Cytochrome oxidase molecules were incorporated into black lipid membranes and into a new form of planar synthetic membrane. Studies of these membranes indicated that the incorporation of large membrane bound enzymes into black lipid membranes involves difficulties fundamental to this technique. On the other hand the new method described in this paper is more promising.
The single channel conductivity of the gramicidin channel has been measured for all the alkali ions using both H2O and 2H2O as a medium. Significant changes in conductivity with medium have been observed in all cases except lithium.
A simple model for the gramicidin ion pore is analyzed in which the potential energy of the ion in the pore can be resolved into three parts: a series of equal potential barriers, the contribution from the applied potential, and a parabolic variation leading to either a potential maximum or a potential minimum at the center of the membrane. For low ion concentrations, the particular case corresponding to a potential minima 2kT below the surface potential predicts experimentally observed behavior.
The synthesis of two new variants of gramicidin is described. It is shown that the changes in the aromatic side groups do not influence the single channel conductivity. Experiments in which solutions having different molarities on the two sides of the bilayer lipid membrane are described and their results compared with a rate theory analysis. It is concluded that the gramicidin pore contains approximately 10 equal potential maxima.
