerosis, the disturbances of serum proteinases inhibitors: alpha-2-antiplasmin, alpha-1-antitrypsin and alpha-2-macroglobulin. In humans with hypercholesterolemia the decrease of serum activity of alpha-2-antiplasmin was observed. In humans with hypercholesterolemia and increased serum concentration of triacylglycerols no significant changes in activity of alpha-1-antitrypsin and alpha-2-macroglobulin were found--in comparison with control subjects.

Cytokines participate in the control of: proliferation and chemotaxis of various cells, thrombocyte adhesion and aggregation, vascular tonus, protein synthesis--among other acute phase protein synthesis, lipoprotein metabolism, and also exert influence on smooth myocyte phenotype transformation and conversion of fibroblasts to myocytes. All these mechanisms are important in the development of atherosclerotic lesions.
In 31 subjects with an occupational exposure to dust pesticides the levels of PZ-peptidase, alpha-1-antitrypsin, alpha-2-macroglobulin were determined. A decrease of PZ-peptidase and an increase of alpha-1-antitrypsin, alpha-2-macroglobulin were found in comparison with a control group of 30 healthy volunteers.
