Two groups of diabetic KK-CAy mice specifically bred for high and low sensitivity to exogenous acetylcholine and beta 1-adrenergic stimulation: interaction of higenamine and aconitine on pulse rate.

Diabetic KK-CAy mice were specifically bred for high and low sensitivity to the addition of exogenous acetylcholine (ACh). The sensitivity to ACh was measured by the change in pulse rate 2 min after the administration of ACh (10 mg/kg, s.c.). The two groups of mice, with high and low sensitivity to ACh, were specially selected and mated sequentially until the 12th filial generation. Although higenamine (100 micrograms/kg, i.p.), a beta 1-adrenergic agonist (a compound derived from aconite), had no effect per se, it inhibited aconitine (another compound derived from aconite extract)-induced bradycardia within 30 s of administration in ACh-low sensitive mice but not in ACh-high sensitive mice. The effects of aconitine and higenamine alone did not differ between these two groups of mice. This demonstrates that the high muscarinic and high beta 1-adrenergic sensitive mice may be stratified into two groups based upon an antagonistic interaction between higenamine and aconitine.

Expression of major histocompatibility complex in mouse peritoneal macrophages increasingly depends on plasma corticosterone levels: stimulation by aconitine.

Influence of aconite extract and a main component of aconite, aconitine were investigated on the plasma corticosterone level for the expression of major histocompatibility complex class II (Ia antigen) of macrophage. In peritoneal macrophage exposed to corticosterone (2.9-87 microns) in vitro the interferon (IFN)-gamma (0.1 unit/ml)-induced Ia antigen expression was inhibited in a dose-dependent manner. Ia antigen expression in macrophages harvested from the corticosterone (1.25mg/mouse/d, i.m., 4d)-excessive mice and aconitine (3 micrograms/kg, i.p., 7d)-treated mice was significantly stimulated, whereas that of adrenalectomized (7d) mice was inhibited. In macrophage exposed to aconitine up to 46.5 microns in culture Ia antigen expressed was not affected. Administration of aconite extract (3 mg/kg, i.p., 7d) and aconitine (3 micrograms/kg, i.p., 7d) increased plasma corticosterone levels. These result demonstrate that high levels of corticosterone increases the macrophage response to ifn-gamma on Ia antigen expression and that low levels of corticosterone decrease it. Aconite extract and aconitine stimulate the response to IFN-gamma-activated expression of Ia antigen by macrophages which is caused by increasing the plasma corticosterone level.

The relationship between serum gamma-glutamyl transpeptidase levels and hypertension: common in drinkers and nondrinkers.

A significant association between elevations of serum gamma-glutamyl transpeptidase (gamma-GTP) levels and those of blood pressure and hypertension has been reported separately in drinkers and nondrinkers. The aim of the present study is to evaluate whether the relationship between serum gamma-GTP and the prevalence of hypertension is the same or similar in both drinkers and nondrinkers. The study subjects comprised 4,920 male nondrinkers, 9,390 male daily drinkers, 8,081 female nondrinkers, and 278 female daily drinkers, who were aged 40 to 59 years. The prevalence of hypertension in the male and female daily drinkers was 1.5 and 1.3 times, respectively, higher than in the nondrinkers. Mean systolic blood pressure in the male and female drinkers was 4.4 and 3.1 mmHg, respectively, higher than in the nondrinkers. After adjusting for age, body mass index, and serum gamma-GTP levels, the differences in the prevalence of hypertension and the mean systolic blood pressure level between the drinkers and nondrinkers decreased to 1.2 times and 2.7 mmHg, respectively. Although these small differences remained statistically significant, the association between serum gamma-GTP and hypertension appears to be quite similar in both drinkers and nondrinkers, suggesting that hepatic steatosis may play a common, pathogenetic role in the development of hypertension.

Serum bone-type alkaline phosphatase activity in women living in a cadmium-polluted area.

To clarify the significance of elevated serum total alkaline phosphatase activity (t-ALP) in persons exposed to environmental cadmium (Cd), the fraction of ALP originating from bone (b-ALP) was assayed using a wheat-germ agglutinin method in 23 men and 20 women in a Cd-polluted area who showed excessive urinary beta 2-microglobulin excretion, and in 21 men and 44 women in a non-polluted area, in addition to 7 patients with itai-itai disease. The fraction of b-ALP increased linearly with the increase in t-ALP in the women, irrespective of Cd-exposure. Elevations of both t-ALP and b-ALP in the Cd-exposed women, including inhabitants of the Cd-polluted area and patients with itai-itai disease, were found with decreases in serum calcium and bone density. It is concluded that elevated serum ALP levels found in Cd-exposed persons reflect the development of Cd-induced bone damage.

In-vitro enhancement of mouse embryonic development by co-culture with peritoneal macrophages.

The effects of peritoneal macrophages on mouse fertilization and embryonic development in vitro were examined by a co-culture method. In experiment 1, oocytes from F1 strain mice were inseminated and cultured with a monolayer of peritoneal macrophages to determine the effects of the macrophages on fertilization. In experiment 2, 1-cell embryos, fertilized in vivo from either a non-blocking or a blocking strain, were also incubated with peritoneal macrophages to determine whether these cells overcame the developmental arrest. Although the peritoneal macrophages had an adverse effect on fertilization in the F1 strain, embryonic development was significantly improved by the co-culture. This beneficial effect on embryonic development was also observed in the embryos fertilized in vivo from the strain that exhibited a 2-cell block. These results suggest that peritoneal macrophages provide an effective co-culture system for the in-vitro development of mouse embryos.

[Tetracycline-resistant plasmid of Salmonella enteritidis isolated in Kumamoto City].

We isolated Salmonella choleraesuis subsp. choleraesuis serovar. Enteritidis (S. Enteritidis) from a mass of cases which broke out in September, 1991 in Kumamoto City. The isolates were shown to hold plasmids on which a tetracycline (TC)-resistant gene was located. The plasmid, about 9 kb in size, was capable of expressing the gene in Escherichia coli, unstable in S. Enteritidis and Escherichia coli. Thus in order to investigate the creeping prevalence of the TC-resistant plasmid in Kumamoto City and its spreading in other areas, the strains, a total of 41, including 37 isolates from sporadic cases with diarrhea in Kumamoto City, 3 isolates from 3 cases of food poisoning in Kitakyushu, Fukuoka and Chiba, and one strain isolated from egg solution in Kumamoto City, were examined by the hybridization test with this plasmid as a probe, as well as for their drug sensitivity and plasmid profile. As a result, 28 strains of 37 tested from the sporadic cases, the strains from food poisoning in Kitakyushu, Chiba and the one from egg solution, all harbored the same 9 kb TC-resistant plasmid. From the above observations, it was assured that, at the time of investigation, July to September in 1992, the prevalence of S. Enteritidis in Kumamoto City was Predominated by its strain carrying the TC-resistant plasmid. Furthermore, considering the fact that the strain harboring this plasmid was also found in the food poisoning in Kitakyushu and Chiba, it was suggested that spreading of this strain was not restricted to localities and was also associated with Chicken eggs according to its detection from the egg solution.

[A study of thyroid hormone levels of inhabitants of the cadmium-polluted Kakehashi River basin].

We compared thyroid hormone levels of inhabitants (19 men and 16 women) of the cadmium (Cd)-polluted Kakehashi River basin in Ishikawa Prefecture, with those of subjects (23 men and 47 women) living in a non-polluted area. In addition, we investigated the relationships between the thyroid hormone levels and indices of renal dysfunction induced by Cd exposure. The following results were obtained: 1) The free T4 level of females was significantly lower than that of controls. 2) The T3 level of inhabitants of both sexes was significantly higher than that of controls. 3) The level of free T4 among females became lower with the increases of urinary beta 2-microglobulin (beta 2-MG), urinary protein, urinary sugar, urinary amino acids and serum creatinine (Cr) levels, and with decreases of creatinine-clearance (CCr) and %TRP. 4) We could not find any relationship between the increase of T3 and the indices of renal dysfunction induced by Cd exposure in either sex.

Erythrocyte Na+/K(+)-ATPase and membrane and serum lipid profiles: as related to alcohol, body mass index and blood pressure.

Erythrocyte Na+/K(+)-pump activities have been measured in hypertensives, alcohol consumers and obese persons, but the results have been variously reported as decreased, increased or unchanged. We analyzed the relationships between erythrocyte Na+/K(+)-ATPase activities and the membrane and serum lipid profiles in 83 middle-aged men, to clarify the reasons for these inconsistencies. Increases in erythrocyte Na+/K(+)-ATPase activity related closely to decreases in cholesterol to phospholipid (C/P) ratio of the erythrocyte membrane. Decreases in the C/P ratio in turn related closely to elevations of serum triglycerides (TG) with increasing body mass index, and weakly to the volume of alcohol consumed. Thus, erythrocyte Na+/K(+)-ATPase activities depend largely on the membrane and serum lipid profiles as related to body weight and alcohol consumption, and which may be a cause of the previous conflicting findings. Erythrocyte Na+/K(+)-ATPase showed a positive association with blood pressure, independently of age, body mass index and serum gamma-glutamyl transpeptidase levels. Although the biological link of elevated erythrocyte Na+/K(+)-ATPase with the rise in blood pressure remains unclear, it may be a reflection of hyperinsulinemia in the subjects with a higher blood pressure due to overweight or excessive alcohol consumption.

Serum gamma-glutamyl transpeptidase level and blood pressure in nondrinkers: a possible pathogenetic role of fatty liver in obesity-related hypertension.

Relationships between body mass index, blood pressure (BP) and serum gamma-glutamyl transpeptidase levels were analysed in 859 male and 2,456 female nondrinkers aged 18-59 years. Serum gamma-GTP levels correlated with BP independently of age and body mass index in both men and women. The changes in BP of 391 male nondrinkers aged 35-54 years during a five year period correlated with the changes in serum gamma-GTP levels, independently of BP levels, age, body mass index and serum gamma-GTP at the beginning of the follow-up, and of the changes in body mass index. As the rise in serum gamma-GTP levels in nondrinkers depends largely on the progression in fatty change in the liver cells accompanying increases in the size of body mass, these results suggest a close association between the development of fatty liver and hypertension in obese persons.

[The role of phosphorylation cascade in insulin action].

Insulin induces a wide variety of growth and metabolic response in many cell types. Insulin initiates its biological effects by activation of tyrosine kinase in the beta-subunit and phosphorylates several proteins, such as insulin receptor substrate-1 (IRS-1), Shc. thereby activating phosphatidyl inositol 3-kinase activity, and ras activity. MAP kinase cascade activated by ras, 70kDaS6 kinase lying downstream of PI3-kinase, and the regulation of glycogen synthase have been discussed.

[Clinical treatment of urinary tract stones with Alexandrite laser].

Clinical treatment of urolithiasis with Alexandrite-laser which is a solid pulse laser system developed in Switzerland is reported. Two cases of renal stones, 3 cases of vesical stones, and 35 renal units in 34 cases of ureteral stones were treated. The approach to the aimed stone was, percutaneous to the renal stones, and transurethral to the vesical and ureteral stones. The clinical success rate was 92.5% (37/40), when we defined success as the endoscopically observed fragmentation. The uric acid stone in the bladder was not disintegrated even when the plasma occurred, but the ureteral uric acid stone was easily fragmented. The difference was thought to be attributed to the condition of the surface. The Alexandrite-laser system generates a 750 nm wavelength, and this wavelength is neither absorbed in water nor hemoglobin, so is thought to be harmless to the tissue. Actually, if misirradiation to the ureteral mucosa during lithotripsy was done, neither plasma formation nor perforation occurred. In conclusion, the Alexandrite-laser is thought to be safe and effective equipment for the treatment of urolithiasis.

Structural analysis of the CD3 zeta/eta locus of the rat. Expression of zeta but not eta transcripts by rat T cells.

We analyzed the structure and pattern of expression of rat TCR zeta- and eta-chains to investigate if these components function in activation and development of rat T cells. The rat zeta cDNA contained the complete open reading frame coding for a polypeptide of 164 amino acids and the 5' and 3' noncoding sequences. Comparison of the amino acid sequence to those of mouse and human counterparts revealed a high degree of similarity, more than 85% homology among all three species except for the signal peptide, which was especially high in the cytoplasmic domain including the nucleotide binding site and the possible tyrosine phosphorylation sites. Furthermore, we determined the nucleotide sequences of a rat genomic eta-like sequence located in the 3' region of the rat zeta-gene. Although it showed a high level of nucleotide similarity to mouse and human counterparts, 90.4 and 78.9%, respectively, the deduced polypeptide was very short (only 28 residues) and markedly divergent from the mouse and human eta-specific polypeptides due to frameshift mutations. Transcription of rat zeta was shown to be highly restricted to T cells; abundantly in thymocytes and scarcely in peripheral T cells. Surprisingly, the rat eta transcript could not be detected in any rat tissues so far tested by Northern blot analysis and even by the sensitive reverse transcription-polymerase chain reaction method, whereas it was readily detected in mouse thymus. These findings suggest that the zeta-chain has conserved roles in the TCR assembly and TCR-mediated signaling. However, the eta-chain seems not to be indispensable because of its structural diversity among these three species characterized to date and the apparent lack of eta expression in the rat.

Dose-response relationship between total cadmium intake and metallothioneinuria using logistic regression analysis.

The dose-response relationship for environmental cadmium exposure was assessed using logistic regression analysis. The prevalence of metallothioneinuria was employed as a response variable, while age and total cadmium intake, calculated from the average cadmium concentration in rice and duration of residence in the cadmium-polluted area, were used as explanatory variables. The target population comprised of 1843 cadmium-exposed and 240 non-exposed inhabitants of Ishikawa, Japan. The individuals were divided into 96 subgroups by sex, age (4 categories), cadmium concentrations in rice (3 categories) and length of residence in the polluted area (4 categories). Only total cadmium intake had a significant association with the prevalence of metallothioneinuria. In the non-exposed subjects total cadmium intakes corresponding to 2.5% prevalence of metallothioneinuria were calculated. Based on metallothionein levels expressed as either microgram/l urine or microgram/g creatinine, the total intakes were: 2.221 or 2.207 g in men and 2.365 or 0.319 g in women, respectively. Most of these values were similar to those reported by us previously, employing simple regression analysis. It is concluded, therefore, that a maximum allowable intake of about 2 g cadmium is a reasonable estimate for preventing the cadmium-induced renal dysfunction.

Dephosphorylation of human p34cdc2 kinase on both Thr-14 and Tyr-15 by human cdc25B phosphatase.

In mammalian cells, p34cdc2 kinase undergoes phosphorylation at threonine-14, tyrosine-15 and threonine-161 in the S and G2 phases of the cell cycle. At the onset of mitosis, the kinase becomes dephosphorylated at threonine-14 and tyrosine-15, resulting in activation. Cdc25 phosphatase has been shown to dephosphorylate tyrosine-15 in vitro, but whether it also does at threonine-14 remains unclear. In this study, we have found that human cdc25B phosphatase dephosphorylates both threonine-14 and tyrosine-15 but not threonine-161.

Serum gamma-glutamyl transpeptidase levels and hypertension in non-drinkers: a possible role of fatty liver in the pathogenesis of obesity related hypertension.

The relationships between increases in body mass index (BMI) and increases in hypertension were compared between non-drinkers with elevated serum gamma-glutamyl transpeptidase (gamma-GTP) levels (> or = 50 U/l) and those with normal levels, who comprised 10,952 men and 22,107 women aged 40-59 years recruited from an occupational health clinic. Hypertension was found in 16.1% and 13.5% of the men and women, and elevated serum g-GTP was found in 10.8% and 2.8% of the men and women, respectively. The prevalences of hypertension and elevated serum gamma-GTP levels were both increased with increased BMI. Hypertension was, however, shown to be 1.5 times more prevalent in the persons with elevated serum gamma-GTP levels than in those with normal levels in both sexes, even after adjusting for BMI by a multiple logistic analysis. It can be concluded that elevations of serum gamma-GTP, which are probably a reflection of fatty liver in the non-drinkers, are closely related to the development of hypertension associated with increased obesity.

Elevated cytosolic free calcium concentrations in platelets of hypertensive drinkers: a weak association with alcohol consumption.

Although the mechanisms of alcohol related hypertension remain unknown, it has been hypothesised that alcohol causes the accumulation of calcium ions (Ca2+) in vascular smooth muscle cells, resulting in the elevation of vascular tonus. To evaluate this contention, intra platelet free calcium concentrations (platelet [Ca2+]i) were determined in 20 young adult men, i.e. five each of hypertensive heavy drinkers, hypertensive light drinkers, normotensive heavy drinkers, and normotensive light drinkers. Platelet [Ca2+]i were higher in the ten hypertensives than in the ten normotensives, irrespective of alcohol consumption. The hypertensive drinkers also showed higher levels of serum gamma-glutamyl transpeptidase (gamma-GTP) and parathyroid hormone (PTH). Platelet [Ca2+]i in the total 20 subjects did not correlate with alcohol consumption, but correlated positively with serum gamma-GTP levels and SBP. These preliminary study results do not support the contention that Ca2+ accumulates in the vascular smooth muscle cells of humans as a result of chronic alcohol consumption. The significance of elevated platelet [Ca2+]i observed in the hypertensive heavy drinkers in the pathogenesis of alcohol related hypertension remains obscure.
