Differential targeting of nicotinic acetylcholine receptors by novel alphaA-conotoxins.

We describe the isolation and characterization of two peptide toxins from Conus ermineus venom targeted to nicotinic acetylcholine receptors (nAChRs). The peptide structures have been confirmed by mass spectrometry and chemical synthesis. In contrast to the 12-18 residue, 4 Cys-containing alpha-conotoxins, the new toxins have 30 residues and 6 Cys residues. The toxins, named alphaA-conotoxins EIVA and EIVB, block both Torpedo and mouse alpha1-containing muscle subtype nAChRs expressed in Xenopus oocytes at low nanomolar concentrations. In contrast to alpha-bungarotoxin, alphaA-EIVA is inactive at alpha7-containing nAChRs even at micromolar concentrations. In this regard, alphaA-EIVA is similar to the previously described alpha-conotoxins (e.g. alpha-MI and alpha-GI) which also selectively target alpha1- versus alpha7-containing nAChRs. However, alpha-MI and alpha-GI discriminate between the alpha/delta versus alpha/gamma subunit interfaces of the mouse muscle nAChR with 10,000-fold selectivity. In contrast, alphaA-conotoxin EIVA blocks both the alpha/gamma site and alpha/delta site with equally high affinity but with distinct kinetics. The alphaA-conotoxins thus represent novel probes for the alpha/gamma as well as the alpha/delta binding sites of the nAChR.

A noncompetitive peptide inhibitor of the nicotinic acetylcholine receptor from Conus purpurascens venom.

A paralytic peptide, psi-conotoxin Piiie has been purified and characterized from Conus purpurascens venom. Electrophysiological studies indicate that the peptide inhibits the nicotinic acetylcholine receptor (nAChR). However, the peptide does not block the binding of alpha-bungarotoxin, a competitive nAChR antagonist. Thus, psi-conotoxin Piiie appears to inhibit the receptor at a site other than the acetylcholine-binding site. As ascertained by sequence analysis, mass spectrometry, and chemical synthesis, the peptide has the following covalent structure: HOOCCLYGKCRRYOGCSSASCCQR* (O = 4-trans hydroxyproline; * indicates an amidated C-terminus). The disulfide connectivity of the toxin is unrelated to the alpha- or the alphaA-conotoxins, the Conus peptide families that are competitive inhibitors of the nAChR, but shows homology to the mu-conotoxins (which are Na+ channel blockers).

Cutaneous granulomas as a presenting sign in ataxia-telangiectasia.

Telangiectasia is the classic cutaneous finding of ataxia-telangiectasia (AT) and is often the physical finding that suggests the diagnosis. We report a patient in whom noninfectious cutaneous granulomas were the presenting cutaneous feature of AT and discuss immunodeficiency syndromes that are associated with similar cutaneous granulomas.

Genetic population structure of the prosobranch snail Potamopyrgus antipodarum (Gray) in Denmark using PCR-RAPD fingerprints.

Parthenogenetic species are often more widely distributed geographically than their sexual relatives. This success in colonizing can be explained either by dispersal of one or a few clones of wide physiological tolerance or by the distribution of many locally adapted clones. Here we test the hypothesis that successfully invading clones of Potamopyrgus antipodarum (Gray) are composed of a few broadly adapted genotypes by using polymerase chain reaction random amplified polymorphic DNA (PCR-RAPD) fingerprinting on six different populations of P. antipodarum from Denmark and three morphotypes of P. antipodarum from Britain. We detected two genotypes of P. antipodarum in six populations examined across Denmark using four decamer primers. The two genotypes were found to be morphologically and genetically indistinguishable from British P. antipodarum. In five of the six Danish populations only one genotype was found; at the remaining site, the two genotypes occurred sympatrically. The present study suggests that P. antipodarum successfully invaded Europe by the proliferation of very few clones.

Knowledge of HIV infection and the fear of mixing socially with HIV-infected and AIDS-sick patients among STD patients in Bergen, Norway.

This is a study of data from 599 STD (sexually transmitted disease) clinic patients in Bergen, Norway who visited the clinic in April-June 1989. We studied the patients' knowledge of modes of HIV transmission and their degree of ignorance of how HIV is not transmitted. Particular interest was focused on how misconceptions influenced the respondents' perception of HIV/AIDS sick persons and the importance of confidence in AIDS information from the health authorities. The objective of the study was to compare the data from this investigation of STD patients with data from similar studies in the general public conducted in Norway between 1986 and 1991. HIV-infected blood (98.3%; 589/599) and unprotected vaginal intercourse (91%; 545/599) were frequently suggested as a mode of HIV transmission, while 22.4% (134/599) did not recognize anal sex as an unsafe sexual practice. There was no statistically significant difference in their knowledge of modes of HIV transmission between the following patient subgroups: homosexuals/bisexuals, intravenous drug users (IVDUs)/prostitutes, sex customers and respondents classified as 'other heterosexuals'. Regarding ways in which HIV is not transmitted, ignorance was first and foremost related to whether the HIV virus could be passed on by oral kissing (wrong: 38.5%). Knowledge of how HIV is not transmitted was positively correlated with education (p < 0.05) and confidence in the HIV/AIDS information from health authorities (p < 0.05), and negatively correlated with age (p < 0.05). Respondents knowing how HIV is not transmitted considered social contact with HIV/AIDS sick persons to be harmless (p < 0.001). The present information campaigns on HIV/AIDS have been less effective than expected. In the future we must find new ways of informing the public by identifying the essential components that are missing from the information campaigns we have today.

[A campaign against home accidents--accident prevention efforts in the communities].

A nation-wide campaign aimed at preventing accidents in the home is being implemented in Norway. 95% of the municipalities acknowledge having received information material from the campaign, 33% report having established accident prevention committees, and 26% report having introduced preventive measures as a result of this national campaign. The study indicates that accidents are not recognized as yet as a major health problem in many municipalities. Identification of accidents as a health problem seems to be an important factor in the preventive efforts. To enhance further accident prevention efforts it seems important to increase awareness of accident as a health problem, and to increase the involvement of key personnel within the community.

[Occurrence of muscle contraction headache in children with abnormal incisor occlusion].

The principal result of the investigation indicates that children with abnormal incisor occlusion must be regarded as a risk group concerning muscle contraction headache (MCH). The need for orthodontic treatment of children with abnormal incisor occlusion is usually by layman regarded as being based upon psychosocial considerations. These considerations shall not be underestimated. For children as well as for grown up persons an appearance dominated by a visible abnormal incisor occlusion will be loading in contact with other people. From a professional point of view need for orthodontic treatment of children with abnormal incisor occlusion has usually been defined from the demand for stability. Grown up people with abnormal incisor relations are frequently referred to orthodontic specialists because of problems, such as migration of the incisors, extreme attrition or functional disturbances in the masticatory system. These problems are closely related to the abnormal incisor occlusion. With our present knowledge of the importance of the incisor relation for the function of the masticatory system, this condition must have high priority in the evaluation of need for orthodontic treatment of children. The treatment will, by improving the functional condition of the masticatory system and normalizing the function, have importance as prophylaxis and/or early treatment of a risk group concerning muscle contraction headache.

Electrodermal recovery data on children of schizophrenic parents.

Half-amplitude recovery of electodermal responses is compared in children of schizophrenic parents (high-risk subjects) and children of depressed or normal parents. The results are dissimilar to those reported by Mednick and colleagues on a Danish high-risk sample. No significant differences emerged among the three groups in our study. Recovery did not differ according to sex or severity of illness of the schizophrenic parent. Recoveries of high-risk subjects separated from their homes were not shorter than recoveries of subjects who remained home. Recovery time recorded in childhood was unrelated to global adjustment in adolescence.

Irrational ideas. Older vs. younger inpatients.

The relationship to age of irrational beliefs among psychiatric inpatients has not been explored using the rational-emotive model. This study addressed the following two questions: 1) Do older and younger psychiatric inpatients differ in irrational beliefs? 2) Do older depressives differ from older nondepressives in irrational beliefs? Upon admission to a large medical center, 58 younger (less than 45 years old) and 54 older (greater than 55 years old) subjects were assessed on a battery of psychological tests, including the Idea Inventory and the Beck Depression Inventory. Results showed that older and younger inpatients did not differ on irrational beliefs. Results also showed that older and younger groups of depressives did not differ on the irrationality scores. When a correlational analysis was used, depression was related to irrationality within the older group but not within the younger group.

Age-related norms for the Hooper Visual Organization Test.

Explored the effects of age, education, and IQ on Hooper Visual Organization Test (VOT) scores and age-related norms for psychiatric patients. Ss were 211 male, veteran, psychiatric inpatients, grouped into six age decades, who were administered routinely the VOT and Shipley Institute of Living Scale plus other tests upon admission. Analyzing the relation between age decade and VOT, a one-way analysis of variance produced an F significant at the .001 level. An intercorrelation matrix for all the study variables disclosed that age, IQ, and education all were correlated significantly with VOT scores, as well as with each other, except for IQ and age. However, a multiple regression analysis revealed that only age and IQ accounted for significant amounts of the variance in VOT scores. Thus, these results argue for the use of age-related norms as well as norms that correct for IQ.
