Characterization of an anion-permeable channel from sugar beet vacuoles: effect of inhibitors.

The vacuole occupies 25-95% of the plant cell volume and plays an essential role in maintaining cytoplasmic homeostasis of nutrients and ions. Recent patch-clamp studies identified ion channels and electrogenic pumps as pathways for the movement of ions and metabolites across the vacuolar membrane (tonoplast). At high cytoplasmic Ca (>10 M) and negative potentials (inside the vacuole) non-selective channels of the ;slow-vacuolar (SV)-type' were activated resulting in anion release or cation influx. In the present study these vacuolar channels were characterized pharmacologically by ion channel inhibitors. The cation-transport inhibitors Ba, TEA and amiloride caused only partial and reversible block of the ;SV-type'channels, whereas anion-transport inhibitors strongly affected the vacuolar channels. Pyridoxalphosphate and the dimethylaminecarboxylate derivates anthracene-9-carboxylic acid and C 144 reversibly blocked the channels up to 70% and Zncl(2) up to 95%. DIDS and SITS inhibited this channel irreversibly up to 95%. The block developed under a variety of experimental conditions using solutions containing combinations of permanent cations and anions. The DIDS binding site is located on the cytoplasmic surface of the tonoplast, as intravacuolar DIDS did not block the channels. DIDS concentrations in the micromolar range, efficient in blocking 70-80% of the ;SV-type' channels did not significantly affect ATP-induced or pyrophosphate-induced proton-pumps. Stilbene derivatives may therefore be useful tools for studies on the substrate binding site on this vacuolar channel and for channel isolation.

Single channel recordings of reconstituted ion channel proteins: an improved technique.

Single channel recording of reconstituted ion channels is possible by patch clamp measurements of giant liposomes formed by dehydration-rehydration of lipid films. This "hydration technique" consists of carefully controlled dehydration of a suspension of small vesicles followed by rehydration of the residue resulting in formation of large liposomes. Patch pipettes can be attached to the liposome surface, yielding stable, high resistance seals between membranes and glass pipettes. This method allows the study of the properties of reconstituted ion channels from different tissues. The hydration technique was used to characterize the reconstituted K+-channel of sarcoplasmic reticulum from rabbit skeletal muscle. In a solution of 100 mM KCl, the sarcoplasmic reticulum K+-channel studied displays a conductance gamma K+ of 145 pS. The single channel conductance in 100 mM Rb+ and Na+ is gamma Rb+ = 98 pS and gamma Na+ = 65 pS respectively. A concentration of 0.5 mM decamethonium causes a flickering channel block. These properties are in good agreement with the ones found in sarcoplasmic reticulum K+-channels characterized by other methods. Other ion channels have also been reconstituted and studied by this technique. This improved method is compared with previous approaches and its applicability for the characterization of reconstituted ion channel proteins is discussed.

A role for fructose 2,6-bisphosphate in regulating carbohydrate metabolism in guard cells.

Fructose 2,6-bisphosphate (Fru2,6P(2)) appears to function as a regulator metabolite in glycolysis and gluconeogenesis in animal tissues, yeast, and the photosynthetic cells of leaves. We have investigated the role of Fru2,6P(2) in guard-cell protoplasts from Vicia faba L. and Pisum sativum L. (Argenteum mutant), and in epidermal strips purified by sonication from all cells except for the guard cells. Guard-cell protoplasts were separated into fractions enriched in cytosol and in chloroplasts by passing them through a nylon net, followed by silicone oil centrifugation. The cytosol contained a pyrophosphate: fructose 6-phosphate phosphotransferase (involved in glycolysis) which was strongly stimulated by Fru2,6P(2). A cytosolic fructose 1,6-bisphosphatase (a catalyst of gluconeogenesis) was inhibited by Fru2,6P(2). There was virtually no fructose 1,6-bisphosphatase activity in guard-cell chloroplasts of V. faba. It is therefore unlikely that the starch formed in these chloroplasts originates from imported triose phosphates or phosphoglycerate. The level of Fru2,6P(2) in guard-cell protoplasts and epidermal strips was about 0.1 to 1 attomole per guard cell in the dark (corresponding to 0.05 to 0.5 nanomole per milligram chlorophyll) and increased three- to tenfold within 15 minutes in the light. Within the same time span, hexose phosphate levels in guard-cell protoplasts declined to approximately one-half, indicating that acceleration of glycolysis involved stimulation of reactions using hexose phosphates. The level of Fru2,6P(2) in guard cells appears to determine the direction in which carbohydrate metabolism proceeds.
