Comparative chromosome painting in mammals: human and the Indian muntjac (Muntiacus muntjak vaginalis).

We have used human chromosome-specific painting probes for in situ hybridization on Indian muntjac (Muntiacus muntjak vaginalis, 2n = 6, 7) metaphase chromosomes to identify the homologous chromosome regions of the entire human chromosome set. Chromosome rearrangements that have been involved in the karyotype evolution of these two species belonging to different mammalian orders were reconstructed based on hybridization patterns. Although, compared to human chromosomes, the karyotype of the Indian muntjac seems to be highly rearranged, we could identify a limited number of highly conserved homologous chromosome regions for each of the human chromosome-specific probes. We identified 48 homologous autosomal chromosome segments, which is in the range of the numbers found in other artiodactyls and carnivores recently analyzed by chromosome painting. The results demonstrate that the reshuffling of the muntjac karyotype is mostly due to fusions of huge blocks of entire chromosomes. This is in accordance with previous chromosome painting analyses between various Muntjac species and contrasts the findings for some other mammals (e.g., gibbons, mice) that show exceptional chromosome reshuffling due to multiple reciprocal translocation events.

Glycosylation of the GLP-1 receptor is a prerequisite for regular receptor function.

The GLP-1 receptor on RINm5F cells is a glycoprotein with a M(r) of 63,000. Treatment of the receptor with glycopeptidase F generated a protein with a M(r) of 51,000, indicating that the GLP-1 receptor contains N-linked glycans. Tunicamycin pretreatment concentration-dependently decreased GLP-1 binding to RINm5F cells due to a decreased receptor number without change of receptor affinity. Tunicamycin exerted no effect on the GLP-1 receptor mRNA expression. The stimulation of cAMP production was decreased in tunicamycin-treated cells. Our data show that glycosylation of the GLP-1 receptor is a precondition for regular receptor function.

Body position affects recumbent postprandial reflux.

There is a lack of consensus as to whether there is a difference in the amount of gastroesophageal reflux when lying with the right versus the left side down. In an attempt to answer this question we conducted a two-part study. The first consisted of a questionnaire distributed to 103 gastroenterologists with a special interest in the esophagus. The second was a randomized study in 20 normal subjects given a high-fat meal to induce reflux. Postprandial reflux was monitored by esophageal pH testing for 4 h after random assignment to either the left or right lateral decubitus position (left or right side down) on different days. Of the 87 gastroenterologists responding to our survey, 45 were uncertain as to which position was associated with worse reflux. Among the 42 who did specify, 27 (64%) favored the right lateral decubitus position as associated with more reflux symptoms. The direct comparison of the two positions demonstrated that the total amount of reflux time was significantly (p < 0.05) greater with subjects in the right decubitus position (231 min) than in the left (117 min). Although total episodes of reflux were greater when lying on the right side (227), compared with the left (198), the difference was not significant. In addition, average overall acid clearance was significantly (p < 0.001) prolonged with right side down (0.77 +/- 0.14 min/episode) compared with left side down (0.29 +/- 0.08 min/episode).

[3 cases of acute transverse syndrome caused by a dissecting aneurysm of the aorta].

A relatively rare cause of the acuto transverse syndrome, aneurysma dissecans of the aorta followed by disturbance of the circulation in the spinal cord, is described on the basis of three cases. The special features of the blood supply of the spinal cord and the etiopathogenesis, symptomatology and prognosis of aneurysma dissecans are discussed. Attention is drawn to the necessity of unobtrusive methods for case taking and clinical examination to save the patient unnecessary stress and to improve the chances of survival.

[Whipple disease of the CNS].

By the example of two cases that could only be cleared up by a postmortem examination the affection of the central nervous system by Whipple's disease is dealt with. The granulomatous inflammatory process which is almost exclusively localised in the grisea and shows a particular concentration in the diencephalon, in the cortex of the frontal, temporal and islet regions as well as in the deeper brain stem, had resulted in a variety of neurological-psychiatric symptoms, in most cases a systemic psychosyndrome, myoclonism and paresis of ocular movement. In both cases the diagnoses was verified by the electromicroscopic identification of the bacteria in the typical SPC cells.
