C1-esterase inhibitor prevents early pulmonary dysfunction after lung transplantation in the dog.

The success of lung transplantation to a large extent depends on effective protection of the graft from ischemic injury after reperfusion. Although mechanisms have not been clarified, the pathologic findings of ischemic injury after reperfusion are similar to adult respiratory distress syndrome, a condition in which the blood coagulation contact system is activated. This study evaluates the effect of C1-esterase inhibitor (C1-INH), the main inhibitor of the blood coagulation contact system, on short-term lung function in a dog model of orthotopic lung transplantation. Twelve lung transplantations were performed after 24 h of ischemic time. Dogs were randomly assigned to receive either vehicle (Control) or C1-INH. After the lung transplantation in the control group, Pao2 decreased by 84% and both the AaPO2 and the Qs/Qt% increased (340 and 530%, respectively, p < 0.01); these parameters remained unchanged in the C1-INH group. The hypoxemia observed in control animals was associated with decreased blood coagulation contact factors, complement consumption, increased expression of adhesion glycoproteins in leukocytes, and extensive intraalveolar and interstitial neutrophil infiltration. In contrast, C1-INH administration prevented hypoxemia, the decrease in blood coagulation contact factors, the activation of the complement system, the increase in expression of leukocyte adhesion molecules, and inflammatory cell infiltrate. This study has demonstrated that in a dog model of lung transplantation, the administration of C1-INH prevents early pulmonary dysfunction, and it suggests that activation of blood coagulation contact system and complement are important mechanisms causing ischemic injury after reperfusion.

New biochemical markers of bone resorption derived from collagen breakdown in the study of postmenopausal osteoporosis.

The aim of this work was to perform a comparative study between three recently developed biochemical markers of bone resorption derived from collagen metabolism--(1) total urinary free pyridinolines (Pyr), (2) serum pyridinoline cross-linked carboxy-terminal telopeptides of type I collagen (ICTP) and (3) a urinary-specific sequence for a part of the C-telopeptide of the alpha 1 chain of type I collagen (CTX)--in the diagnosis and follow-up of postmenopausal osteoporosis. Results were also evaluated relative to the classical biochemical marker urinary hydroxyproline (Hyp). The study included 20 untreated osteoporotic postmenopausal women (OSP), age 60 +/- 6 years, range 46-69 years; 27 osteoporotic postmenopausal women treated (OSP-T) by cyclic therapy with disodium etidronate, 25-hydroxyvitamin D and calcium for a period between 3 months and 4 years (25 +/- 15 months), age 59 +/- 7 years, range 41-67 years; 17 osteopenic postmenopausal women, age 57 +/- 6 years, range 46 +/- 68 years; and 29 healthy control postmenopausal women, age 56 +/- 7 years, range 41-70 years. The diagnostic criterion for postmenopausal osteoporosis was a bone mineral density (BMD) (Hologic QDR-1000) in lumbar spine and/or femoral neck more than 2 SD below the mean value corresponding to an age- and sex-matched healthy control group. For inclusion in the osteopenic group BMD values had to be between 1 and 2 SD below the mean BMD value corresponding to the control group. We found a significant increase (p < 0.01) in the levels of Pyr/Cr and CTX/Cr (Cr = creatinine) in OSP patients with respect to the control group and we did not observe any significant difference between control and OSP-T or osteopenic women. It is interesting to note that there was a mean increase in CTX/Cr in OSP patients of 101% of the control values, while the mean increase found in Pyr/Cr concentration was only 33%. However, we did not find significant differences in the concentrations of ICTP and Hyp/Cr between groups. In a comparison of Pyr/Cr and CTX/Cr, urinary CTX showed the higher diagnostic accuracy, as can be deduced from the receiver operating characteristic (ROC) curves. CTX sensitivity was 40% with a specificity of 100%, whereas the sensitivity was 25% for urinary Pyr/Cr. In conclusion, the results of the present work suggest that in osteoporotic women CTX has the highest diagnostic accuracy among the markers of bone resporption studied.

High degree of similarity between Chromatium vinosum and Chromatium minutissimum as revealed by riboprinting.

The riboprinting technique (restriction fragment length polymorphism [RFLP] analysis of PCR-amplified ribosomal DNA) was used to study five strains representing three species of the genus Chromatium. An RFLP analysis following digestion of the amplified small-subunit ribosomal DNA with 25 restriction enzymes revealed that the patterns obtained for all strains of Chromatium vinosum were identical. Chromatium gracile was different from C. vinosum with seven enzymes. On the other hand, Chromatium minutissimum produced the same patterns as C. vinosum with all enzymes, indicating that these organisms have a high degree of similarity. An RFLP analysis of the PCR-amplified spacer sequence between the 16S and 23S ribosomal DNAs gave similar results except that there was a larger number of differences between C. gracile and the other organisms examined.

Cellular content of storage inclusions in purple sulfur bacteria determined by ultrathin sections.

Phototrophic sulfur bacteria accumulate storage inclusions as a mechanism to adapt to several types of environmental stress. We compare the content of elemental sulfur and poly-beta-hydroxybutyrate (PHB) found in cultures growing under laboratory conditions with the content found in microorganisms in natural environments. Since natural communities are extremely heterogeneous in composition (they do not contain only phototrophic sulfur bacteria) and physiological state, it was not possible to apply conventional chemical analyses for the determination of the contents of sulfur and PHB. The study was performed by means of transmission electron microscopy, which turned out to be an excellent tool for this purpose. The results indicate that, in natural environments, cells have an extremely high content of storage inclusions, much higher than their laboratory grown counterparts, probably as a consequence of less favorable environmental conditions.

3-Hydroxybutyric Acid

Oswaldocruzia venezuelensis sp. n. (Nematoda:Trichostrongylina, Molineoidea), a parasite of Bufo marinus from Venezuela.

A new species of Oswaldocruzia Travassos, 1917, a parasite of Bufo marinus L. from Venezuela, is described. Like most Neotropical Oswaldocruzia, Oswaldocruzia venezuelensis sp. n. is characterized by spicules with three principal branches: blade, shoe and fork, and by a division of the fork within the distal third of the spicule length. O. vaucheri Ben Slimane et Durette-Desset, 1993 is the most closely related species due to its caudal bursa of type II and its cervical alae of the same shape but it differs in the following characters: the position of the papillae of rays 4 situated nearer the papillae of rays 3 rather than rays 5, a higher percentage of the ridges in the oesophageal region, the cervical alae three times longer and sharp and the spicular fork divided less deeply.

Genomic heterogeneity in Chlorobium limicola: chromosomic and plasmidic differences among strains.

Chromosome analysis by pulsed-field gel electrophoresis shows a high level of genetic heterogeneity between the two subspecies of the green sulfur bacteria Chlorobium limicola analysed: C. limicola and C. limicola f. s. thiosulfatophilum. Currently, they are differentiated only by the ability to utilize thiosulfate as photosynthetic electron donor, and by their %G + C content (51% and 58.1%, respectively). However, the capacity to utilize thiosulfate as photosynthetic electron donor does not appear to be a useful criterion to differentiate between some strains of this species, because this ability is encoded by plasmids that are different depending on the thiosulfatophilum strain analysed. In contrast, this study reveals that the comparison of chromosomal restriction patterns is very useful as an additional aid for the differentiation and identification of C. limicola strains.

Induced mutagenesis by bleomycin in the purple sulfur bacterium Thiocapsa roseopersicina.

Cell death and mutagenesis in bleomycin-treated cells of Thiocapsa roseopersicina (a purple sulfur bacterium) was studied by cultivation in a semisolid medium (agar-shake technique). This technique has also proven useful in assessing the frequency of antibiotic mutations by detecting and counting individual colonies of Thiocapsa roseopersicina. The frequencies of spontaneous mutants resistant to ampicillin, rifampicin, cloramphenicol, tetracycline, kanamycin, streptomycin, and neomycin were also studied: they ranged between 2 x 10(-9) and 9 x 10(-8). Bleomycin (4 micrograms/ml) sharply increased the frequency of ampicillin-resistant mutants, from 10(-8) (spontaneous) to 4 x 10(-4) (induced), in 17 h. An inducible, error-prone mechanism of DNA synthesis seems to be responsible for this enhancement of the mutagenic effect. this is the first report on the sensitivity to several antibiotics, and capacity of lethality and mutagenesis by bleomycin has been studied in a purple sulfur bacterium.

Ampicillin Resistance

Differential characteristics of AIDS patients with a history of imprisonment.

BACKGROUND: AIDS is among the leading causes of death in prisons, but there is little information about AIDS patients with a history of imprisonment. METHODS: AIDS patients diagnosed in Barcelona between 1988 and 1993 were studied. Those with prison histories were compared to those without, with respect to epidemiological variables, including survival analysis. RESULTS: 28.5% of 2336 AIDS patients, 49.4% of intravenous drug users (IVDU) and only 2.6% of homosexuals who were not IVDU had a prison history. Those with prison histories, compared to those without, were younger (median age of 30.6 versus 36.4, P < 0.0001), more often IVDU than homosexuals (87.8% versus 35.8%, OR = 36.9, 95% CI: 22.6-60.8, P < 0.0001), and diagnosed with AIDS because of extrapulmonary tuberculosis (32.0% versus 14.7%, P < 0.001). Among IVDU, those with prison histories were more frequently males (OR: 2.2; 95% CI: 1.6-2.9), lived in the poorest district of Barcelona more frequently than in the richest district (OR: 6.6; 95% CI: 3.4-12.9) and presented with extra-pulmonary tuberculosis more frequently than Pneumocystis carinii pneumonia (OR: 1.7; 95% CI: 1.2-2.4). Longer survival in the prison group did not persist when adjusted for age and AIDS-defining disease. Those with prison histories who presented with AIDS with only extrapulmonary tuberculosis had better probability of survival than those who presented only with P. carinii pneumonia (P < 0.001). CONCLUSIONS: AIDS patients in Barcelona with prison histories tended to be younger, more likely to be IVDU, and to present with extrapulmonary tuberculosis as an AIDS-defining illness than other patient groups. Better survival appears to be related to age and AIDS-defining illnesses in the prison group. The fact that half the IVDU AIDS cases had prison histories has important implications for the care and prevention of HIV, tuberculosis, and drug abuse in comparable prison settings.

Analysis of bacterial genomes by pulsed field gel electrophoresis.

The present article reviews the main achievements reached in bacterial genetics through the application of pulsed field gel electrophoresis. In particular, it summarizes the main subjects where the use of this technique has significant relevance for the analysis of bacterial genomes. With this purpose, it sums up some of the most relevant examples which illustrate the applicability of this technique in bacterial taxonomy, in epidemiological work and in the study of bacterial genome organization (including the assessment of the number of genomic elements and the size and topology of all of them). This technique, which makes it possible to make genomic comparisons in an easy, fast and repetitive way, has been demonstrated to be especially useful for those bacteria where the application of classical genetic techniques has not been possible.

Susceptibility of various purple and green sulfur bacteria to different antimicrobial agents.

Several purple and green sulfur bacteria (genera Chromatium, Thiocapsa and Chlorobium) were tested for their sensitivity to different antimicrobial agents by a disc diffusion assay, using thioacetamide as a source of hydrogen sulfide for plate growth. Chlorobium limicola strains were more sensitive to amoxicillin, erythromycin and nalidixic acid, whereas gentamicin and netilmicin were more active against the purple bacteria tested. None of the organisms were sensitive to oxacillin and trimethoprim+sulfamethoxazole. The critical concentrations at the edge of the inhibition zone were also calculated for three organisms and the antimicrobials colistin, mitomycin C, penicillin G, rifampicin, and streptomycin. The results obtained suggest that colistin, mitomycin C, penicillin G would provide selective conditions against the growth of Chlorobium limicola strains, while streptomycin and other aminoglycoside antibiotics would select against purple bacteria.

Transformation of Chlorobium limicola by a plasmid that confers the ability to utilize thiosulfate.

A modified transformation method for Chlorobium limicola confirms the role of an endogenous plasmid in thiosulfate metabolism. The plasmid was present in two forma specialis thiosulfatophilum (Tio+) strains and absent in one non-thiosulfate-utilizing (Tio-) strain. The plasmid (size, 14 kb) was transferred by transformation from Tio- to Tio+. The chromosomal restriction patterns, analyzed by pulsed-field gel electrophoresis, were used to distinguish between the different strains.

Hypersensitivity of Rhodobacter sphaeroides ribosomes to protein synthesis inhibitors: structural and functional implications.

The elongation cycle of protein synthesis systems of purple nonsulfur photosynthetic bacteria Rhodobacter sphaeroides, grown both phototrophically and chemotrophically, was studied using 33 inhibitors with different chemical structures and functional and domain specificities. No functional differences between phototrophic and chemotrophic ribosomal systems were detected. Rhodobacter sphaeroides ribosomes exhibited strong hypersensitivity to nine functional inhibitors when compared with Escherichia coli ribosomes. Most of the R. sphaeroides ribosomal hypersensitivities corresponded to peptidyltransferase inhibitors, implying that this important functional neighborhood must be somehow different in the two organisms.
